Homogenous preparations of XenB of Pseudomonas putida, pentaerythritol tetranitrate reductase of Enterobacter cloacae, and N-ethylmaleimide reductase of Escherichia coli, all type II hydride transferases of the Old Yellow Enzyme family of flavoproteins, are shown to reduce the polynitroaromatic compound 2,4,6-trinitrotoluene (TNT). The reduction of this compound yields hydroxylaminodinitrotoluenes and Meisenheimer dihydride complexes, which, upon condensation, yield stoichiometric amounts of nitrite and diarylamines, implying that type II hydride transferases are responsible for TNT denitration, a process with important environmental implications for TNT remediation.  相似文献

The Sierra de la Plata is an Aljibe yellow sandstone formation from the Acheulian period. There are a few shelters, some of them with rock art paintings. The most representative one, and subjected to anthropogenic pressure, is that of Atlanterra, situated in a residential area. This shelter contains some rock art paintings made with iron oxides. The bacteria present in these paintings were isolated and identified using an automatic method: fatty acid methyl esters profiling. Most of the bacteria belong to the Bacillus genus, B. megaterium being the most abundant species. The isolated strains are able to reduce hematite. This is significant due to the fact that Fe(III)-(hydr)oxides are the most abundant pigments in rock art.  相似文献

Female teleosts do not have oviducts because Müllerian ducts do not develop. Instead, the caudal region of the ovary, the gonoduct, connects to the exterior. Because of the lack of oviducts in viviparous teleosts, the embryos develop in the ovary, as an intraovarian gestation, unique in vertebrates. This is the first study to address the histology of the gonoduct in a viviparous teleost. The gonoduct of Poecilia reticulata was analyzed during previtellogenesis, vitellogenesis, and gestation. The gonoduct lacks germinal cells. From deep to superficial, the wall has simple cuboidal or columnar epithelium, loose connective tissue, longitudinal layer of smooth muscle, and visceral peritoneum. Cells of the immune system occur in the lumen and in the mucosa. The gonoduct was divided in three regions: 1) cephalic, 2) middle, and 3) caudal. At the initial part of each region, thin mucosal folds extend into the lumen. The cephalic region forms a tubular structure with light and irregular folds. The middle region has a wider lumen and is more irregular due to ventral invaginations and irregular and short mucosal folds; beneath the epithelium there are melano‐macrophage centers. The caudal region is delimited from the middle region by folds; however, they are thinner than these of the other regions. Ventral invaginations form exocrine glands, and the smooth muscle is thicker than in the other regions. During gestation, cells of the immune system are abundant; melano‐macrophage centers become larger and the glands exhibit desquamated cells. These observations suggest roles of the gonoduct in reducing the diameter of the lumen; receiving sperm during vitellogenesis; producing secretions, more abundant during vitellogenesis; and in immunological activity throughout the reproductive cycle. Developing diatom-based transfer functions for Central Mexican lakes   总被引:2,自引:2,他引:0  
This paper is the first attempt to produce diatom-based transfer functions for the northern tropical Americas. A dataset of 53 modern diatom samples and associated hydrochemical variables from 31 sites in the volcanic highlands of central Mexico is presented. The relationship between diatom species distribution and water chemistry is explored using canonical correspondence analysis (CCA) and partial CCA. Variance partitioning indicates that ionic strength and ion type both account for significant and independent portions of this variation. Transfer functions are developed for electrical conductivity (r 2 = 0.91) and alkalinity (as a percentage of total anions) (r 2 = 0.90), reflecting ionic strength and ionic composition respectively. Prediction errors, estimated using jack-knifing, are low for the conductivity model, but the carbonate transfer function performs less well. This study highlights the potential for diatom-based quantitative palaeoenvironmental reconstructions in central Mexico. However, a number of key diatom species found in fossil material are not represented in the modern flora. Sampling of additional sites may resolve this, but it is thought that the lack of modern analogues may reflect the high degree of anthropogenic disturbance in many of the catchments. This highlights the problem of trying to reconstruct pre-disturbance environmental changes in highly modified ecosystems. One possible solution is to merge the central Mexican data with the African dataset, which includes sites of similar chemical composition, but which have not suffered the same degree of disturbance.  相似文献

