The long-range, pheromone-mediated, flight behaviour of male moths under natural and mating disruption conditions was monitored by means of harmonic radar. Individual male turnip moths, Agrotis segetum (Denis & Schiffermüller) (Lepidoptera: Noctuidae), tagged with radar transponders, were released and tracked in plots with or without disruptive doses of sex pheromone. In addition, male attraction to pheromone-baited traps and mating of calling females in treated and untreated plots was investigated. High doses of a four-component pheromone blend reduced trap catch by 79% and mating of females by 62% when compared with control plots in pre-radar experiments. Surprisingly, this effect was not associated with any pronounced differences in flight behaviour of males between a treatment and a control plot as revealed by harmonic radar recordings. In total, 20 flight tracks from a control plot and 22 flight tracks from a treatment plot were analysed. Moths could be followed for up to 77 min, corresponding to a track length of 7350 m. Mean ground speed ranged from 0.7 m s–1 to 5.4 m s–1. There was a strong trend (P = 0.06) for a greater number of male orientations to traps from downwind in the control field compared to the treatment field. Many flight tracks were fragmented due to radar shadow. Advantages and constraints using harmonic radar to study the pheromone-mediated flight behaviour of nocturnal moths are discussed. 相似文献

Transgenic Bt (expressing the cry1Ac endotoxin gene) and conventional oilseed rape plants grown in different soils were used to study nutrient uptake and emission of volatiles after herbivore damage. All plants were greenhouse-grown in soils representing low-, medium- and high-nutrient levels. The concentrations of N, P, K, Mg and Zn were significantly affected by the transgene, while the main effect of soil type appeared in N, P, Ca, Mg, B, Mn and Zn concentrations in the plants. Plants with four to five leaves were infested with the third instar larvae of Bt-susceptible Plutella xylostella for 48 h, and samples of volatiles were collected and analysed. In the first experiment, the soil nutrient level had a significant effect on the emissions of (Z)-3-hexen-1-ol, (Z)-3-hexenyl acetate, hexyl acetate, (E)-4,8-dimethyl-1,3,7-non-atriene (DMNT), beta-elemene, gamma-bisabolene, alpha-bisabolene and (E)-nerolidol. The induction of these volatiles was significantly higher in infested conventional plants grown at a high-soil nutrient level compared to infested conventional plants at a low-soil nutrient level. In the second experiment, the soil nutrient level had a significant effect on the emissions of (Z)-3-hexen-1-ol, (Z)-3-hexenyl acetate and beta-elemene and, again, this was significantly higher in infested conventional plants grown at high-soil nutrient levels in comparison with infested plants at a low-soil nutrient level. In both experiments, the transgene effect was significant on the emissions of DMNT and (E,E)-alpha-farnesene. The differences in emissions between the two separate experiments suggest that growth conditions (particularly daylength) and sampling procedure may affect the ratio of compounds detected in the emission blend, even though the response to herbivory, nutrient availability and the transgene were similar. 相似文献

Animal and plant diseases pose a serious and continuing threat to food security, food safety, national economies, biodiversity and the rural environment. New challenges, including climate change, regulatory developments, changes in the geographical concentration and size of livestock holdings, and increasing trade make this an appropriate time to assess the state of knowledge about the impact that diseases have and the ways in which they are managed and controlled. In this paper, the case is explored for an interdisciplinary approach to studying the management of infectious animal and plant diseases. Reframing the key issues through incorporating both social and natural science research can provide a holistic understanding of disease and increase the policy relevance and impact of research. Finally, in setting out the papers in this Theme Issue, a picture of current and future animal and plant disease threats is presented. 相似文献

The interaction of hemoglobin with phospholipid bilayer vesicles (liposomes) has been analyzed in several studies to better understand membrane-protein interactions. However, not much is known on hemoglobin interactions with the aminophospholipids, predominantly localized in the inner leaflet of erythrocytes, e.g., phosphatidylserine (PS), phosphatidylethanolamine (PE) in membranes containing phosphatidylcholine (PC). Effects of cholesterol, largely abundant in erythrocytes, have also not been studied in great details in earlier studies. This work therefore describes the study of the interactions of different hemoglobin variants HbA, HbE and HbF and the globin subunits of HbA with the two aminophospholipids in the presence and absence of cholesterol. Absorption measurements indicate preferential oxidative interaction of HbE and alpha-globin subunit with unilamellar vesicles containing PE and PS compared to normal HbA. Cholesterol was found to stabilize such oxidative interactions in membranes containing both the aminophospholipids. HbE and alpha-globin subunits were also found to induce greater leakage of membrane entrapped carboxyfluorescein (CF) using fluorescence measurements. HbE was found to induce fusion of membrane vesicles containing cholesterol and PE when observed under electron microscope. Taken together, these findings might be helpful in understanding the oxidative stress-related mechanism(s) involved in the premature destruction of erythrocytes in peripheral blood, implicated in the hemoglobin disorder, HbE/beta-thalassemia. 相似文献

