Impacts of climate change on organisms are already apparent, with effects ranging from the individual to ecosystem scales. For organisms engaged in mutualisms, climate may affect population performance directly or indirectly through mediated effects on their mutualists. We tested this hypothesis for two stink bugs, Acrosternum hilare and Murgantia histrionica, and their gut-associated symbionts. We reared these species at two constant temperatures, 25 and 30°C, and monitored population demographic parameters and the presence of gut-associated symbionts with diagnostic PCR primer sets. Both stink bugs lost their respective gut symbionts within two generations at 30°C. In addition, the insect survivorship and reproductive rates of both A. hilare and M. histrionica at 30°C were lower than at 25°C. Other demographic parameters also indicated a decrease in overall insect fitness at the high temperature. Collectively our data showed that the decrease in host fitness was coupled with, and potentially mediated by, symbiont loss at 30°C. This work illustrates the need to better understand the biology of animal-symbiont associations and the consequences of local climate for the dynamics of these interactions.The effect of climate on organisms, communities, and the environment at large has become a pressing issue for biologists and environmental scientists. Recent studies indicate that previous forecasts were conservative in their predictions for the magnitude of global warming (29). Up-to-date models suggest that the global mean surface temperature will increase by 1.8 to 4°C by the year 2100 (11). The ecological impact of such warming is already apparent (35) in the effects seen on species fitness (24), range shifts (22), species interactions (10), and community structure (32).It is important to note that many macroorganisms live in symbiosis with microbes and that host fitness may be affected indirectly by higher temperatures due to the disruption of mutualistic relationships. Some corals, for example, have symbiotic relationships with photosynthesizing dinoflagellates (zoothanxellae) that provide them with nutrients (31). Higher water temperatures in reef ecosystems, among other factors, induce the expulsion of microbial symbionts by the host, resulting in coral bleaching (15). Therefore, it is plausible that observed effects of climate on species distribution or performance might stem from disruption of symbiotic interactions as much as from direct effects on host biology.Despite the current interest in insect-microbe symbioses, the vast majority of such systems have been poorly studied. A group of insects that has recently received some attention are the true bugs (Hemiptera, Pentatomomorpha). Studies in the early 1900s suggested that mutualistic bacteria colonized a portion of the gut of insects in different pentatomomorphan families (9). More specifically, monocultures of bacteria were present in high densities in the crypt- (or cecum-) bearing organ preceding the hindgut of hosts, with different bacterial taxa associated with different true bug families. Furthermore, studies have shown that these symbionts are mutualistic (1, 8, 13, 14, 26). Among these bug families, the stink bugs (Pentatomidae) have been shown to depend on their gut symbionts (1). Pentatomid symbionts are polyphyletic and closely related to plant-associated bacteria in the genera Erwinia and Pantoea (25). Although the mechanism of symbiont vertical transmission is poorly understood, females seem to smear the surface of eggs with bacteria while ovipositing (3). Aposymbiotic first instars hatch but remain on the surface of eggs and acquire the symbiont by probing on the egg surface, as evidenced by the fact that surface sterilization of egg masses generates aposymbiotic individuals (1, 26, 28).Climate change has already affected stink bug performance and geographic range (19, 20). In Japan, populations of two pentatomid species, Nezara viridula and N. antennata, have shifted northwards and to higher elevations, respectively, over the last 50 years (33). However, we found previously that for one of these species high temperature eliminated gut symbionts, without any clear decrease in host fitness (27). Thus, it remains unclear whether temperature change played a role, either directly or indirectly, in these geographic shifts. To better understand the extent to which temperature mediates stink bug ecology and prevalence of their gut bacteria, we conducted laboratory studies with two pentatomid species, Acrosternum hilare and Murgantia histrionica. We show that high temperature affects the symbiotic relationship, with concomitant reduction in insect fitness.  相似文献

Introduction

The purpose of this study was to evaluate the effects of L-4F, an apolipoprotein A-1 mimetic peptide, alone or with pravastatin, in apoE-/-Fas-/-C57BL/6 mice that spontaneously develop immunoglobulin G (IgG) autoantibodies, glomerulonephritis, osteopenia, and atherosclerotic lesions on a normal chow diet.

Methods

Female mice, starting at eight to nine weeks of age, were treated for 27 weeks with 1) pravastatin, 2) L-4F, 3) L-4F plus pravastatin, or 4) vehicle control, followed by disease phenotype assessment.

