The immunocytochemically observed intracellular redistribution of phytochrome as a function of its molecular form is described by utilizing color photomicrography. The reversible change from a diffuse to a discretely localized distribution following photoconversion of the red-absorbing Pr form to the far-red-absorbing Pfr form observed with etiolated oat (Avena sativa L., cv. Garry) coleoptile parenchyma cells is not seen with etiolated wheat (Triticum sativum L., cv. unknown), barley (Hordeum vulgare L., cv. Harrison), or rye (Secale cereale L., cv. Balbo). Whether redistribution in these latter cases does not occur or is below the limit of detection is not known. Upon continuous actinic irradiation, phytochrome, which is discretely localized as Pfr, rapidly disappears by both immunocytochemical and spectral assay. However, after about 90 min irradiation, a new association of phytochrome with nuclei is evident which is more pronounced after 4 or 8 h of irradiation. With longer irradiation times there is a total loss of antigenically detectable phytochrome at the resolution employed in these experiments.  相似文献

Mesenchyme cells derived from embryonic mouse limb buds were cultured at high cell density. During the first 24 h in culture, groups of mesenchyme cells condensed and formed cell contacts and specialized junctions. These condensations were the nodule primordia which gave rise to cartilage nodules. The cell contacts were lost as the mesenchyme cells in the primordia developed into cartilage nodules. The mature nodules contained chondrocytes isolated from one another by an extensive extracellular matrix consisting of cartilage type collagen fibrils and proteoglycan granules. The differentiation of the mesenchyme cells to chondrocytes was also characterized by the loss of a 240,000-MW cell surface glycoprotein and the appearance of an 80,000-MW surface protein. The addition of vitamin A to the medium on Day 1 inhibited chondrogenesis. The cells were closely packed together, and the limited extracellular space contained thick, banded collagen fibrils with no proteoglycan granules. The cells exhibited extensive areas of close membrane contact and specialized junctions. Vitamin A-treated cultures also retained the 240,000-MW surface glycoprotein and retarded the appearance of the 80,000-MW cell surface protein. The results of this study suggest that cell surface features normally present on mesenchyme cells are maintained and exaggerated by vitamin A.  相似文献

Rats were given a single dose of saline, saline supplemented with xanthine oxidase (XO), half cream and half milk (H/H) and H/H supplemented with XO. XO was determined by a spectrophotometric method at 297 nm in serum at 0, 2, 4 and 6 hours after administration. The method is rapid, reliable and compares favorably with reported assays. No significant difference was obtained between the two saline treatments. The XO activity in serum of animals receiving the H/H increased significantly at 2 hours and then decreased. The H/H supplemented with XO demonstrated a maximum activity in serum at 4 hours and then declined to a value similar to that of the H/H treatment and below the XO level at 0 time. The initial increase in XO activity in serum of rats receiving the H/H treatments may indicate that XO is absorbed in the gastrointestinal tract or that the H/H materials stimulated endogenous XO activity.  相似文献

l-(U-(14)C)-5-oxoproline (pyrollidone carboxylic acid or pyroglutamic acid) was infiltrated into detached leaves of a number of species and incubated for 1 to 6 hours. In every case, conversion to labeled glutamate and glutamine was observed. The amount converted varied from 1 to 64% of the total label fed depending on the species. The ratio of glutamate-(14)C to glutamine-(14)C ranged from 5 in Vicia faba to 1 in sugar beet. This ratio could be affected by preinfiltrating various compounds before allowing the uptake of the 5-oxoproline. When l-methionine-dl-sulfoximine was prefed to sugar beet leaves, the glutamate-glutamine ratio increased from 1 to 10. Prior treatment of V. faba leaves with azaserine resulted in essentially only labeled glutamine being recovered. Preinfiltration with NaF or ATP gave similar results in that the glutamate-glutamine ratio was greatly decreased. The results are consistent with glutamate being produced from the 5-oxoproline and then being converted to glutamine.  相似文献

