Variation in food availability impacts the performance of insects in terms of their size and age to maturity and fecundity. Age at maturity determines how quickly individuals in a population can start to reproduce and how much they can reproduce. Results from studies on various insect species show that food availability influences the size and fecundity of adult females. It is predicted that under poor growth conditions, variation in size is low, but variation in age at maturity is considerable. This prediction was examined in a widely distributed lady beetle species, Harmonia axyridis (Pallas) (Coleoptera: Coccinellidae), a predator of aphids and coccids. Using a food gradient from low to high aphid prey density, performance of females that were reared on excess food was recorded for pre‐reproductive duration, size at reproductive maturity, number of aphids consumed, and fecundity in the first 10 days of their reproductive period. Results suggested that female H. axyridis that were reared on surplus food when kept at low prey density (poor growth condition) took, on average, three times longer to attain maturity and produced, on average, 14 times fewer eggs than females that were also reared on surplus food, but kept at high prey density (good growth condition). Females performed best at a prey density of 30 aphids per female per 150 cm2. Results suggested that the current food availability significantly influenced the age and size of females at maturity and their fecundity. Age and size at maturity of female lady beetles showed non‐linear responses to prey density as well as the occurrence of a minimum size of females, below which H. axyridis females fail to mature. The steep slope recorded at lower prey densities suggests relatively high variation in age at maturity but low variation in size.  相似文献

The bacterial replicative helicases known as DnaB are considered to be members of the RecA superfamily. All members of this superfamily, including DnaB, have a conserved C- terminal domain, known as the RecA core. We unearthed a series of mycobacteriophage encoded proteins in which the RecA core domain alone was present. These proteins were phylogenetically related to each other and formed a distinct clade within the RecA superfamily. A mycobacteriophage encoded protein, Wildcat Gp80 that roots deep in the DnaB family, was found to possess a core domain having significant sequence homology (Expect value < 10-5) with members of this novel cluster. This indicated that Wildcat Gp80, and by extrapolation, other members of the DnaB helicase family, may have evolved from a single domain RecA core polypeptide belonging to this novel group. Biochemical investigations confirmed that Wildcat Gp80 was a helicase. Surprisingly, our investigations also revealed that a thioredoxin tagged truncated version of the protein in which the N-terminal sequences were removed was fully capable of supporting helicase activity, although its ATP dependence properties were different. DnaB helicase activity is thus, primarily a function of the RecA core although additional N-terminal sequences may be necessary for fine tuning its activity and stability. Based on sequence comparison and biochemical studies we propose that DnaB helicases may have evolved from single domain RecA core proteins having helicase activities of their own, through the incorporation of additional N-terminal sequences.  相似文献

Littleseed canarygrass (Phalaris minor Retz.) is a major weed in wheat fields, and has developed resistance to the commonly used herbicide isoproturon. This study explores the potential use of isoxaflutole, a pre-emergence herbicide, to control littleseed canarygrass. Greenhouse studies were carried out to determine the phytotoxicity of isoxaflutole in relation to shoot height, fresh shoot biomass and leaf chlorophyll concentration of wheat and littleseed canarygrass. Electron microscopy was used to examine any damage to leaf chloroplast at ultrastructural level. Results indicate that isoxaflutole (0.5 mg/L) significantly reduced the shoot height of littleseed canarygrass (39.6%), but no significant reduction in the shoot height of wheat was observed (9.6%) when compared to control. None of the concentrations (0.05, 0.1, 0.5 and 1 mg/L) of isoxaflutole altered soil chemistry in relation to pH, organic matter, macro or micro inorganic ions. While untreated littleseed canarygrass leaves had elongated chloroplast, starch grains and small number of plastoglobuli; treated littleseed canarygrass leaves had swollen chloroplast, large number of plastoglobuli, and a lack of starch grains. We conclude that isoxaflutole can be an effective herbicide for controlling littleseed canarygrass.  相似文献

