In the preceding article, we described physicochemical and kinetic properties of parathyroid hormone (PTH) receptors in clonal rat osteosarcoma cells (ROS 17/2.8) using photoaffinity ligand labeling and showed that the physiologically relevant receptor-ligand complex has an apparent Mr = 80,000. In this study, the photoaffinity labeled Mr = 80,000 receptor was localized exclusively on the cell surface plasma membrane and its glycoprotein nature was demonstrated through the use of lectin affinity-chromatography and specific exo- and endoglycosidases. Rinsing ROS cells, preincubated in the dark with 125I-labeled [Nle8, N-epsilon-(4-azido-2-nitrophenyl)Lys13,Nle18,Tyr34]bovine PTH-(1-34)-NH2 (NAP-NlePTH) (4 h, 15 degrees C, equilibrium conditions) with acidic phosphate-buffered saline (pH 2.5, 30 s, 4 degrees C) before photolysis resulted in selective and nearly total disappearance of the labeled Mr = 80,000 receptor. PTH receptor integrity to acid rinsing and photolysis was shown by relabeling the Mr = 80,000 receptor after a second incubation of these cells with 125I-labeled NAP-NlePTH, followed by photolysis. Adsorption of Triton X-100-solubilized, 125I-labeled NAP-NlePTH receptors to wheat germ agglutinin-agarose is nearly complete and highly selective, and elution with N-acetylglucosamine resulted in virtually total recovery of the labeled receptors from the column. The wheat germ agglutinin-retarded PTH receptors show increased electrophoretic mobility upon treatment with neuraminidase which was inhibited by simultaneous addition of 2,3-dehydro-3-desoxy-N-acetylneuraminic acid, a specific neuraminidase inhibitor. Endoglycosidase F treatment of the Mr = 80,000 receptors generated a single, labeled polypeptide with a Mr = 59,000 which migrated as a narrow band. PTH receptors on ROS 17/2.8 cells appear to be monomeric plasma membrane glycoproteins with an apparent Mr of 80,000 which contain a Mr = 59,000 polypeptide backbone and a polymeric arrangement of N-acetylglucosamine with N-acetylneuraminic acid as major terminal sugar residues.  相似文献

Pea plants ( Pisum sativum L.) possessing the slender phenotype (conferred by the gene combination la crys ) have extremely long, thin internodes and are phenotypically similar to dwarf plants (possessing genes La and/or Cry ) that have been treated with a non-limiting dose of gibberellin (GA3). In contrast to tall and dwarf plants, slender plants are virtually insensitive to treatment with AMO 1618, PP333 or GA3 and addition of the "gibberellin-less" mutant gene na does not alter the phenotype of slender plants. Na slender segregates possessed lower levels of gibberellin-like substances than comparable dwarf segregates when extracts from shoots were assayed using the lettuce hypocotyl or rice seedling bioassays. In addition, na slenders possessed little or no gibberellin-like activity even though they possessed a slender phenotype. Thus the gene combination la crys causes slender plants to respond as if they are saturated with gibberellins for growth. In addition, the gene combinations la crys and le la cryc (allele cryc is less extreme in effect than crys ) are shown to be almost completely epistatic to the alleles at the na locus. All these results suggest that gibberellin levels are not important in determining the internode length of slender peas (genotype la crys ). The possible mechanisms by which this could occur are discussed.  相似文献

RNA pools were extracted from cells of Nostoc commune UTEX 584 in exponential growth (liquid cultures) and from cells which had been immobilized and dried rapidly at -99.5 MPa. Levels of incorporation of 35S-methionine, five- to sixfold higher than the endogenous level, were obtained after in vitro translation of the RNA preparations in a heterologous S30 cell-free system purified from Escherichia coli Q13. The levels of incorporation, obtained with a homologous N. commune UTEX 584 S30 system, were much lower. The requirement for magnesium in the heterologous system was 15–21 mM, translation of N. commune UTEX 584 RNA was inhibited when the RNA concentration was greater than 0.3 mg ml–1, and translation was stimulated significantly by the presence of ammonium chloride. Few qualitative differences were observed between the pattern of proteins (SDS-PAGE) obtained after translation of the RNA pools from cells in exponential growth, and from those cells subjected to immobilization and rapid drying. The data suggest that short-term desiccation of N. commune UTEX 584 does not have a marked selective effect on the composition of the mRNA pool. In contrast, preparations of RNA from field materials of Nostoc commune HUN (desiccated for 5 years) were unable to drive high rates of translation in any of the systems tested and optimized for use in this study.  相似文献

