Elevated triglycerides (TG) contribute towards increased risk for cardiovascular disease. Lipoprotein lipase (LPL) is an enzyme that is responsible for the metabolism of core triglycerides of very-low density lipoproteins (VLDL) and chylomicrons in the vasculature. In this study, we explored the structure-activity relationships of our lead compound (C10d) that we have previously identified as an LPL agonist. We found that the cyclopropyl moiety of C10d is not absolutely necessary for LPL activity. Several substitutions were found to result in loss of LPL activity. The compound C10d was also tested in vivo for its lipid lowering activity. Mice were fed a high-fat diet (HFD) for four months, and treated for one week at 10 mg/kg. At this dose, C10d exhibited in vivo biological activity as indicated by lower TG and cholesterol levels as well as reduced body fat content as determined by ECHO-MRI. Furthermore, C10d also reduced the HFD induced fat accumulation in the liver. Our study has provided insights into the structural and functional characteristics of this novel LPL activator. 相似文献

Azadirachta indica (A. Juss) commonly known as Neem is an important source of valuable natural products and occupies an important place in traditional healthcare system. Naturally, this plant synthesizes a number of tetranortriterpenoids utilizing isoprenoid as substrate flux. Although various phytochemical and pharmacological studies in A. indica have been carried out, but very limited information is available about the biosynthetic pathway as well as structural and regulatory genes involved in synthesis of bioactive molecules. In this study, we have cloned and characterized two genes, AiHMGR1 and AiHMGR2, encoding 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR; EC 1.1.1.34) catalyzing the rate limiting step of the isoprenoid biosynthesis. Two isoforms, AiHMGR1 and AiHMGR2, contain an open reading frame of 1707 and 1695 bp encoding polypeptides of 568 and 545 amino acid residues, respectively. The nucleotide and encoded amino acid sequence analyses suggest that both genes encode polypeptides with necessary structural domains present in other plant HMGRs, however, have different genomic organization. The relative expression analysis suggests that two genes express differentially in various tissues. Out of the two genes, expression of AiHMGR2 showed a direct correlation with azadirachtin accumulation in fruit tissue. The common as well as unique cis-regulatory elements present in both genes might be responsible for differential expression of both the genes in various tissues. The color complementation assay in Escherichia coli suggests that though both AiHMGR1 and AiHMGR2 encode functional proteins, AiHMGR2 is more active as compared to AiHMGR1. 相似文献

Frankincense, the oleo‐gum resin of Boswellia species, has been an important element of traditional medicine for thousands of years. Frankincense is still used for oral hygiene, to treat wounds, and for its calming effects. Different Boswellia species show different chemical profiles, and B. carteri, in particular, has shown wide variation in essential oil composition. In order to provide insight into the chemical variability in authentic B. carteri oleoresin samples, a hierarchical cluster analysis of 42 chemical compositions of B. carteri oleo‐gum resin essential oils has revealed at least three different chemotypes, i) an α‐pinene‐rich chemotype, ii) an α‐thujene‐rich chemotype, and iii) a methoxydecane‐rich chemotype. 相似文献

Diabetic nephropathy is a complication of diabetes mellitus. This present study investigates the status of copper and magnesium
in diabetic nephropathy cases to establish a possible relation. Forty patients of diabetic nephropathy participated in the
study as cases. Forty age- and sex-matched healthy individuals served as controls. Blood samples were collected from both
cases and controls for determination of FBS, PPBS, HbA1c, microalbumin, copper, and magnesium levels. The mean concentrations
of FBS, PPBS, HbA1c, and microalbumin of cases were significantly higher than that of controls. The mean magnesium levels
of cases (1.60 ± 0.32 meq/L) were significantly lower than controls 2.14 ± 0.16 meq/L (p < 0.05). But the mean copper levels of cases, 165.42 ± 5.71 μg/dl, shows no significant difference with controls, 166.6 ± 5.48 μg/dl, (p > 0.05).The findings in the present study suggest that hypomagnesemia may be linked with development of diabetic nephropathy. 相似文献

We investigated whether sn-1,2-dioctanoylglycerol (diC8) activates highly purified human T cells. diC8's signaling activity was also compared with that of 12-O-tetradecanoylphorbol-13-acetate (TPA). diC8 and ionomycin were synergistic in promoting T-cell proliferation. The proliferative response was dependent upon an operational interleukin-2 (IL-2) system and exhibited a high degree of specificity; sn-1,2-diC8 was twice as active as racemic-1,2-diC8, and diC8 and TPA were not synergistic. diC8's signaling activity differed from that of TPA. diC8, unlike TPA, failed to elicit IL-2 receptors or proliferation, independently of ionomycin. diC8 also failed to promote the proliferation of T cells signaled with anti-CD3 or -CD2 monoclonal antibodies. Two different inhibitors of PKC, 1-(5-isoquinolinylsulfonyl)-2-methylpiperazine or staurosporine, inhibited T-cell proliferation induced with diC8 and ionomycin, but not with TPA and ionomycin. These observations, in addition to demonstrating the differential activity of diC8 and TPA, document a signaling role for diacylglycerol in the activation of normal T cells. 相似文献