Many crops transformed with insecticidal genes isolated from Bacillus thuringiensis (Bt) show resistance to targeted insect pests. The concentration of Bt endotoxin proteins in plants is very important in transgenic crop efficacy and risk assessment. In the present study, changes in levels of Cry1Ac protein in the leaves of transgenic Bt oilseed rape (Brassica napus) carrying a Bt cry1Ac gene under the control of the cauliflower mosaic virus 35S promoter were quantified during vegetative growth by enzyme‐linked immunosorbent assay. Plants were grown in a glasshouse, sampled at 2, 4, 5 and 6 weeks, and the concentration of Cry1Ac was quantified in basal, top and previous top leaves. The mean concentration differed between sowing dates when Cry1Ac concentration was expressed as ng g?1 fresh leaf weight but not when expressed as ng mg?1 total soluble protein. It was demonstrated that Cry1Ac concentration increased significantly as the leaf aged, while the total soluble plant protein decreased significantly. Levels of Cry1Ac were therefore higher in leaves at the base of the plants than in leaves close to the growing point. However, even young leaves with very low Cry1Ac concentrations caused high mortality in the larvae of a Cry1Ac‐susceptible laboratory strain of the diamondback moth. The feeding area of leaves consumed by larvae in vivo and in situ was similar. Leaf damage caused by sampling (i.e. artificially) or by feeding of larvae did not affect the levels of Cry1Ac in the leaves under the experimental conditions in this study. 相似文献

We hypothesized that Shewanella oneidensis MR-1, a model dissimilatory metal-reducing bacterium, could utilize environmentally relevant concentrations of tyrosine to produce pyomelanin for enhanced Fe(III) oxide reduction. Because homogentisate is an intermediate of the tyrosine degradation pathway, and a precursor of a redox-cycling metabolite, pyomelanin, we evaluated the process of homogentisate production by S. oneidensis MR-1, in order to identify the key steps involved in pyomelanin production. We determined that two enzymes involved in this pathway, 4-hydroxyphenylpyruvate dioxygenase and homogentisate 1,2-dioxygenase are responsible for homogentisate production and oxidation, respectively. We used genetic analysis and physiological characterization of MR-1 strains either deficient in or displaying substantially increased pyomelanin production. The relative significance imparted by pyomelanin on solid-phase electron transfer was also addressed using electrochemical techniques, which allowed us to extend the genetic and physiological findings to biogeochemical cycling of metals. Based on our findings, environmental production of pyomelanin from available organic precursors could contribute to the survival of S. oneidensis MR-1 when dissolved oxygen concentrations become low, by providing an increased capacity for solid-phase metal reduction. This study demonstrates the role of organic precursors and their concentrations in pyomelanin production, solid phase metal reduction and biogeochemical cycling of iron. 相似文献

plant which had been previously damaged byA. pisum also increased their response to an undamaged plant and a host-damaged plant, whereas the experience of searching on an undamaged
plant did not significantly change their responses to undamaged plants. However, when parasitoids were allowed to search on
an undamaged plant which had been sprayed withA. pisum honeydew, this significantly increased their response to an undamaged plant. Oviposition experience on the plant-host complex
and foraging experience on a host-damaged plant or an undamaged plant sprayed with honeydew also significantly reduced the
mean time taken by the parasitoids to respond in the wind tunnel. The behavioral changes associated with such experience were
acquired within 30 min and persisted for at least 3 days. The results demonstrate the capacity ofA. ervi to learn associatively olfactory cues from plants, host-damaged plants, and plant-host complexes and confirm the role of

The host preference behaviour of the generalist aphid parasitoid Aphidius colemaniwas investigated using a Y-tube olfactometer. Female A. colemanishowed a preference for the host-plant complex on which they had been reared, even though the same aphid host was involved, demonstrating a host plant preference. This preference was not evident when the parasitoids were dissected from their mummies prior to adult emergence. Host plant preference exhibited during host selection appeared to be induced by chemical cues encountered on the mummy case at the time of emergence, but preferences could be changed by subsequent foraging experiences. It is concluded that plant chemical cues play a major role in determining initial preferences through a process of emergence conditioning but that learning processes, involving cues encountered during oviposition in or contact with the host, can modify these initial preferences. 相似文献