Results

In preliminary studies, dysfunctional, proinflammatory high-density lipoproteins (piHDL) were decreased six hours after a single L-4F, but not scrambled L-4F, injection in eight- to nine-week old mice. After 35 weeks, L-4F-treated mice, in the absence/presence of pravastatin, had significantly smaller lymph nodes and glomerular tufts (PL, LP < 0.05), lower serum levels of IgG antibodies to double stranded DNA (dsDNA) (PL < 0.05) and oxidized phospholipids (oxPLs) (PL, LP < 0.005), and elevated total and vertebral bone mineral density (PL, LP < 0.01) compared to vehicle controls. Although all treatment groups presented larger aortic root lesions compared to vehicle controls, enlarged atheromas in combination treatment mice had significantly less infiltrated CD68+ macrophages (PLP < 0.01), significantly increased mean α-actin stained area (PLP < 0.05), and significantly lower levels of circulating markers for atherosclerosis progression, CCL19 (PL, LP < 0.0005) and VCAM-1 (PL < 0.0002).

Conclusions

L-4F treatment, alone or with pravastatin, significantly reduced IgG anti-dsDNA and IgG anti-oxPLs, proteinuria, glomerulonephritis, and osteopenia in a murine lupus model of accelerated atherosclerosis. Despite enlarged aortic lesions, increased smooth muscle content, decreased macrophage infiltration, and decreased pro-atherogenic chemokines in L-4F plus pravastatin treated mice suggest protective mechanisms not only on lupus-like disease, but also on potential plaque remodeling in a murine model of systemic lupus erythematosus (SLE) and accelerated atherosclerosis.  相似文献

Patterns of spatial variation of molluscan communities associatedwith coralline algal turfs were evaluated over 1,000 kmof the coast of Argentinean Patagonia. A hierarchically-nestedexperimental design was used to determine the relative importanceof molluscan assemblage variation at three different spatialscales (shores, sites and cores). Hypotheses were also testedabout the potential role of habitat variables (frond density,frond length, sediment and epiphytes) for determining molluscancommunity structure. In total, 38 molluscan species were foundcomprising 16, 18 and 4 species of bivalves, gastropods andpolyplacophorans, respectively. Densities of molluscs in corallineturfs reached ca 77,000 individuals per m2 and were dominatedby mussels, especially Perumytilus purpuratus. Multivariateand univariate analyses of assemblage structure consistentlyshowed that variation at scales of metres and hundreds of kilometresdominated, with sites 20–50 m apart always contributingless than 24% of the total. Significant associations betweenmolluscan community structure and both frond density and frondlength demonstrated the potential importance of habitat structurein determining community structure at local scales. Variationin molluscan assemblages at the scale of shores, however, didnot appear to correlate with latitudinal, temperature or waveexposure gradients, indicating that other processes must beoperating. The compositions of molluscan assemblages in corallineturfs on the coast of Argentina were similar to those reportedfor central Chile. Comparisons of the richness of these SouthAmerican assemblages to other parts of the world revealed somestriking biogeographical patterns that warrant further investigation.Overall, this work highlights the general importance of small-scalevariation in molluscan assemblages on rocky shores and the consistentinfluence of habitat complexity in determining the structureof diverse molluscan communities associated with mat-like habitats. The isocoumarins (1-50 microM) paepalantine (9,10-dihydroxy-5,7-dimethoxy-1H-naptho(2,3c)pyran-1-one), 8,8'-paepalantine dimer, and vioxanthin isolated from Paepalanthus bromelioides, were assessed for antioxidant activity using isolated rat liver mitochondria and non-mitochondrial systems, and compared with the flavonoid quercetin. The paepalantine and paepalantine dimers, but not vioxanthin, were effective at scavenging both 1,1-diphenyl-2-picrylhydrazyl (DPPH(*)) and superoxide (O(2)(-)) radicals in non-mitochondrial systems, and protected mitochondria from tert-butylhydroperoxide-induced H(2)O(2) accumulation and Fe(2+)-citrate-mediated mitochondrial membrane lipid peroxidation, with almost the same potency as quercetin. These results point towards paepalantine, followed by paepalantine dimer, as being a powerful agent affording protection, apparently via O(2)(-) scavenging, from oxidative stress conditions imposed on mitochondria, the main intracellular source and target of those reactive oxygen species. This strong antioxidant action of paepalantine was reproduced in HepG2 cells exposed to oxidative stress condition induced by H(2)O(2).  相似文献

