Possibility of perchlorate reduction by microbes raises hope for an eco-friendly mode of degradation of this toxic rocket fuel. This study reports 3 isolates (A1, A2 and A3) capable of molybdenum-independent degradation of perchlorate under aerobic conditions. The rate of degradation was the highest when perchlorate concentration was 17 mM, and then 3.2 mM, 4.7 mM and 4.1 mM of perchlorate was reduced by isolates A1, A2 and A3 (respectively) after 72 h at 28°C under aerobic conditions. Presence of perchlorate at a concentration higher than 17 mM resulted in some inhibition of perchlorate reduction. 16S ribosomal RNA gene analysis revealed isolate A1 to bePseudomonas stutzeri (Proteobacteria) while isolates A2 ad A3 where found to belong to the genusArthrobacter (Actinobacteria). The study, apart from demonstrating ribotyping as a rapid method of identification of economically important soil microbes, also raised prospects for using artificial consortia for environmental degradation of perchlorate, without apparent domination ofDechloromonas spp. (a group of microbes known for perchlorate remediation in the environment).  相似文献

Although the production of poly-β-hydroxybutyrate (PHB) has many biological, energetic and environmental advantages over chemically synthesized polymers, synthetic polymers continue to be produced industrially since the productivities of fermentation processes fr PHB are not yet economically competitive. Abnormal protein kinetics could be a cause of several diseases associated with essential life processes. An accurate understanding of protein dynamics and turnover is essential for developing diagnostic or therapeutic tools to monitor these changes. Raman spectroscopy in combination with stable isotope probes (SIP) such as carbon-13, and deuterium has been a breakthrough in the qualitative and quantitative study of various metabolites. In this work, we are reporting the utility of Raman-SIP for monitoring dynamic changes in the proteome at the community level. We have used 13C-labeled glucose as the only carbon source in the medium and verified its incorporation in the microbial biomass in a time-dependent manner. A visible redshift in the Raman spectral vibrations of major biomolecules such as nucleic acids, phenylalanine, tyrosine, amide I, and amide III were observed. Temporal changes in the intensity of these bands demonstrating the feasibility of protein turnover monitoring were also verified. Kanamycin, a protein synthesis inhibitor was used to assess the feasibility of identifying effects on protein turnover in the cells. Successful application of this work can provide an alternate/adjunct tool for monitoring proteome-level changes in an objective and nondestructive manner.  相似文献

We present evidence from experiments on overwintering populations of two Palearctic-Indian latitudinal migratory birds, the black-headed bunting (Emberiza melanocephala) and the red-headed bunting (E. bruniceps), that the bird’s clock in interaction with day length regulates seasonal rhythms of migration and reproduction such that physiological conflict between them is reduced. Initiation and termination of the body mass and testicular cycles are separately regulated photoperiodic events. For example, under stimulatory photoperiods the response curve of body mass does not overlap with that of the testicular growth. A response-specific photoperiodism is adaptive, since gain in body mass, critical to spring migration, precedes gonadal recrudescence. Finally, migration as indicated by the night-time migratory restlessness under experimental situations (e.g., intense locomotion under caged condition-called Zugunruhe) appears to be regulated by a separate circadian oscillator.  相似文献
