We introduce a new approach to detect individual microparticles that contain NIR fluorescent dye by multispectral optoacoustic tomography in the context of the hemoglobin-rich environment within murine liver. We encapsulated a near infrared (NIR) fluorescent dye within polystyrene microspheres, then injected them into the ileocolic vein, which drains to the liver. NIR absorption was determined using multispectral optoacoustic tomography. To quantitate the minimum diameter of microspheres, we used both colorimetric and spatial information to segment the regions in which the microspheres appear. Regional diameter was estimated by doubling the maximum regional distance. We found that the minimum microsphere size threshold for detection by multispectral optoacoustic tomography images is 78.9 µm.  相似文献

The in vitro effects of the analgesic drugs, lornoxicam, indomethacin, tenoxicam, diclofenac sodium, ketoprofen and lincomycine, on the activity of purified human serum paraoxonase (hPON1) (EC 3.1.8.1.) were evaluated. hPON1 was purified from human serum with a final specific activity of 3840 U mg?1 and a purity of 25.3 % using simple chromatographic methods, including DEAE-Sephadex anion exchange and Sepharose 4B-L-tyrozine-1-napthylamine hydrophobic interaction chromatography. SDS-polyacrylamide gel electrophoresis indicated a single protein band corresponding to hPON1. The six analgesics dose-dependently decreased in vitro hPON1 activity, with IC50 values for lornoxicam, indomethacin, tenoxicam, diclofenac sodium, ketoprofen and lincomycine of 0.136, 0.195, 0.340, 1.639, 6.23 and 9.638 mM, respectively. Ki constants were 0.009, 0.097, 0.306, 0.805, 13.010 and 11.116 mM, respectively. Analgesics showed different inhibition mechanisms: lornoxicam, diclofenac sodium and lincomycine were uncompetitive, indomethacin and tenoxicam were competitive, ketoprofen was noncompetitive. According to the results, inhibition potency was lornoxicam>indomethacin>tenoxicam> diclofenac sodium>ketoprofen> lincomycine.  相似文献

We have previously shown microarchitectural tissue changes with cellular modifications in osteocytes following high chronic alcohol dose. The aim of this study was to assess the dose effect of alcohol consumption on the cytoskeleton activity, the cellular lipid content and modulation of differentiation and apoptosis in osteocyte. Male Wistar rats were divided into three groups: Control (C), Alcohol 25% v/v (A25) or Alcohol 35% v/v (A35) for 17 weeks. Bone mineral density (BMD) was assessed by DXA, osteocyte empty lacunae, lacunae surface, bone marrow fat with bright field microscopy. Osteocyte lipid content was analysed with transmission electron microscopy (TEM) and epifluorescence microscopy. Osteocyte apoptosis was analysed with immunolabelling and TEM. Osteocyte differentiation and cytoskeleton activity were analysed with immunolabelling and real time quantitative PCR. At the end of the protocol, BMD was lower in A25 and A35 compared with C, while the bone marrow lipid content was increased in these groups. More empty osteocyte lacunae and osteocyte containing lipid droplets in A35 were found compared with C and A25. Cleaved caspase‐3 staining and chromatin condensation were increased in A25 and A35 versus C. Cleaved caspase‐3 was increased in A35 versus A25. CD44 and phosphopaxillin stainings were higher in A35 compared with C and A25. Paxillin mRNA expression was higher in A35 versus A25 and C and sclerostin mRNA expression was higher in A35 versus C. We only observed a dose effect of alcohol consumption on cleaved caspase‐3 osteocyte immunostaining levels and on the number of lipid droplets in the bone marrow.  相似文献

At least half of the world's population resides in the coastal zone and the livelihoods of billions of people are affected either directly or indirectly by the production and sustainability of nearshore fisheries. Landscape change, specifically development of tree plantations, is accelerating worldwide as developing countries integrate into global markets to sell goods, offer climate‐mitigation services (carbon), and/or provide renewable energy. These changes can release excess nutrients into adjacent coastal waters causing eutrophication that alters the structure and function of coastal ecosystems. This study examined the relationship between coastal drainage basin land use/land cover change (LCLUC), specifically development of tree plantations, patterns of chlorophyll‐a in nearshore coastal waters, and the biological condition of commercially important shellfish, Concholepas concholepas (loco) in southern Chile. Locos (N = 1374) were sampled across 13 watersheds (35 853 km2) and 42 fisheries management areas (spanning 250 km of coastline). Locos harvested from management areas influenced by tree plantations had approximately 30% more endobiont (shell‐boring) phoronids, almost twice as many endobiont polychaetes and twice as many epibiont (shell‐attaching) barnacles than locos from areas in close proximity to watersheds dominated by native forests (15–20% of the watershed). Phoronid infested locos from coastal waters adjacent to watersheds with tree plantations were of relatively poor biological condition (smaller and narrower in width) and of reduced market value. Our study suggests that tree plantations result in indirect ecological impacts to coastal fisheries (more nutrients and higher phytoplankton biomass, resulting in smaller, low quality locos), and costs are born by coastal fishers (lower prices for locos). Increases in tree plantations could thus potentially significantly impact coastal fisheries worldwide and such problems should be managed as an interconnected network of land use change, oceanic ecosystems, and economic systems that are considered an integrated socio‐ecological system.  相似文献
