This study examined the dynamics of the middlecerebral artery (MCA) blood flow response to hypocapnia in humans(n = 6) by using transcranial Dopplerultrasound. In a control protocol, end-tidalPCO2(PETCO2) was heldnear eucapnia (1.5 Torr above resting) for 40 min. In ahypocapnic protocol, PETCO2was held near eucapnia for 10 min, then at 15 Torr below eucapnia for20 min, and then near eucapnia for 10 min. During both protocols,subjects hyperventilated throughout andPETCO2 and end-tidalPO2 were controlled by using thedynamic end-tidal forcing technique. Beat-by-beat values werecalculated for the intensity-weighted mean velocity (IWM),signal power (), and theirinstantaneous product(IWM).A simple model consisting of a delay, gain terms, time constants(f,on, f,off) and baseline levels offlow for the on- and off-transients, and a gain term(gs) and time constant(s) for a second slower component was fitted to the hypocapnic protocol. The cerebral bloodflow response to hypocapnia was characterized by a significant (P < 0.001) slowprogressive adaptation inIWM, with gs = 1.26 %/Torr ands = 427 s, that persistedthroughout the hypocapnic period. Finally, the responses at the onsetand relief of hypocapnia were asymmetric(P < 0.001), withf,on (6.8 s) faster thanf,off (14.3 s).

A GnRH antagonist (Antarelix) treatment was used during the breeding season of Romanov ewes, to investigate whether LH pulses are required the day before the preovulatory surge for normal early embryo development in vivo (Expt 1) and in vitro (Expt 2). In Expt 1, at the onset of oestrus after removal of a fluorogestone acetate sponge, group A0.5 (n = 22) received a subcutaneous injection of 0.5 mg Antarelix, and ovulation was induced with an intravenous injection of 3 mg pig LH 24 h later. The control group (group C, n = 20) were untreated. All ewes were mated naturally at 36 and 48 h after oestrus and embryos were recovered 8 days after sponge removal. There were significant differences in the decrease in LH and in the increase in FSH concentration after Antarelix treatment between treated and control groups. The ovulation rate and embryo recovery rate were not significantly different between the two groups but the blastocyst rate was lower (P < 0.0001) in group A0.5 than in group C, with more unfertilized or degenerated oocytes in group A0.5 (69.2%). In Expt 2, 24 h after sponge removal, group A (n = 10) and group B (n = 10) received one subcutaneous injection of 0.5 mg Antarelix. The control group (group C, n = 10) was left untreated. LH pulsatility was re-established in group B with hourly intravenous injections of 5 micrograms ovine LH for 24 h. Oocytes were collected by flushing the oviducts 28 h after the LH surge, and were fertilized and cultured in vitro for 7 days. Ovulation and cleavage rates were not significantly different among the three groups but a higher rate of blastocysts (P < 0.01) was obtained after Antarelix treatment when LH pulsatility was re-established (group B). Oestradiol concentration was strongly depressed (P < 0.0003) after Antarelix treatment in group A, but was maintained after injection of LH pulses in group B, although at a lower value than before the preovulatory surge in the control group. In conclusion, inhibition of endogenous LH pulses 1 day before the preovulatory surge was not essential for ovulation and in vitro fertilization but was associated with a decrease in plasma oestradiol concentrations and inferior embryo development both in vivo and in vitro. When LH pulsatility was re-established, oestradiol concentrations increased and embryo development was restored.  相似文献

Chondracanthid copepods parasitise many teleost species and have a mobile larval stage. It has been suggested that copepod parasites, with free-living infective stages that infect hosts by attaching to their external surfaces, will have co-evolved with their hosts. We examined copepods from the genus Chondracanthus and their teleost hosts for evidence of a close co-evolutionary association by comparing host and parasite phylogenies using TreeMap analysis. In general, significant co-speciation was observed and instances of host switching were rare. The prevalence of intra-host speciation events was high relative to other such studies and may relate to the large geographical distances over which hosts are spread.  相似文献

The levels of trypanothione, a glutathione-spermidine conjugate, are increased in the protozoan parasite Leishmania selected for resistance to the heavy metal arsenite. The levels of putrescine and spermidine were increased in resistant mutants. This increase is mediated by overexpression of ornithine decarboxylase (ODC), the rate-limiting enzyme in polyamine biosynthesis. Gene overexpression is generally mediated by gene amplification in Leishmania but, here, the mRNA and the enzymatic activity of ODC are increased without gene amplification. This RNA overexpression is stable when cells are grown in the absence of the drug and does not result from gene rearrangements or from an increased rate of RNA synthesis. Transient transfections suggest that mutations in the revertant cells contribute to these elevated levels of RNA. Stable transfection of the ODC gene increases the level of trypanothione, which can contribute to arsenite resistance. In addition to ODC overexpression, the gene for the ABC transporter PGPA is amplified in the mutants. The co-transfection of the ODC and PGPA genes confers resistance in a synergistic fashion in partial revertants, also suggesting that PGPA recognizes metals conjugated to trypanothione.  相似文献