An efficient microprogation protocol has been developed for Dendrobium densiflorum Lindl. ex Wall., a traditional medicinal plant, through protocorm-like bodies (PLBs) from nodal stem segments using 6-benzylamino-purine (BAP) and the lanthanoid neodymium. The highest percentage of explants producing PLBs (72%), with an average of 15 PLBs per explant, was induced by culturing stem segments on Murashige and Skoog (MS) medium supplemented with 5.0 mg l−1 BAP. The newly formed PLBs proliferated well on the basal MS medium and completely converted into shoots on MS medium containing 2.0 mg l−1 BAP. Shoots produced an average of 22 roots per plantlet when cultured on MS medium supplemented with 2.0 mg l−1 neodymium nitrate. Healthy plantlets with well-developed roots were successfully acclimatized. The obtained result suggests that the lanthanoids can be used to effectively initiate rooting in the micropropagation and conservation of D. densiflorum.  相似文献

The photoelectrochemical and spectral studies of riboflavin have been carried out in aqueous and phosphatidylcholine (PC) liposome media in presence of different reducing agents such as I-, Br-, Cl-, Fe2+, Fe(CN)6(4-) and Cu+. The results from both the studies support the photoinduced electron transfer from the reducing agent to the excited riboflavin dye. Moreover, a good correlation between photovoltages/Stern-Volmer quenching constants versus reduction potentials of the reducing agents also confirms the above electron transfer in the photoexcited state. An alternative method has been developed to determine the Stern-Volmer quenching constant.  相似文献

Summary In the type-strainLactobacillus acidophilus ATCC 4356 -galactosidase (-gal) was inducible; lactose, galactose, melibiose and probably maltose, but not glucose, fructose, mannose, sucrose and cellobiose, induced -gal synthesis. Glucose partially inhibited -gal-induction by lactose but not by isopropyl--D-thiogalactoside. -gal synthesis during cell growth was maximal at 0.4% lactose, stimulated by Ca2+ but inhibited by Mg2+ and Mn2+. -gal in the cell-free extract had optimum activity at pH 6.5 and at 45°C. The enzyme activity was stimulated by Mg2+, inhibited by Ca2+, destroyed by oxidizing agents and protected by reducing agents.  相似文献

Synovial fluid samples of goat kids inoculated (ip) with 5 ml of 48 hr log phase culture of Mycoplasma mycoides sub sp. mycoides (large colony type) containing 10(7) cfu/ml were analysed for physical, cytological and biochemical properties. The synovial effusions were exudative in nature with increased volume. Gross appearances were serofibrinous, haemorrhagic and turbid containing flocculent materials with immediate clot formation. Mucinous precipitate quality was very poor having friable precipitates with cloudy supernatant. There were high total leucocytic and erythrocytic counts with significant high numbers of both neutrophils and lymphocytes. Synovial fluid sugar contents were significantly reduced, whereas total protein contents were significantly increased with concomitant reduction in albumin:globulin ratio. The alkaline phosphatase and transaminase values were also markedly increased in the synovial fluids of mycoplasma induced polyarthritic goat kids. The results may provide a clinical guideline for diagnosis, chemotherapy and prognosis of different joint diseases in domesticated animals.  相似文献

An efficient method for gene replacement in Lactobacillus helveticus CNRZ32 was developed by utilizing pSA3 as an integration vector. This plasmid is stably maintained in CNRZ32 at 37 degrees C but is unstable at 45 degrees C. This method consisted of a two-step gene-targeting technique: (i) chromosomal integration of a plasmid carrying an internal deletion in the gene of interest via homologous recombination and (ii) excision of the vector and the wild-type gene via homologous recombination, resulting in gene replacement. By using this procedure, the chromosomal X-prolyl dipeptidyl aminopeptidase gene (pepXP) of CNRZ32 was successfully inactivated.  相似文献
