North American prairie pothole wetlands are known to be important carbon stores. As a result there is interest in using wetland restoration and conservation programs to mitigate the effects of increasing greenhouse gas concentration in the atmosphere. However, the same conditions which cause these systems to accumulate organic carbon also produce the conditions under which methanogenesis can occur. As a result prairie pothole wetlands are potential hotspots for methane emissions. We examined change in soil organic carbon density as well as emissions of methane and nitrous oxide in newly restored, long-term restored, and reference wetlands across the Canadian prairies to determine the net GHG mitigation potential associated with wetland restoration. Our results indicate that methane emissions from seasonal, semi-permanent, and permanent prairie pothole wetlands are quite high while nitrous oxide emissions from these sites are fairly low. Increases in soil organic carbon between newly restored and long-term restored wetlands supports the conclusion that restored wetlands sequester organic carbon. Assuming a sequestration duration of 33 years and a return to historical SOC densities we estimate a mean annual sequestration rate for restored wetlands of 2.7 Mg C ha−1year−1 or 9.9 Mg CO2 eq. ha−1 year−1. Even after accounting for increased CH4 emissions associated with restoration our research indicates that wetland restoration would sequester approximately 3.25 Mg CO2 eq. ha−1year−1. This research indicates that widescale restoration of seasonal, semi-permanent, and permanent wetlands in the Canadian prairies could help mitigate GHG emissions in the near term until a more viable long-term solution to increasing atmospheric concentrations of GHGs can be found.  相似文献

Three lakes were studied in the High Tatra Mountains at altitudes from 2000 to 2157 m a.s.l., which represent three categories of acidity status recognised in the Tatra lakes in the 1980s: non-acidified, acidified, and strongly acidified. Subfossil chironomid remains from dated sediment cores covering ca. throughout 200 years were analysed. The chironomid thanatocoenoses of all cores layers reflected ultra-oligotrophic non-acidified conditions in L’adové pleso. Nevertheless, the finding of the acid-tolerant species Zalutschia tatrica in littoral samples in 1980s indicates that the littoral zone was more influenced by acidification than deeper areas of the lake. Three stages of lake developmental history can be distinguished in the acidified lake Vy?né Wahlenbergovo pleso based on the chironomid subfossil record: the pre-acidification stage before the 1920s, the anthropogenic acidification stage between the 1920–1980s, and the recovery of the lake from the end of the 1980s. The period of acidification was accompanied by a decline in chironomid numbers, while chironomid fauna composition was unchanged during the whole investigated history of the lake. None of the chironomid taxa present prior to the acidification period disappeared during the peak of acidification. The most apparent change in the sediment record taken from the strongly acidified lake Starolesnianske pleso was recognised in the layers corresponding to the period 1960–1980. It is characterised by the disappearance of the dominant acid-sensitive Tanytarsus lugens group and the dominance of acidtolerant Tanytarsus gregarius group. The most recent chironomid assemblage probably benefits from the amount of food resources as a result of increased lake productivity induced by acidification.  相似文献

The “holy grail” of regenerative medicine is the identification of an undifferentiated progenitor cell that is pluripotent, patient specific, and ethically unambiguous. Such a progenitor cell must also be able to differentiate into functional, transplantable tissue, while avoiding the risks of immune rejection. With reports detailing aberrant genomic imprinting associated with assisted reproductive technologies (ART) and reproductive cloning, the idea that human embryonic stem cells (hESCs) derived from surplus in vitro fertilized embryos or nuclear transfer ESCs (ntESCs) harvested from cloned embryos may harbor dangerous epigenetic errors has gained attention. Various progenitor cell sources have been proposed for human therapy, from hESCs to ntESCs, and from adult stem cells to induced pluripotent stem cells (iPS and piPS cells). This review highlights the advantages and disadvantages of each of these technologies, with particular emphasis on epigenetic stability.  相似文献

