Anti-idiotypic antibody mimics proteolytic function of parent antigen   总被引:1,自引:0,他引:1  
Functional imaging of subtilisin Carlsberg active center by the idiotypic network yielded a catalytic anti-idiotypic antibody with endopeptidase, amidase, and esterase activities. A monoclonal antibody inhibitory to subtilisin (Ab1 5-H4) was employed as the template for guiding the idiotypic network to produce the catalytic anti-idiotypic Ab2 6B8-E12. Proteolytic activity of 6B8-E12 was demonstrated by zymography using self-quenched fluorescein-BSA conjugate and in a coupled assay detecting Ab2-dependent RNase A inactivation. Cleavage of peptide substrates by 6B8-E12 revealed distinct patterns of hydrolysis with high preference for aromatic residues before or after the scissile bond. Catalytic activity of Ab2 was inhibited by phenylmethylsulfonyl fluoride, a mechanism-based inhibitor of serine hydrolases. 5-H4 and 6B8-E12 were cloned, produced in Escherichia coli as single-chain variable fragments (scFvs), and purified. Kinetic parameters for amidolytic and esterolytic activities were similar in Ab2 and its scFv derivative. Although the antigen-specific portion of 6B8-E12 possesses no primary structure similarity to subtilisin, it mimics proteolytic and amidolytic functions of the parental antigen, albeit with 4 orders of magnitude slower acceleration rates. The lack of detectable endopeptidase activity of 6B8-E12 scFv raises interesting issues concerning general evolution of catalytic activity. The in silico 3D models of Ab1 and Ab2 revealed strong structural similarity to known anti-protease antibodies and to abzymes, respectively. These results indicate that the idiotypic network is capable, to a significant extent, of reproducing catalytic apparatus of serine proteases and further validate the use of imaging of enzyme active centers by the immune system for induction of abzymes accelerating energy-demanding amide bond hydrolysis.  相似文献

The Gracillariidae fauna of Siberia, the region that occupies almost half of the territory of the Russian Federation, remains poorly studied. During a DNA barcoding study of Gracillariidae in Siberia, based on analysis of larvae and pupae collected from their leaf mines on woody plants, we identified 41 species. Three gracillariids were identified only to genus: Caloptilia sp. (host plant Prunus padus), Parornix sp. (Malus sp.), and Phyllonorycter sp. (Crataegus sp.), representing poorly studied or undescribed species. Six species are reported here for the first time for Siberia: Callisto insperatella (from Novosibirsk and Tomsk provinces), Caloptilia alnivorella (Buryatia), Phyllonorycter ermani (Irkutsk Province), Ph. lantanella (Novosibirsk Province), Ph. pumilae (Omsk and Irkutsk provinces), and Ph. viciae (Krasnoyarsk Territory). Parornix pfaffenzelleri, found in Khakassia, is reported as a species new to Russia. Other 15 gracillariid species previously known from Siberia were recorded in new administrative regions. The invasive lime leafminer Phyllonorycter issikii was first documented in Tomsk Province and Krasnoyarsk Territory in 2017 and 2018, respectively. Seven new gracillariid-host plant associations were found: Parectopa ononidis on Lupinaster pentaphyllus, Sauterina hofmanniella on Lathyrus gmelinii, Caloptilia stigmatella on Salix kochiana, Callisto insperatella on Prunus virginiana, Parornix scoticella on Amelanchier sp., Phyllonorycter ermani on Alnus alnobetula subsp. fruticosa, and Phyllonorycter viciae on Vicia unijuga. The Gracillariidae fauna of Siberia has 51% similarity with that of European Russia and only 38% similarity with that of the Russian Far East.

Directions and formats of work with medical personnel, including those invited from other medical institutions, as well as their information support were determined. Organic photovoltaic (OPV) technology offers many advantages, although no commercial applications have been achieved after more than a decade of intensive research and development. Several challenges have yet to be overcome including high power conversion efficiency (PCE), good processability, low cost, and excellent long‐term stability, and so on. In this article, these fundamental challenges are significantly addressed by surveying and analyzing a new merit factor for material applied accessibility containing three parameters: synthetic complexity, device efficiency, and photostability. Thirty‐five donor small molecules are introduced to assess their synthetic accessibility. Furthermore, the PCEs and device photostability of these molecules are carried out, and further measured under one sun illumination within 200 h, respectively. Combining with the characteristics of these three factors, investigated molecules are ranked according to an industrial figure of merit (i‐FOM), while some guidelines for the material design and synthesis are given. It is suggested that a PCE of >14% and an i‐FOM of >20% via active material engineering are realistic for possible industry future of OPV. Along with the systematic study, it is believed that this i‐FOM can be taken into consideration at an early stage of molecular design and provides valuable insight for efficient evaluation of photovoltaic materials for possible commercial applications.  相似文献
