The reproductive biology, growth and diet of Galaxiella munda McDowall in a south-western Australian river are described. Monthly trends in gonadosomatic indices, stages in ovarian development and the size and maturity of oocytes show that spawning extended from July to October and peaked in late August to early September. Histology of ovaries indicated that G. munda produced clutches of eggs which it released at intervals. Data on length-frequencies, otoliths and gonads demonstrate that G. munda typically died in the few months after spawning. By age I, the females and males of G. munda had reached 47 mm (≡ 0.54 g) and 43 mm (≡ 0.42 g), respectively. The respective von Bertalanffy growth curve parameters for L., K and to were 48.6 mm, 3.702 and -0.0014 for females and 44.3 mm, 4.217 and -0.0012 for males. Galaxiella munda fed predominantly on terrestrial fauna on the water surface, cladocerans and copepods in the water column, and dipteran larvae in the benthos. Comparisons are made between the above aspects of the biology of G. munda and those recorded for three other locally endemic species ( Galaxias occidentalis Ogilby, Bostockia porosa Castelnau and Edelia vittata Castelnau) and wherever possible with other Galaxiella species. In current models, protein translocation in the endoplasmic reticulum (ER) occurs in the context of two cycles, the signal recognition particle (SRP) cycle and the ribosome cycle. Both SRP and ribosomes bind to the ER membrane as a consequence of the targeting process of translocation. Whereas SRP release from the ER membrane is regulated by the GTPase activities of SRP and the SRP receptor, ribosome release from the ER membrane is thought to occur in response to the termination of protein synthesis. We report that ER-bound ribosomes remain membrane-bound following the termination of protein synthesis and in the bound state can initiate the translation of secretory and cytoplasmic proteins. Two principal observations are reported. 1) Membrane-bound ribosomes engaged in the synthesis of proteins lacking a signal sequence are released from the ER membrane as ribosome-nascent polypeptide complexes. 2) Membrane-bound ribosomes translating secretory proteins can access the translocon in an SRP receptor-independent manner. We propose that ribosome release from the ER membrane occurs in the context of protein translation, with release occurring by default in the absence of productive nascent polypeptide-membrane interactions. 相似文献

Five chemotherapeutic agents which inhibit mitosis caused an effect on the colchicine bound to the isolated tubulin from mouse lymphoma L5178Y cells. These effects were examined over a time course of 4, 12, 24, 48, and 72 hours after a single administration of each drug. Vinblastine, oncodazole, and bleomycin decreased the amount of colchicine bound per mg protein; procarbazine and chloroambucil increased the amount bound. All of the drugs except procarbazine required more than 4 hours to cause an effect on colchicine binding and in the case of bleomycin and oncodazole some recovery occurred after 48 hours. The mitotic index was affected by 4 hours by all drugs: procarbazine, chlorambucil and bleomycin caused a decrease; vinblastine and oncodazole, an increase. 相似文献

Aim
Invasive species occurrence is often related to the anthropogenic context of a given area. Quantifying the effects of roads is of particular interest as roads are a major vector for invasion. Our objective was to further quantify the effects of roads on forest plant invasion through a macroscale, high‐resolution investigation to assist effective invasion control and mitigation.Location
Eastern United States.Methods
Using invasive plant data from 23,039 forest inventory plots in 13 ecological provinces, we employed logistic regression to relate the odds of invasion to distance from a road, with adjustments for broadscale differences attributable to ecological provinces, and local scale differences in productivity, forest fragmentation and land use.Results
The overall proportion (P) of invaded plots was 0.58 (0.65 for plots within 50 m of a road), and the highest odds (P/1 ? P) of invasion were found in relatively more productive, fragmented forest in landscapes with more than 10% agriculture or developed land cover. Wald chi‐square statistics indicated the best predictor of the odds of invasion was ecological province, followed by land use, productivity, forest fragmentation and distance from a road. Depending on the province, the adjusted odds of invasion decreased by up to 23% (typically 4%–10%) per 100 m distance from a road. The adjusted probability of invasion approached zero in only three provinces, for the least productive, least fragmented forest that was at least 2,000 m from a road in landscapes with less than 10% agricultural or developed land cover.Main conclusions
In the eastern United States, the existence of a nearby road is less important than the landscape context associated with the road. A purely road‐mediated effect has little practical meaning because anthropogenic activities and roads are pervasive and confounded.28.
The chloroplast enzyme phosphoribulokinase is reversibly deactivated by oxidation of Cys16 and Cys55 to a disulfide. Although not required for catalysis, Cys16 is an active-site residue positioned at the nucleotide-binding domain (Porter and Hartman, 1988). The hyperreactivity of Cys16 has heretofore limited further active-site characterization by chemical modification. To overcome this limitation, the partially active enzyme,S-methylated at Cys16, has been probed with a potential affinity reagent. Treatment of methylated enzyme with bromoacetylethanolamine phosphate results in essentially complete loss of catalytic activity. Inactivation follows pseudo-first-order kinetics and exhibits a rate saturation with an apparentK d of 3–4 mM. ATP, but not ribulose 5-phosphate, affords substantial protection. Complete inactivation correlates with incorporation of 1 mol of [14C]reagent per mole of enzyme subunit. Amino acid analysis of the [14C]-labeled enzyme demonstrates that only cysteine is modified, and mapping of tryptic digests shows that Cys55 is a major site of alkylation. Experimentally induced lesions of cutaneous leishmaniasis and the effect of concurrent bacterial infection on the development of these lesions were studied in the golden hamster. Male outbred golden hamsters received intradermal injections at the base of the tail with approximately 10(7) promastigotes of Leishmania braziliensis panamensis, or promastigotes combined with Staphylococcus aureus or Pasteurella multocida or both, bacteria only, or sterile Eagle's minimal essential medium (MEME). The size of the resulting lesions was measured at least twice each week. Hamsters were killed at postinoculation Days 6, 13, 20, 27, 41, or 48, and each lesion was measured, aseptically excised, and bisected; half was used for bacteriologic culture and the other half was prepared for light microscopic examination. Lesions resulting from L. b. panamensis alone progressed from initial erythema to a granulomatous nodule and finally to a necrotic granuloma, often capped by a crateriform ulcer. Lesions resulting from a suspension of L. b. panamensis with added S. aureus or S. aureus and P. multocida, were initially larger, more erythemic and contained a greater proportion of neutrophils up to postinoculation Days 14-21 than did lesions resulting from L. b. panamensis alone. Concurrent infections with bacteria such as S. aureus and P. multocida had little effect on the development of ulcerating characteristics of lesions, but when S. aureus was present it appeared to enhance the severity of the early lesions. Between postinoculation Days 14-28, lesions produced by L. b. panamensis, with or without added bacteria had similar developmental progression of sufficient size for optimal testing of antileishmanial compounds. 相似文献
