Since 1942, when paracoccidioidomycosis was first identified in the state of Rio Grande do Sul, paracoccidioidal pulmonary
lesions became a great concern to physicians. The present study focuses on 53 patients diagnosed over a seven-year period
who presented paracoccidioidal lesions circumscribed to the lungs. These patients presented clinical and radiological features
that simulated several pulmonary infectious and non-infectious conditions. Four unusual cases are briefly discussed. A sequence
of laboratorial tests should be established for the diagnosis of pulmonary paracoccidioidomycosis.

Zinc has been shown to disturb the innate host defense response by interfering in the activation of neutrophils and subsequent oxidative burst, although the exact role of this metal, either as an activator or inhibitor, remains a matter of controversy among research groups. These apparent discrepancies may be due to experimental settings, through modification of zinc availability to neutrophils, or to inaccurate detections of reactive species. Thus, the main objective of the present study was to provide clarification on the role of zinc on the activation of human neutrophils and the subsequent oxidative burst. For that purpose, different detection methods and incubation media were used. The obtained results showed that phosphate buffers (PBS and HBSS) complex with zinc and interfere with the results obtained with this metal. By using Tris-G, it was clearly demonstrated that zinc, at low concentrations (5–12.5 μM), activates NADPH oxidase, mainly via protein kinase C, leading to the formation of superoxide radical (O2 •−). Higher concentrations of zinc results on a rapid dismutation of O2 •− to oxygen and hydrogen peroxide, which in turn is used by myeloperoxidase to generate hypochlorous acid (HOCl). 相似文献

Methods: A total of 180 circular specimens (30 mm × 6 mm) were fabricated using three hard chairside reliners (Jet; n = 60, Kooliner; n = 60, Tokuyama Rebase II Fast; n = 60), which were immersed in deionised water (control), and five disinfectant solutions (1%, 2%, 5.25% sodium hypochlorite; 2% glutaraldehyde; 4% chlorhexidine gluconate). They were tested for Knoop hardness (KHN) and surface roughness (μm), before and after 30 simulated disinfecting cycles. Data was analysed by the factorial scheme (6 × 2), two‐way analysis of variance (anova ), followed by Tukey’s test. Results: For Jet (from 18.74 to 13.86 KHN), Kooliner (from 14.09 to 8.72 KHN), Tokuyama (from 12.57 to 8.28 KHN) a significant decrease in hardness was observed irrespective of the solution used on all materials. For Jet (from 0.09 to 0.11 μm) there was a statistically significant increase in roughness. Kooliner (from 0.36 to 0.26 μm) presented a statistically significant decrease in roughness and Tokuyama (from 0.15 to 0.11 μm) presented no statistically significant difference after 30 days. Conclusions: This study showed that all disinfectant solutions promoted a statistically significant decrease in hardness, whereas with roughness, the materials tested showed a statistically significant increase, except for Tokuyama. Although statistically significant values were registered, these results could not be considered clinically significant. 相似文献

Domestic animals are unique in that they have been organised into managed populations called breeds. The strength of genetic divergence between breeds may vary dependent on the age of the breed, the scenario under which it emerged and the strength of reproductive isolation it has from other breeds. In this study, we investigated the Gulf Coast Native breed of sheep to determine if it contains lines of animals that are sufficiently divergent to be considered separate breeds. Allele sharing and principal component analysis (PCA) using nearly 50,000 SNP loci revealed a clear genetic division that corresponded with membership of either the Florida or Louisiana Native lines. Subsequent analysis aimed to determine if the strength of the divergence exceeded that found between recognised breed pairs. Genotypes from 14 breeds sampled from Europe and Asia were used to obtain estimates of pair-wise population divergence measured as F
ST. The divergence separating the Florida and Louisiana Native (F
ST = 6.2%) was approximately 50% higher than the average divergence separating breeds developed within the same region of Europe (F
ST = 4.2%). This strongly indicated that the two Gulf Coast Native lines are sufficiently different to be considered separate breeds. PCA using small SNP sets successfully distinguished between the Florida and Louisiana Native animals, suggesting that allele frequency differences have accumulated across the genome. This is consistent with a population history involving geographic separation and genetic drift. Suggestive evidence was detected for divergence at the poll locus on sheep chromosome 10; however drift at neutral markers has been the largest contributor to the genetic separation observed. These results document the emergence of populations that can be considered separate breeds, with practical consequences for bio-conservation priorities, animal registration and the establishment of separate breed societies. 相似文献

Iron is an essential nutrient in several biological processes such as oxygen transport, DNA replication and erythropoiesis. Plasma iron normally circulates bound to transferrin. In iron overload disorders, however, iron concentrations exceed transferrin binding capacity and iron appears complexed with low molecular weight molecules, known as non-transferrin-bound iron (NTBI). NTBI is responsible for the toxicity associated with iron-overload pathologies but the mechanisms leading to NTBI uptake are not fully understood. Here we show for the first time that T lymphocytes are able to take up and accumulate NTBI in a manner that resembles that of hepatocytes. Moreover, we show that both hepatocytes and T lymphocytes take up the oligomeric Fe3Cit3 preferentially to other iron-citrate species, suggesting the existence of a selective NTBI carrier. These results provide a tool for the identification of the still elusive ferric-citrate cellular carrier and may also open a new pathway towards the design of more efficient iron chelators for the treatment of iron overload disorders. 相似文献

