The localization of lipid globules was studied in the adrenal glands from 35 fetuses and 10 adult humans by histochemistry. The fetal adrenal showed the presence of numerous lipid globules in the zona glomerulosa, whereas in the adult the lipid globules were mainly found in the zona fasciculata. These lipid globules were confirmed as cholesterol in the fetal adrenals.  相似文献

Seed germination of Melaleuca ericifolia (swamp paperbark) is delayed by submergence in water but the final percentage germination after removing seeds from water is unaffected; some seeds will germinate and the cotyledons emerge whilst submerged. Germination is totally inhibited at salinities of 14% and a population which naturally occurs adjacent to coastal saltmarsh was inhibited more by salinities of 6–12% than two other populations from freshwater habitats. Seedling growth may be increased by water-logging conditions and the degree of response differs for different seed sources. Inundation stimulates root growth and new roots are thick and aerenchymatous. Root systems in the field run horizontally and are shallow, apparently being restricted by clay at depth. All populations studied showed a similar seedling growth-response to salinity treatments; growth declined at salinities ≤ 13% and was severely restricted at 21%, although no seedlings died after 48 days’exposure to the latter treatment.  相似文献

Procathepsin H in kidney and liver microsomal lumen was identified to have a molecular mass of 41 kDa by immunoblot analysis. The proenzyme was then concentrated by applying the microsomal contents to a concanavalin A-Sepharose column. When the concanavalin A-adsorbed fraction was incubated at pH 4.0 at 20 degrees C, the activity measured with synthetic substrate increased 3.5 times over that of the control after 24 h incubation. Immunoblot analysis showed that acidic treatment caused the disappearance of procathepsin H. Thus the proenzyme might be processed to the mature enzyme under acidic conditions. The marked increase of enzymatic activity and the conversion of proenzyme were completely blocked with pepstatin which is a potent inhibitor of aspartic proteases. These results suggested that a protease for processing procathepsin H might be cathepsin D, a major lysosomal aspartic protease. Therefore, procathepsin H seems to be synthesized first in the enzymatically inactive form in endoplasmic reticulum and successively converted into the active form in lysosomes during biosynthesis.  相似文献

1. Time-curves of insulin effects on energy-producing systems in different cellular compartments of rat diaphragm muscle have revealed: (a) a rapid (within minutes) and transient stimulatory effect of insulin on cytoskeletal phosphofructokinase and aldolase and mitochondrial hexokinase. (b) A slower and consistent stimulatory effect on glucose 1,6-bisphosphate level, with concomitant gradual activation of cytosolic phosphofructokinase. Fructose 2,6-bisphosphate levels were not changed by insulin. (c) Lactate concentration correlated with the stimulation of cytoskeletal and cytosolic glycolysis. 2. Calmodulin antagonists, trifluoperazine or CGS 9343B, prevented all these effects of insulin. 3. These results suggest that cytoskeletal glycolysis and mitochondrial oxidation are the source of ATP for the rapid actions of insulin, whereas cytosolic glycolysis is the source of ATP for the slow actions of insulin. Calmodulin is involved in all these effects of insulin.  相似文献