Spastic paraplegia 46 refers to a locus mapped to chromosome 9 that accounts for a complicated autosomal-recessive form of hereditary spastic paraplegia (HSP). With next-generation sequencing in three independent families, we identified four different mutations in GBA2 (three truncating variants and one missense variant), which were found to cosegregate with the disease and were absent in controls. GBA2 encodes a microsomal nonlysosomal glucosylceramidase that catalyzes the conversion of glucosylceramide to free glucose and ceramide and the hydrolysis of bile acid 3-O-glucosides. The missense variant was also found at the homozygous state in a simplex subject in whom no residual glucocerebrosidase activity of GBA2 could be evidenced in blood cells, opening the way to a possible measurement of this enzyme activity in clinical practice. The overall phenotype was a complex HSP with mental impairment, cataract, and hypogonadism in males associated with various degrees of corpus callosum and cerebellar atrophy on brain imaging. Antisense morpholino oligonucleotides targeting the zebrafish GBA2 orthologous gene led to abnormal motor behavior and axonal shortening/branching of motoneurons that were rescued by the human wild-type mRNA but not by applying the same mRNA containing the missense mutation. This study highlights the role of ceramide metabolism in HSP pathology.  相似文献

1. Heme regulation before the appearance of hyperplastic nodules was investigated in mice models of hepatocarcinogenesis. 2. With this aim 5-aminolaevulinate synthetase (ALA-S), microsomal heme-oxygenase (MHO), mitochondrial and cytoplasmic rhodanese activities were examined throughout a period of 35 days in animals exposed to dietary p-dimethylaminoazobenzene (DAB). 3. ALA-S activity was significantly diminished (50%) on day 14, then showing a sharply rising profile from day 28 onwards, and reaching 350% on day 35. 4. A similar profile was observed for mitochondrial rhodanese activity. 5. Changes in MHO and cytoplasmic rhodanese activities were almost the opposite to those observed for ALA-S. 6. The distinctive alteration in mitochondrial and cytoplasmic rhodanese would suggest that it plays a subtle role in ALA-S regulation during carcinogenesis initiation through a mechanism that appears to involve subcellular localization controls perhaps by means of the breakage of cystine trisulphide postulated to act as an ALA-S activator. 7. Taking into account the present results, we suggest a probable mechanism for the onset of hepatocarcinogenesis that includes a primary activating liver status, provoking biochemical aberration leading to the stage of initiation of hepatocarcinogenesis involving the whole organ.  相似文献

Three Nucleopolyhedrovirus (NPVs) originally isolated from Heliothis armigera larvae, collected from Portugal (HearNPV-PO) and two places in Spain (HearNPV-SP1 and HearNPV-SP2), and three previously described NPVs were compared biochemically and biologically. Restriction endonuclease analysis of the virus genome with several enzymes revealed that isolates HearNPV-SP1 and HearNVP-SP2 are unique but closely related genotypes which represent two new strains of the NPVs of H. armigera. The DNA fragment profiles of HearNPV-PO were distinct from those of the HearNPV-SP1 and HearNPV-SP2, with all the enzymes used, while they were identical to the Mamestra brassicae NPV strain present in the commercial product MAMESTRIN®. Bioassays in third-instar H. armigera larvae showed that the LD50 value obtained for HearNPV-SP1 (68 occlusion bodies/larva) was about two- and six-times lower than those of HearNPV-SP2 and a Russian isolate, HearNPV-RU, respectively. The corresponding LT50 values were not found to differ significantly between these three virus isolates at comparable doses.  相似文献

Nonrandom mating whereby parents are related is expected to cause a reduction in effective population size because their gene frequencies are correlated and this will increase the genetic drift. The published equation for the variance effective size, Ne, which includes the possibility of nonrandom mating, does not take into account such a correlation, however. Further, previous equations to predict effective sizes in populations with partial sib mating are shown to be different, but also incorrect. In this paper, a corrected form of these equations is derived and checked by stochastic simulation. For the case of stable census number, N, and equal progeny distributions for each sex, the equation is [formula: see text], where Sk2 is the variance of family size and alpha is the departure from Hardy-Weinberg proportions. For a Poisson distribution of family size (Sk2 = 2), it reduces to Ne = N/(1 + alpha), as when inbreeding is due to selfing. When nonrandom mating occurs because there is a specified system of partial inbreeding every generation, alpha can be substituted by Wright's FIS statistic, to give the effective size as a function of the proportion of inbred mates.  相似文献