The influence of factors, both abiotic (light and nutrients) and biotic (meadow structure, herbivory and seagrass shoot length) in the abundance and distribution of epiphytes on the Mediterranean seagrass Posidonia oceanica was investigated by means of a correlational approach over a spatial scale of ca. 500 km and at two different depths (5 and 15 m). Variability was examined from the double perspective of integrative community measures (biomass, species richness and alpha-diversity) and species composition. We assessed the influence of biotic and abiotic factors on integrative community measures using multiple correlation analyses. The influence of these factors in the structure of the whole community was investigated using a distance-based RDA for a linear model. A total of 129 taxa, mostly epiphytic algae, were recorded across all sites and depths. A large part of the variability in species composition (51%) was explained by the variables investigated. In particular, variability caused by differences in grazing pressure was the most important (25%), followed by variables related to nutrient availability (11%), meadow structure (6%), light (5%) and seagrass shoot length (4%). Among integrative community variables investigated, species richness was also best explained by grazing and nutrients.  相似文献

Trafficking of the vesicular acetylcholine transporter (VAChT) to synaptic vesicles has the potential to regulate storage and release of acetylcholine. We used the C-terminal tail of the vesicular acetylcholine transporter as bait for the screening of a brain cDNA library by yeast-two hybrids. Here we report an interaction uncovered in this screening with SEC14L1, a mammalian SEC14-like protein that may function as a phospholipid transfer protein. The interaction of VAChT and SEC14L1 occurred through the GOLD domain found in the latter and was confirmed in mammalian cells. In addition, we also found that SEC14L1 co-immunoprecipitates with the high affinity choline transporter (CHT1), but not with synaptophysin or synaptotagmin. In cultured cells SEC14L1 was predominantly found in the cytosol with little or no localization in defined organelles. In contrast, overexpression of VAChT or CHT1 with SEC14L1 recruited the latter to large intracellular organelles similar to vesicles or vesicle aggregates. Finally, we find that overexpression of SEC14L1 modestly decreases high affinity choline transport activity. We suggest that interaction of cholinergic transporters with proteins containing the GOLD domain may be relevant for transporter function.  相似文献

The decomposition of lipid hydroperoxides (LOOH) into peroxyl radicals is a potential source of singlet molecular oxygen ((1)O(2)) in biological systems. Recently, we have clearly demonstrated the generation of (1)O(2) in the reaction of lipid hydroperoxides with biologically important oxidants such as metal ions, peroxynitrite and hypochlorous acid. The approach used to unequivocally demonstrate the generation of (1)O(2) in these reactions was the use of an isotopic labeled hydroperoxide, the (18)O-labeled linoleic acid hydroperoxide, the detection of labeled compounds by HPLC coupled to tandem mass spectrometry (HPLC-MS/MS) and the direct spectroscopic detection and characterization of (1)O(2) light emission. Using this approach we have observed the formation of (18)O-labeled (1)O(2) by chemical trapping of (1)O(2) with anthracene derivatives and detection of the corresponding labeled endoperoxide by HPLC-MS/MS. The generation of (1)O(2) was also demonstrated by direct spectral characterization of (1)O(2) monomol light emission in the near-infrared region (lambda = 1270 nm). In summary, our studies demonstrated that LOOH can originate (1)O(2). The experimental evidences indicate that (1)O(2) is generated at a yield close to 10% by the Russell mechanism, where a linear tetraoxide intermediate is formed in the combination of two peroxyl radicals. In addition to LOOH, other biological hydroperoxides, including hydroperoxides formed in proteins and nucleic acids, may also participate in reactions leading to the generation (1)O(2). This hypothesis is currently being investigated in our laboratory.  相似文献

A research about arthropode fauna in laying hen excrement was carried out in a poultry house in the State of S?o Paulo, from January 2001 to December 2002. The objective was to verify the frequency of Diptera and Coleoptera collected by Berlese funil and flutuation methods, and to show correlation between the collect methods and sazonality possible in the currents seasons. A total of 29,499 insects were collected from seventy five collects, being 16,702 Diptera (six families) and 12,797 Coleoptera (four families). Some arthropodes presented direct relation with the collect method, the seasons of the years and the rainfall precipitation, with significant increase in the number of insects collected in certain periods of the year.  相似文献