Summary Immobilised, desiccated cells ofNostoc commune UTEX 584 have the capacity to increase the size of their extractable intracellular ATP pool upon rewetting. The time taken to recover the pool size depends on the conditions of storage at a particular water potential and the duration of storage. Under the conditions employed, the rewetting of cells induced an increase in ATP pool size at the expense of photophosphorylation or electron transport (oxidative) phosphorylation. The rise in the ATP pool size was instantaneous and was shown to be due to ATP synthesis. This increase did not occur when cells were rewetted in the presence of sodium azide (10 mmol/l), while a partial inhibition was observed with CCCP (carbonyl cyanidem-chlorophenylhydrazone; 2 mol/l). For cells dried at more extreme water potentials, the lag ofc 48 h observed before the ATP pool reached control values is of similar duration to that observed in the recovery of nitrogenase upon rewetting. Chloramphenicol (10 mol/l) stimulated significantly the upshift in the size of the ATP pool ofNostoc cells upon rewetting, yet inhibited completely the rise in nitrogenase activity.  相似文献

Recently it has been recognized that IL-1 plays an important role in hematopoietic regulation. Administration of 5-fluorouracil (5-FU) to mice causes prolonged neutropenia. rHIL-1 injected to mice after 5-FU, accelerated the recovery of hematopoietic progenitors and blood neutrophils. The combination of rhIL-1 and rhG-CSF reduced the neutropenic period significantly. Sublethal irradiation of mice induced profound neutropenia for 3 weeks which was associated with 80% mortality. Administration of rhIL-1 20 hours prior to or 2 hours post irradiation resulted in a significantly improved survival and rapid recovery of the neutrophil count. IL-1 administered alone or in combination with other colony stimulating factors to spontaneous breast tumor bearing mice following 5-FU therapy resulted in a rapid recovery of neutrophils, improved survival, and markedly reduced the tumor mass. Experiments in primates demonstrated that rhIL-1 administered to 5-FU treated animals shortened the neutropenic period from 30 to 17 days and increased the number of marrow progenitors responsive to other CSFs. Prolonged administration of IL-1 (14 days) to these animals resulted in a delayed neutrophil recovery as compared to animals receiving short courses of IL-1. rhIL-1 administered to primates receiving marrow grafts after lethal irradiation, did not result in rapid hematopoietic recovery. In humans, studies with CD-34 positive marrow cells showed that IL-1 had a radioprotective effect on a committed and early marrow progenitors. These data show the therapeutic potential of IL-1 in the treatment of chemoradiotherapy induced myelosuppression.  相似文献

There are three main types of plumage in the Shag, the juvenal which is brown, the post-juvenal which is dark brown and the nuptial which is black-green. The replacement of these plumages and the ontogeny of the Staffelmauser was studied in a sample of 566 Shags mainly from northeast England. The replacement of the juvenal primaries starts at the age of eight months with the loss of the innermost primary and continues outwards at a rate of about one primary per 17 days. Another cycle starts when the first reaches the 8th primary but both cycles soon pause for the winter. Successive cycles are established annually in August or September in this way, so that cycles which take longer than one year to complete result eventually in a Staffelmauser in which each feather is replaced once annually. Most individual Shags in a breeding population will retain feathers from two or three cycles, but a large sample of breeding adults of the same age will retain the feathers of four cycles. The winter pause is considered an adaptation to the increased probability of adverse conditions at this time. The system described for the ontogeny of the Staffelmauser is applicable to those other sea-birds which have been studied in detail, and is probably advantageous since it combines slow moult during the difficult early years with a highly efficient moult during the breeding years, especially where moult and breeding occur together.  相似文献

A meridic diet for culturing the coffee bean weevil, Araecerus fasciculatus was formulated and freeze-dried into pellets. When exposed to an atmosphere of 80 ± 5%r.h. the diet reached a moisture equilibrium that invoked oviposition and was satisfactory for growth and development of the insect. Larvae ingested larger amounts of the freeze-dried diet than of the natural food, green coffee beans, reaching equivalent weight and pupation in 31.8 ± 0.4 days on the diet as compared to 46.8 ± 0.8 days on the coffee beans. Approximate digestibilities of the two foods were not significantly different. However, the coffee beans were converted more efficiently to body mass than was the diet. Faeces of larvae on the diet contained more uric acid than those of larvae on coffee beans. Continuous culture and mass rearing of A. fasciculatus on the diet is possible.  相似文献

Four strains of Chroococcidiopsis and one Chroococcus, all isolated from extreme arid desert rocks, and one marine Chroococcus, were subjected to water stress using both matric and osmotic control methods. For all Chroococcidiopsis strains, photosynthetic rates decreased with decreasing water potential. After 24h preincubation the decrease was linear but after 72h there was a sharp drop below-3400 kPa (a w0.976). In contrast, the two Chroococcus strains showed optimum photosynthesis between-3000 and-4000 KPa. It appears, therefore, that Chroococcidiopsis in deserts may have a different survival strategy in response to aridity than Chroococcus (rare in deserts).Absolute rates of 14CO2 uptake were higher in matric than in osmotic control systems. It is suggested that, in a matric experimental system, the water status is more representative of the natural conditions in arid environments.The consistent differences between different strains in their response to water stress suggest that this character in Cyanobacteria may be of taxonomic significance.  相似文献
