Intratracheal inoculation of young quail chicks with Aspergillus fumigatus spores resulted in the development of characteristic gross and microscopic lesions. The lesions were restricted to respiratory tract and there was no dissemination of infection to other tissues of the body.Gross changes in lungs and air sacs were observed within 24 hours and continued up to 20 days while in trachea these were noticed from the 3rd to the 9th day post-infection. The lesions, in general, included congestion and focal haemorrhages in the first 2 days followed by the development of varying-sized greyish-white nodules in the lungs, air sacs and trachea.Microscopic changes consisted of congestion, haemorrhages and a diffuse cellular infiltration in the first 2 days followed by granulomatous reaction with well developed granulomas in lungs, air sacs and trachea. Spores and developing hyphae of Aspergillus could be demonstrated in sections from 24 hours to 20 days of infection.Reisolation of the fungus was consistently achieved from the lungs, air sacs and trachea up to 14 days. 相似文献

Enteropathogenic Yersinia expresses several invasins that are fundamental virulence factors required for adherence and colonization of tissues in the host. Within the invasin‐family of Yersinia adhesins, to date only Invasin has been extensively studied at both structural and functional levels. In this work, we structurally characterize the recently identified inverse autotransporter InvasinE from Yersinia pseudotuberculosis (formerly InvasinD from Yersinia pseudotuberculosis strain IP31758) that belongs to the invasin‐family of proteins. The sequence of the C‐terminal adhesion domain of InvasinE differs significantly from that of other members of the Yersinia invasin‐family and its detailed cellular and molecular function remains elusive. In this work, we present the 1.7 Å crystal structure of the adhesion domain of InvasinE along with two Immunoglobulin‐like domains. The structure reveals a rod shaped architecture, confirmed by small angle X‐ray scattering in solution. The adhesion domain exhibits strong structural similarities to the C‐type lectin‐like domain of Yersinia pseudotuberculosis Invasin and enteropathogenic/enterohemorrhagic E. coli Intimin. However, despite the overall structural similarity, the C‐type lectin‐like domain in InvasinE lacks motifs required for Ca2+/carbohydrate binding as well as sequence or structural features critical for Tir binding in Intimin and β1‐integrin binding in Invasin, suggesting that InvasinE targets a distinct, yet unidentified molecule on the host‐cell surface. Although the biological role and target molecule of InvasinE remain to be elucidated, our structural data provide novel insights into the architecture of invasin‐family proteins and a platform for further studies towards unraveling the function of InvasinE in the context of infection and host colonization. 相似文献

A fractal analysis of a confirmative nature only is presented for cellular analyte-receptor binding kinetics utilizing biosensors. Data taken from the literature can be modeled by using a single-fractal analysis. Relationships are presented for the binding rate coefficient as a function of the fractal dimension and for the analyte concentration in solution. In general, the binding rate coefficient is rather sensitive to the degree of heterogeneity that exists on the biosensor surface. It is of interest to note that examples are presented where the binding coefficient, k exhibits an increase as the fractal dimension (D(f)) or the degree of heterogeneity increases on the surface. The predictive relationships presented provide further physical insights into the binding reactions occurring on the surface. These should assist in understanding the cellular binding reaction occurring on surfaces, even though the analysis presented is for the cases where the cellular "receptor" is actually immobilized on a biosensor or other surface. The analysis suggests possible modulations of cell surfaces in desired directions to help manipulate the binding rate coefficient (or affinity). In general, the technique presented is applicable for the most part to other reactions occurring on different types of biosensor or other surfaces. 相似文献

Biol. Chem. 272 (1997) 11384], and for the cooperative binding of human 1,25-dihydroxyvitamin D(3) receptor (VDR) to DNA with the 9-cis-retinoic acid receptor (RXR) [Biochemistry 35 (1996) 3309]. Ligands were also used to modulate the first reaction. Data taken from the literature may be modeled by using a single- or a dual-fractal analysis. Relationships are presented for the binding rate coefficient as a function of either the analyte concentration in solution or the fractal dimension that exists on the biosensor surface. The binding rate expressions developed exhibit a wide range of dependence on the degree of heterogeneity that exists on the surface, ranging from sensitive (order of dependence equal to 1.202) to very sensitive (order of dependence equal to 12.239). In general, the binding rate coefficient increases as the degree of heterogeneity or the fractal dimension of the surface increases. The predictive relationships presented provide further physical insights into the reactions occurring on the biosensor surface. Even though these reactions are occurring on the biosensor surface, the relationships presented should assist in understanding and in possibly manipulating the reactions occurring on cellular surfaces. 相似文献

Resveratrol, a polyphenol found in fruits, possesses chemopreventive and chemotherapeutic properties and has been shown to increase lifespan in yeast and metazoans, including mice. Genetic evidence and in vitro enzymatic measurements indicate that the deacetylase Sir2/SIRT1, an enzyme promoting stress resistance and aging, is the target of resveratrol. Similarly, down-regulation of insulin-like pathways, of which PI3K (phosphoinositide 3-kinase) is a key mediator, promotes longevity and is an attractive strategy to fight cancer. We show here that resveratrol inhibits, in vitro and in cultured muscle cell lines, class IA PI3K and its downstream signalling at the same concentration range at which it activates sirtuins. Our observations define class IA PI3K as a target of resveratrol that may contribute to the longevity-promoting and anticancer properties and identify resveratrol as a natural class-specific PI3K inhibitor. 相似文献

Understanding of the mechanisms of Mn supply from the soil and uptake by the plants can be improved by using simulation models
that are based on basic principles. For this, a pot culture experiment was conducted with a sandy clay loam soil to measure
Mn uptake by summer wheat (Triticum aestivum L. cv. Planet), maize (Zea mays L. cv. Pirat) and sugar beet (Beta vulgaris L. cv. Orbis) and to simulate Mn dynamics in the rhizosphere by means of a mechanistic model. Seeds of three crops were sown
in pots containing 2.9 kg soil in a controlled growth chamber. Root and shoot weight, Mn content of plants, root length and
root radius were determined 8 (13 days in case of sugar beet) and 20 days after germination. Soil and plant parameters were
determined to run nutrient uptake model calculations. Manganese content of the shoot varied from 25 mg kg-1 for sugar beet to 34 mg kg-1 for maize. Sugar beet had the lowest root length/shoot weight ratio but the highest relative shoot growth rate, resulting
in the highest shoot demand on the root. This is reflected by the Mn influx which was 0.9 × 10-7, 1.7 × 10-7 and 2.5 × 10-7 nmol cm-1 s-1 for wheat, maize and sugar beet, respectively. Nutrient uptake model calculations predicted similar influx values. Initial
Mn concentration of 0.2 μM in the soil solution decreased to only 0.16 μM for wheat, 0.13 μM for maize and 0.11 μM for sugar beet at the root surface. This shows that manganese transport to the root was not a limiting step. This was confirmed
by the fact that an assumed 20 times increase in maximum influx (Imax) increased the calculated Mn influx by 3.7 times. Sensitivity analysis demonstrated that for controlling Mn uptake the initial
soil solution concentration (C
Li), the root radius (r0), Imax and the Michaelis constant (K
m) were the most sensitive factors in the listed order.
