Saddle gall midge Haplodiplosis marginata (Diptera: Cecidomyiidae) is a pest of cereals across Europe. The occasional nature of this pest has resulted in limited and sporadic research activity. There remain important gaps in knowledge due either to a genuine lack of research or to previous research being difficult to access. These knowledge gaps make the development of effective control options difficult. Here, we review the existing literature in an attempt to consolidate the information on H. marginata from research which spans several decades and encompasses many different countries. The current distribution and pest status of this insect are updated, along with the methods of cultural and chemical control available to growers. The biology and life history of the insect are described in detail and the ecological processes governing them are discussed. A forecasting model is presented which allows the emergence of this pest in the UK to be predicted from degree day data, and the potential application of this model in management decisions is discussed. Finally, the areas in most need of further research are identified, along with suggestions of how this information can be used to help develop effective and sustainable management solutions for this pest. 相似文献

Background
Evolution is a difficult subject for students, with well-documented confusion about natural selection, tree thinking, and genetic drift among other topics. Here we investigate the effect of a simulation-based module about the conservation of black-footed ferrets, a module designed with pedagogical approaches that have been demonstrated to be effective, for teaching genetic drift. We compared performance on the Genetic Drift Inventory (GeDI) of students who completed the module and students who were in classes that used other methods for teaching genetic drift.Results
Students in 19 courses using the simulation-based module improved their understanding of genetic drift significantly after completing the Ferrets module, as measured by the GeDI. Students in five control courses actually performed significantly worse on the GeDI after instruction. The lower scores in the control courses were driven by a decrease in these students’ understanding of key concepts.Conclusions
The Ferrets module appears to be an effective way to teach genetic drift. In the control courses, students’ progress in understanding genetic drift may pass through a stage where their understanding of key concepts is worse than it was prior to instruction. However, students who learned genetic drift in courses that used the Ferrets module showed a more rapid increase in their understanding of key concepts related to genetic drift. This result suggests that the paths that students can take to move from novice to expert understanding may be more varied than was previously predicted.173.

In this study, we explore the relationship between moments in the frontal and sagittal planes, generated by a lifting task, vs the electromyographic (EMG) activity of right and left trunk muscle groups. In particular, we postulate that the functional dependence between erector spinae muscle activity and the applied lifting moments about the spine is as follows: the sum of left and right erector spinae processed EMG depends on the sagittal plane moment, and the difference of left and right erector spinae processed EMG depends on the frontal plane moment. A simple out-of-sagittal plane physical model, treating the lumbar spine as a two degree-of-freedom pivot point is discussed to justify these hypotheses. To validate this model, we collected surface EMG and lifting moment data for ten males performing a grid of frontal and sagittal plane lifting tasks. A digital RMS-to-DC algorithm was developed for processing raw EMG. For these tests, we measured EMG for the left and right erector spinae and for the left and right external oblique muscles. The processed EMG signals of the left and right erector spinae muscles are summed and differenced for comparison to the measured sagittal and frontal plane moments. A linear correlation (r2) of 0.96 was obtained for the sum of erector spinae EMG vs the sagittal plane moment; a corresponding value of r2 = 0.95 was obtained for the difference vs the frontal plane moment. No correlations (r2 less than 0.004) was found for the sagittal plane moment and the difference of the left and right erector spinae EMG, and the frontal plane moment and the sum of the left and right erector spinae EMG. 相似文献

The effects of anions on inorganicpyrophosphate-dependent H+-transport in isolated tonoplast vesicles from oat (Avena sativa L.) roots were determined. Both fluorescent and radioactive probes were used to measure formation of pH gradients and membrane potential in the vesicles. Pyrophosphate hydrolysis by the H+-translocating pyrophosphatase was unaffected by anions. Nonetheless, some anions (Cl-, Br- and NO3-) stimulated H+-transport while others (malate,
and iminodiacetate) did not. These differential effects were abolished when the membrane potential was clamped at zero mV using potassium and valinomycin. Stimulation of H+-transport by Cl- showed saturation kinetics whereas that by NO3- consisted of both a saturable component and a linear phase. For Cl- and NO3-, the saturable phase had a K
m
of about 2 mol·m-3. The anions that stimulated H+-transport also dissipated the membrane potential (.) generated by the pyrophosphatase. It is suggested that the stimulatory anions cross the tonoplast in response to the positive generated by the pyrophosphatase, causing dissipation of and stimulation of pH, as expected by the chemiosmotic hypothesis. The work is discussed in relation to recent studies of the effects of anions on ATP-dependent H+-transport at the tonoplast, and its relevance to anion accumulation in the vacuole in vivo is considered.Abbreviations and symools BTP