1. Positive correlations between local abundance and distribution on a larger spatial scale are commonly observed among related species.
2. Within parasite species, the same relationship may be expected between prevalence and intensity of infection across host species used. Across parasite species, a positive relationship is expected between average abundance in a host population and the number of host species that can be exploited based on the resource breadth hypothesis. Trade-offs between the ability to exploit many host species and the potential for heavy infections, however, could result in a negative relationship.
3. Intraspecifically, using data on 51 helminth species parasitic in birds, prevalence and intensity of infection among host species used are generally only weakly correlated. Only in nematodes is there an overall positive relationship between prevalence and intensity.
4. A comparative analysis was performed on data from 389 species of cestodes, trematodes and nematodes parasitic in birds to determine how host specificity covaries interspecifically with abundance, measured both as prevalence and intensity of infection.
5. After controlling for phylogenetic influences and sampling effort, the number of host species used correlated positively with prevalence in all three parasite taxa, and with intensity of infection in trematodes only.
6. These results do not support the existence of a trade-off between abundance and the use of many host species, as has been found for fish parasites. Instead, whatever makes helminth parasites of birds abundant within a host population may facilitate their successful colonization of new host species.  相似文献

Alterations in host phenotype induced by parasitic infection are often interpreted as either host or parasite adaptations, depending on which of the two appears to benefit. Mermithid nematodes typically castrate their insect hosts and, therefore, any change in host behavior has no further fitness consequences for the host; the adaptive value of the modified behavior must be assessed with respect to parasite fitness only. In a New Zealand stream, mermithid-infected nymphs of mayflies in the genus Deleatidium were disproportionately represented in drift samples compared with benthic samples, suggesting that infection by mermithids results in an increased tendency to drift. Drifting mayflies face a higher predation risk from trout, and the mermithid nematodes they harbor die if ingested by a fish. The change in mayfly behavior induced by mermithids thus appears to have negative fitness effects for the parasite, and one possible explanation for this phenomenon is that it is a nonadaptive, pathological side effect of infection.  相似文献

Almost every animal trait is strongly associated with parasitic infection or the potential exposure to parasites. Despite this importance, one of the greatest challenges that researchers still face is to accurately determine the status and severity of the endoparasitic infection without killing and dissecting the host. Thus, the precise detection of infection with minimal handling of the individual will improve experimental designs in live animal research. Here, we quantified extracellular DNA from two species of endoparasitic worm that grow within the host body cavity, hairworms (phylum Nematomorpha) and mermithids (phylum Nematoda), from the frass of their insect host, a cave wētā (Orthoptera: Rhaphidophoridae) and an earwig (Dermaptera: Forficulidae), respectively. Frass collection was done at two successive time periods, to test if parasitic growth correlated with relative DNA quantity in the frass. We developed and optimized two highly specific TaqMan assays, one for each parasite-specific DNA amplification. We were able to detect infection prevalence with 100% accuracy in individuals identified as infected through post-study dissections. An additional infection in earwigs was detected with the TaqMan assay alone, probably because some worms were either too small or degraded to observe during dissection. No difference in DNA quantity was detected between sampling periods, although future protocols could be refined to support such a trend. This study demonstrates that a noninvasive and minimally stressful method can be used to detect endoparasitic infection with greater accuracy than dissection alone, helping improve protocols for live animal studies.  相似文献

In Préalpes de Sud ewes after an autumn lambing, the mean post-partum interval to first LH surge was 10 +/- 1 days and 17 +/- 1 days for non-suckling and suckling ewes, respectively. Post-partum interval to first luteal phase, estimated from plasma progesterone concentrations, was similar in non-suckling and suckling ewes (27 +/- 1 days and 28 +/- 5 days, respectively). Interval to first oestrus was shorter in non-suckling (22 +/- 2 days) than in suckling ewes (35 +/- 2 days) but these first oestrous periods were followed by short luteal phases in 60% (12/20) of non-suckling ewes and in only 7% (2/29) of suckling ewes. Finally, suckling slightly postponed the resumption of the first oestrus followed by a normal oestrous cycle (37 +/- 1 days versus 31 +/- 2 days) because progesterone, essential for oestrus expression, was secreted mainly during normal luteal phases in 70% (21/30) of suckling ewes and during short cycles in 95% (21/22) of non-suckling ewes. Therefore, the primary consequence of suckling is to regulate the conditions of resumption of cyclic ovarian activity after parturition.  相似文献

Aim The ‘abundance optimum’ hypothesis predicts that species abundance peaks in the locality with the most favourable conditions and decreases with an increase of distance from that locality. We tested this prediction for 9 fleas and 13 gamasid mite species. Location We used published data on fleas and gamasid mites that are parasitic on small mammals throughout the Palaearctic. Methods For each ectoparasite, we computed the correlation between the relative abundance on its principal host species in a region and the distance from that region to the region of maximum abundance. Then, the correlation coefficients were used in a meta‐analysis. We also made a cross‐species comparison between relative abundances in localities (a) closest to and (b) furthest from the locality of maximum abundance. Results Although the relationship between the relative abundance in a region and the distance from that region to the region of maximum abundance was negative in 19 out of 22 ectoparasites, it was only statistically significant in three of them. However, a meta‐analysis of coefficients of correlations across all species revealed a significant negative effect of the distance from the region of maximum abundance on relative abundance in a particular region. A cross‐species comparison between relative abundances in the localities closest to and furthest from the locality of maximum abundance demonstrated that the former were significantly higher than the latter. Main conclusions A lack of strict host specificity in the ectoparasites studied, and the absence of any strong spatial correlations among the environmental variables affecting ectoparasite reproduction and abundance, may provide an explanation for the spatial independence in abundance values of most species. However, a preference for a particular host even in host‐opportunistic parasites combined with species‐specific environmental preferences could be the reason behind the weak, but significant, negative abundance–distance relationship across species. The contradiction between results obtained when separate species were considered and when the overall pattern was analysed across species suggests that there exists a general underlying spatial pattern that can often be masked by other factors.  相似文献
