UV and Ethyl Methanesulfonate Effects in Hyperthermophilic Archaea and Isolation of Auxotrophic Mutants of Pyrococcus Strains

The lethal and mutagenic effects of ethyl methanesulfonate (EMS) and UV on nine archaeal strains belonging to each of the two described genera of Thermococcales, Pyrococcus and Thermococcus, were investigated. To test the efficiency of the EMS and UV mutagenesis under a variety of experimental conditions, we chose Pyrococcus abyssi strain GE5 as a model strain. We observed a strong induced mutagenicity in both cases, since the spontaneous mutation frequency (expressed as the frequency of resistance to 5-fluoroorotic acid) increased up to 150-fold with EMS and 400-fold with UV, after mutagen exposure. Although a heterogeneous response to the induced effects caused after EMS and UV exposures was detected for all the other sulfothermophilic archaea tested, an efficient mutagenicity of Pyrococcus-like isolates GE27, GE23, and GE9 was observed. Optimal procedures described for UV mutagenesis yielded a number of useful uracil auxotrophic mutant strains of Pyrococcus abyssi. Received: 2 May 1996 / Accepted: 3 July 1996     

Pelodera (syn. Rhabditis) strongyloides as a cause of dermatitis – a report of 11 dogs from Finland

Background  

Pelodera (Rhabditis) strongyloides is a small saprophytic nematode that lives in decaying organic matter. On rare occasions, it can invade the mammalian skin, causing a pruritic, erythematous, alopecic and crusting dermatitis on skin sites that come into contact with the ground. Diagnosis of the disease is based on case history (a dog living outdoors on damp straw bedding) with characteristic skin lesions and on the demonstration of typical larvae in skin scrapings or biopsy. Pelodera (rhabditic) dermatitis cases have been reported mainly from Central European countries and the United States.     

Seasonal variations in bacterial colonisation of stainless steel,aluminium and polycarbonate surfaces in a sea water flow system

Before the construction of an Ocean Thermal Energy Conversion (OTEC) power plant, an experimental station was built near the entrance of the Bay of Brest (France) in order to perform preliminary studies. Bacterial colonisation of three substrata (stainless steel UB6, aluminium AG4 and polycarbonate filters) was observed in a continuous flow seawater system. The study was performed between May 1983 and May 1984 and included seven experiments of 15 days each.

The early events in bacterial colonisation of these materials may be affected by both water quality and type of substratum. If these two parameters influence the adsorption step of the biological process (0 to 1 h), seasonal variation in water quality seems to be more important during the accumulation step (1 h to 6 days), because of its effect on bacterial metabolism.