Programmed cell death occurs in a defined region of the secondary palatal epithelium (medial edge) during its development in vivo or in vitro. The purpose of this study was to examine the effects of various inhibitors of macromolecular synthesis, particularly glycoprotein synthesis, on the death of palatal medial-edge epithelial cells in vitro. Rat palatal shelves explanted on gestational Day 15 and cultured singly showed medial-edge cell degeneration after 48 hr, whereas in the presence of 6-diazo-5-oxo-l-norleucine (DON), 2-deoxyglucose (DOG), or cycloheximide, cell death was prevented. This was in contrast to the lack of inhibition of cell death in the presence of actionomycin D or cytosine arabinoside. Selectively adding metabolites which bypass the effect of DON on various synthetic pathways demonstrated that the inhibition of cell death by DON was most likely due to a block in glucosamine formation. The synthesis and activity of various lysosomal enzymes were not affected by DON, whereas a marked decrease was observed with both cycloheximide and DOG. These results suggest that palatal epithelial cell death is not a passive event, but an active process requiring the programmed synthesis of specific proteins.  相似文献

Clinocardium nuttallii from Yaquina Bay, Oregon, were found to harbor an endosymbiotic alga. Some aspects of this relationship are presented. The areas of infection include the siphon, mantle, and occasionally the foot. C. nuttallii under 2 years old are not infected; the incidence and intensity of the infection increases in the older age groups. For all ages the incidence averages 35%. Pigment composition, morphology, and growth characteristics of the alga are similar to those found in the genus Chlorella. In situ, the algae form dense, sometimes massive colonies but do not appear to cause any host reaction or enfeeblement. The thick layers surrounding the algal cells in situ, the dense colonies, and the in vitro reaction to host extract suggest the alga is parasitic. However, the presence of chloroplasts, the location of the algal cells only in illuminated tissue, the seemingly unhampered reproduction in situ, and the eventual adaptation to mineral medium suggest the alga is a facultative parasite. Experimental infection was achieved by feeding mature uninfected cockles a diet of only symbiont cells. In vitro observations found the symbiont cells readily engulfed by host blood amoebocytes. It is believed that the animal acquires the infection through phagocytosis of the symbiont cells and subsequent diapedesis across epithelial barriers by host amoebocytic cells.  相似文献

Homozygous brachymorphic (bmbm) mice have a disproportionately short stature, similar to human achondroplasia. We previously showed that each zone of growth in young bmbm epiphyseal cartilages is smaller than normal and that the extracellular matrix appears to contain normal collagen fibrils, but smaller and reduced numbers of proteoglycan matrix granules. Our studies reported here indicate that mutant, like normal cartilage, synthesizes type II collagen and contains normal quantities of glycosaminoglycans as judged by uronic acid content. However, the glycosaminoglycans from the mutant differ from the normal in their chromatographic and electrophoretic properties. Further studies established that glycosaminoglycans from cartilages of brachymorphic animals were undersulfated. Whereas chondroitinase digests of glycosaminoglycans from cartilage of normal C57Bl6J 5-day-old mice contained predominantly disaccharides sulfated in the 4-position, that of the mutant contained appreciable unsulfated disaccharides as well.  相似文献

—In living rats the concentration of insulin in the circulating blood was raised and independently of this the glucose concentration in the blood plasma was varied from hyperglycaemic to hypoglycaemic levels. Hyperglycaemia increased the influx of glucose into the brain and it also, for a limited period, increased the glucose gain by the brain. Insulin, on the other hand, did not affect influx but significantly increased the gain of glucose by the brain. It is suggested that although both hyperglycaemia and insulin can increase glucose gain by the brain they do so in entirely different ways.  相似文献

Male sand fiddler crabs, Uca pugilator, wave a claw to attract females to a breeding burrow. The effect of claw size on the likelihood of attracting mate-seeking females is little studied although in some other species females preferentially approach larger males. We used paired mirrors to reflect different sized images of the same male in a South Carolina (USA) back-beach habitat. Use of mirrors controlled for waving rate (but not velocity), waving motion, claw color, and claw shape. Female choice was attributed to instances in which a female contacted one of two mirrors. Paired mirrors were inclined toward one another in an arena defined by blinds and containing a single male. Two reflections of the male were visible to females moving approximately 50 cm toward the mirrors. The male was behind a small internal blind and not directly visible. In one-half of the trials, a non-magnifying mirror was placed at the bottom of mirrors so that only the elevated claw was magnified. Thus, body and burrow size and apparent distance were controlled. Receptive females preferred the larger reflection whether or not the body of the male was magnified, suggesting the importance of claw size. Non-receptive females did not exercise a choice. Control arenas, without a male, rarely attracted females. The results suggest that females choose on the basis of claw size. Selection on females may favor response to larger-clawed males because use of the claw in contests between males over burrows maintains the honesty of claw size as a signal of burrow quality.  相似文献