Keratitis treatment has become more complicated due to the emergence of bacterial or fungal pathogens with enhanced antibiotic resistance. The pharmaceutical applications of N-heterocyclic carbene complexes have received remarkable attention due to their antimicrobial properties. In this paper, the new precursor, 3,3′-(p-phenylenedimethylene) bis{1-(2- methyl-allyl)imidazolium} bromide (1a) and its analogous PF6 salt (1b) were synthesized. Furthermore, silver(I) and gold(I) -N-heterocyclic carbene (NHC) complexes [Ag2LBr2/Au2LBr2; 2a/3a], [(Ag2L2)(PF6)2/(Au2L2)(PF6)2; 2b/3b] were developed from their corresponding ligands. All compounds were screened for their antimicrobial activities against multiple keratitis-associated human eye pathogens, including bacteria and fungi. Complexes 2a and 3a showed highest activity, and the effectiveness of 3a was also studied, focusing eradication of pathogen biofilm. Furthermore, the structures of 1a, 2a and 3b were determined using single crystal X-ray analysis, 2b and 3a were optimized theoretically. The mechanism of action of 3a was evaluated by scanning electron microscopy and docking experiments, suggesting that its target is the cell membrane. In summary, 3a may be helpful in developing antimicrobial therapies in patients suffering from keratitis-associated eye infections caused by multidrug-resistant pathogens. 相似文献

Lectins constitute a class of glycoproteins, which are capable of selectively and reversibly binding to carbohydrates, distinguishing
small structural differences in complex oligosaccharides. Studies have shown that the binding of lectins to cell-surface carbohydrates
can lead to various effects such as cellular proliferation, histamine release and cytokine production. Canavalia brasiliensis lectin (ConBr) is a (D-mannose) D-glucose lectin. In this study, murine splenocytes were cultured to determine the effect
of ConBr on cell proliferation, nitric oxide (NO) release and cytokine secretion. In addition, cellular viability assays were
performed to evaluate any mitogenic activity induced by this lectin. ConBr significantly increased cell proliferation with
minimal cell damage. This lectin was able to induce an increased production of cytokines such as IL-2, IL-6 and IFN-γ and
a decreased production of IL- 10. The release of NO was also observed. The results of this study indicate that ConBr could

Objectives
To measure the oxygen and ventilatory output across all COPD stages performing 18 common ADL and identify the activities that present the highest metabolic and ventilatory output as well as to compare the energy expenditure within each disease severity.Materials and Methods
Metabolic (VO2 and VCO2), ventilatory (f and VE), cardiovascular (HR) and dyspnea (Borg score) variables were assessed in one hundred COPD patients during the completion of eighteen ADL grouped into four activities domains: rest, personal care, labor activities and efforts.Results
The activities with the highest proportional metabolic and ventilatory output (VO2/VO2max and VE/MVV) were walking with 2.5 Kg in each hand and walking with 5.0 Kg in one hand. Very severe patients presented the highest metabolic, ventilatory output and dyspnea than mild patients (p<0.05).Conclusions
COPD patients present an increased proportion of energy expenditure while performing activities of daily living. The activities that developed the highest metabolic and ventilatory output are the ones associated to upper and lower limbs movements combined. Very severe patients present the highest proportional estimated metabolic and ventilatory output and dyspnea. Activities of daily living are mainly limited by COPD’s reduced ventilatory reserve. 相似文献190.

In this study, the biodegradability of synthetic dyes used in the textile industry by Basidiomycetes collected in woods of
the Atlantic Forest of Pernambuco, Brazil, was evaluated. Among the fungi, the following species were identified: Caripia montagnei, Datronia caperata, Earliella scabrosa, Fomitopsis feei, Ganoderma stiptatum, Hexagonia hydnoides, Pleurotus ostreatus, Pycnoporus sanguineus and Trametes membranacea. Mycelium discs (2 cm2) of each fungal isolate were transferred to 30 ml of King medium supplemented with 0.05% m/v of each
of the following dyes: methyl orange, bromophenol blue (BB), methylene blue, Congo red, phenol red (PR) and methyl green (MG).
All these species showed potential to decolorize the tested dyes, with decolorization efficiency varying from 7.26 to 99.2%.
H. hydnoides and T. membrancea were the only species able to degrade all dyes. BB was effectively metabolized by P. sanguineus (97.4%), H. hydnoides (95.6%), E. scabrosa (96.6%) and T. membranacea (99.2%). E. scabrosa stood out among the isolates in dyes decolorization, allowing for efficiencies >90% for MG, PR, and BB. 相似文献