Resource pulses can have cascading effects on the dynamics of multiple trophic levels. Acorn mast is a pulsed resource in oak-dominated forests that has significant direct effects on acorn predators and indirect effects on their predators, prey, and pathogens. We evaluated changes in acorn mast, rodent abundance, raptor abundance, and reproductive success of a ground-nesting songbird over a 24-year period (1980–2004) in the southern Appalachian Mountains in an effort to determine the relationships among the four trophic levels. In particular, we examined the following: acorn mast from red oaks (Quercus rubra) and white oaks (Q. alba), abundance of white-footed mice (Peromyscus leucopus) and deer mice (P. maniculatus), population estimates of seven raptor species from three feeding guilds, and nest failure and number of juveniles of dark-eyed juncos (Junco hyemalis). Finally, we recorded seasonal temperature and precipitation to determine the effects of weather on each trophic level. We found that weather patterns had delayed effects of up to 3 years on these trophic interactions. Variation in acorn mast, the keystone resource in this community, was explained by weather conditions as far back as 2 years before the mast event. Acorn mast, in turn, was a strongly positive predictor of rodent abundance the following year, whereas spring and summer temperature and raptor abundance negatively affected rodent abundance. Dark-eyed junco nests were more likely to fail in years in which there were more rodents and raptors. Nest failure rate was a strong predictor of the number of juvenile juncos caught at the end of the summer. Our results improve our understanding of the complex ecological interactions in oak-dominated forests by illustrating the importance of abiotic and biotic factors at different trophic levels. Lentiviral vectors have been used for gene transfer into the liver but their ability to efficiently transduce quiescent hepatocytes remains controversial. Lentivirus-mediated gene transfer is more efficient in cycling cells. We determine the effect of H-IL6 in the lentiviral transduction. The lentiviral vector was used to transduce HepG2 cells and mice liver cells, previously treated with H-IL6. The highest transduction level was observed in HepG2 cells treated with 30 ng/mL H-IL6 and in the mice that received 4 μg H-IL6. Our results suggest that H-IL6 is an inducer of lentiviral gene transfer into the liver cells without any toxicity.  相似文献

Though researchers have studied lowland gray langurs extensively, there is little information about the Himalayan populations. We provide foraging data from a field study of Himalayan langurs in Langtang National Park, Nepal at 3000–4000 m elevation. Phenological records show marked seasonality in resource abundance, with extremely low availability in winter, increasing abundance in spring and monsoon, and a reduction in fall. Activity budgets indicated greater time devoted to feeding as total vegetation abundance decreased. Diet included leaf buds, ripe fruit, and evergreen mature leaves in winter; deciduous young leaves in spring; and deciduous mature leaves in the monsoon and fall. Supplemental resources, such as underground storage organs, bark, and herbaceous vegetation, were also seasonally important. Among plant part classes included in the phenological sample, abundance and consumption correlate positively for all primary food resources except evergreen mature leaves and unripe fruit. Daily path lengths varied by season and, when controlled for overall vegetation abundance, positively relate to the consumption of soft underground storage organs, fruits, and deciduous mature leaves. The results contradict the common generalization of leaves as ubiquitous or nonpatchy resources.  相似文献

Transdermal drug delivery system (TDDS) may provide a more reliable method of drug delivery than oral delivery by avoiding gut absorption and first-pass metabolism, but needs a method for efficiently crossing the epidermal barrier. To enhance the delivery through the skin, we have developed a biocompatible, dissolvable microneedle array made from carboxymethyl cellulose (CMC). Using laser ablation for creating the mold greatly improved the efficiency and reduced the cost of microneedle fabrication. Mixing CMC with amylopectin (AP) enhanced the mechanical and tunable dissolution properties of the microneedle for controlled release of model compounds. Using the CMC microneedle array, we observed significant enhancement in the skin permeability of a fluorescent model compound, and also increase in the anti-oxidant activity of ascorbic acid after crossing the skin. Our dissolvable microneedle array provides a new and biocompatible method for delivery of drugs and cosmetic compounds through the skin.  相似文献

Abnormal angiogenesis is critically involved in tumor progression and metastasis including endometrial cancer and is regulated by microRNAs such as microRNA-101 (miR-101). We hypothesize that miR-101 expression is disrupted in endometrial cancer and modulation of miR-101 levels is sufficient to regulate tumor growth through angiogenesis. We examined the expression levels of miR-101 and factors involved in angiogenesis in the patients with endometrial cancer. We also overexpressed or inhibited miR-101 in RL-95-2 cells and examined their effects on cell toxicity and tumor growth. Finally, we determined if miR-101 regulated tumorigenesis through cyclooxygenase-2 (COX-2). We found that miR-101 levels were significantly reduced. Factors involved in angiogenesis included vascular endothelial growth factor-A (VEGF-A), thrombospondin-1 (TSP-1), cyclooxygenase-2 (COX-2), prostaglandin E2 (PGE2), and aromatase (P450arom), which were increased in endometrial carcinoma. Modulation of miR-101 level was sufficient to affect tumor growth. Finally, we found that the effects of miR-101 inhibition on tumor growth were suppressed by COX-2 inhibition. Our results suggest that modulating miR-101 and COX-2 levels or their activity may be a potential therapeutic strategy for endometrial cancer.  相似文献