To study the effect of strain rate on the acoustic emission amplitude signature of bone, bovine cortical bone was milled into standard tensile specimens which were tested at two different strain rates while being monitored with acoustic emission equipment. It was demonstrated that the amplitude distribution of the acoustic events in bone is dependent on strain rate. Greater numbers of events occurred with the slower strain rate (0.0001 s-1), but these events were of lower amplitude than those emitted during the more rapid strain rate (0.01 s-1). The plot of the cumulative event amplitude distribution followed the power-law model, and the slope of this output, the b-value, represented a signature of the amplitude distribution. The mechanical test results were consistent with the behavior of a viscoelastic multi-phase composite material. 相似文献

Binding of pig plasma gelsolin to F-actin and partial fractionation into calcium-dependent and calcium-independent forms 总被引:4,自引:0,他引:4
The interaction of pig plasma gelsolin with F-actin has been studied by a sedimentation assay using 125I-gelsolin in a Beckman Airfuge. Over 90% of the gelsolin bound to F-actin in 0.1 mM CaCl2 in experiments using 24 microM actin and 2-10 nM 125I-gelsolin, but only 40-50% bound in 1 mM EGTA. Addition of more F-actin to the EGTA supernatant does not sediment this gelsolin. Demonstration of this partial calcium sensitivity depends critically on the use of F-actin that has been prepared in the absence of calcium ions. F-actin prepared from G-actin in calcium or pretreated with calcium, binds 125I-gelsolin more completely in EGTA. This suggests that gelsolin activity is influenced by transient exposure of actin to calcium. Further evidence for partial calcium sensitivity in the interactions between gelsolin and F-actin has been obtained by other methods, including viscometry and electron microscopy. The gelsolin present in the EGTA supernatant is complexed to G-actin, predominantly as binary complexes. Very low concentrations of these complexes reduce the viscosity of F-actin in calcium but not in EGTA. Whether this effect is due to severing activity, or capping with consequent depolymerization to establish the new critical concentration, is uncertain. The results suggest the presence of two types of gelsolin, one that requires micromolar concentrations of calcium for binding to F-actin and one that does not. Both bind to G-actin. Partial separation has been achieved using actin-Sepharose. Pig plasma gelsolin is heterogeneous on isoelectric focussing gels in urea, but the two types of gelsolin separated on actin-Sepharose do not correspond to specific isoelectric species. 相似文献

1H, 2H, and 31P NMR methods have been employed in the study of dimyristoyl lecithin bilayers hydrated with D2O in the gel (L beta'), intermediate (P beta') and liquid-crystalline (L alpha) phases. For D2O/lipid molar ratios, n, in the range 7 less than or equal to n less than or equal to 11 discontinuities are observed in the deuterium NMR splittings at both main and pretransitions. A partial phase diagram based on NMR and differential scanning calorimetry data is presented. 1H NMR dipolar splittings are observed for macroscopically oriented samples in all three phases. Changes in the 1H splittings are correlated with 2H and 31P data and interpreted to show that the chain tilt in the gel phase undergoes a discontinuous change on transition to the intermediate phase, which brings the chain axes closer to the bilayer normal. An estimate of chain tilt in the gel phase is made on the basis of NMR data and found to be approximately 23 degrees for a sample with n = 11 at 18 degrees C. 相似文献

Summary We have examined the procollagens and collagens produced by skin fibroblasts from a patient with Ehlers-Danlos syndrome type VII. The patient was heterozygous for an abnormal 2(I) chain migrating with the approximate size of pN2(I) chains after pepsin digestion. Peptide mapping suggested that the abnormality was located at the amino-terminus of the 2(I) chain. Quantitative analysis of the 2(I) mRNA indicated loss of the exon 6 sequences, and subsequent polymerase chain reaction amplification of cDNA demonstrated a deletion of the 54 bp of exon 6 from some of the 2(I) mRNA. Analysis of genomic DNA from the patient revealed a single base change in one COL1A2 allele, substituting an A for a G as the first base of intron 6. This change mutates the obligate GT-dinulceotide splicing signal to AT and leads to exon skipping with splicing from exon 5 to exon 7. Loss of exon 6 sequences results in the loss of the procollagen-N-propeptidase cleavage site and a lysine residue that normally participates in covalent intermolecular crosslinking within collagen fibres. 相似文献
