We used fingerprinting and cloning-sequencing to study the spatiotemporal dynamics and diversity of Planctomycetes in two perialpine lakes with contrasting environmental conditions. Planctomycetes, which are less-abundant bacteria in freshwater ecosystems, appeared to be structured in the same way as the entire bacterial community in these ecosystems. They were more diversified and displayed fewer temporal variations in the hypolimnia than in the epilimnia. Like the more-abundant bacterial groups in aquatic systems, Planctomycetes communities seem to be composed of a very small number of abundant and widespread operational taxonomic units (OTUs) and a large number of OTUs that are present at low abundance. This indicates that the concept of "abundant or core" and "rare" bacterial phylotypes could also be applied to less-abundant freshwater bacterial phyla. The richness and diversity of Planctomycetes were mainly driven by pH and were similar in both of the lakes studied, whereas the composition of the Planctomycetes community seemed to be determined by a combination of factors including temperature, pH, and nutrients. The relative abundances of the dominant OTUs varied over time and were differently associated with abiotic factors. Our findings demonstrate that less-abundant bacterial phyla, such as Planctomycetes, can display strong spatial and seasonal variations linked to environmental conditions and suggest that their functional role in the lakes studied might be attributable mainly to a small number of phylotypes and vary over space and time in the water column.  相似文献

This work describes the generation of novel PHAs (named PHACOS) with a new monomer composition containing thioester groups in the side chain, which confers new properties and made them suitable for chemical modifications after their biosynthesis. We have analyzed the PHACOS production abilities of the wild-type strain Pseudomonas putida KT2442 vs. its derived strain P. putida KT42FadB, mutated in the fadB gene from the central metabolic β-oxidation pathway involved in the synthesis of medium-chain-length PHA (mcl-PHA). Different fermentation strategies based on one- or two-stage cultures have been tested resulting in PHACOS with different monomer composition. Using decanoic acid as inducer of the growth and polymer synthesis and 6-acetylthiohexanoic acid as PHA precursor in a two-stage strategy, the maximum yield was obtained by culturing the strain KT42FadB. Nuclear magnetic resonance and gas chromatography coupled to mass spectrometry showed that polymers obtained from the wild-type and KT42FadB strains, included 6-acetylthio-3-hydroxyhexanoic acid (OH-6ATH) and the shorter derivative 4-acetylthio-3-hydroxybutanoic acid (OH-4ATB) in their composition, although in different ratios. While the polymer obtained from KT42FadB strain contained mainly OH-6ATH monomer units, mcl-PHA produced by the wild-type strain contained OH-6ATH and OH-4ATB. Furthermore, polyesters showed differences in the OH-alkyl derivates moiety. The strain KT42FadB overproduced PHACOS when compared to the production rate of the control strain in one- and two-stage cultures. Thermal properties obtained by differential scanning calorimetry indicated that both polymers have different glass transition temperatures related to their composition.  相似文献

Insulin was found to provoke simultaneous, rapid, biphasic increases in [3H]choline-labeling of phosphatidylcholine and phosphocholine in BC3H-1 myocytes. Phorbol esters increased [3H]choline-labeling of phosphocholine, but not phosphatidylcholine. Both agonists increased diacylglycerol production. These results suggest that: (a) insulin provokes coordinated increases in the synthesis and hydrolysis of PC; and, (b) insulin-induced activation of protein kinase C may activate a PC-specific phospholipase.  相似文献

The 'helpers at the nest' hypothesis suggests that individuals who are not currently reproducing often help kin by caretaking and thereby increase their inclusive fitness. Using a large scale historical dataset (Integrated Public Use Microdata Series sample of 1910; n=13,935), the hypothesis is tested that childless couples are more likely to fulfil such a role by taking care of a niece or nephew, but not a parent, than couples with children. Childless couples were significantly more likely to take care of a niece or nephew than couples with children. In contrast, couples with children and childless couples did not differ in caretaking of parents. Childless couples were also more likely to have more and younger nieces/nephews in their home than couples with children.  相似文献

This study presents data on over 350,000 insured Swedish dogs up to 10 years of age contributing to over one million dog-years at risk (DYAR) during 1995–2000. A total of 43,172 dogs died or were euthanised and of these 72% had a claim with a diagnosis for the cause of death. The overall total mortality was 393 deaths per 10,000 DYAR. Mortality rates are calculated for the 10 most common breeds, 10 breeds with high mortality and a group including all other breeds, crudely and for general causes of death. Proportional mortality is presented for several classifications. Five general causes accounted for 62% of the deaths with a diagnosis (i.e. tumour (18%), trauma (17%), locomotor (13%), heart (8%) and neurological (6%)). Mortality rates for the five most common diagnoses within the general causes of death are presented. These detailed statistics on mortality can be used in breed-specific strategies as well as for general health promotion programs. Palms can grow at sites exposed to high winds experiencing large dynamic wind and gust loads. Their stems represent a system of stiff fibrous elements embedded in the soft parenchymatous tissue. The proper design of the interface of the stiffening elements and the parenchyma is crucial for the functioning of the stem. The strategy of the palm to compromise between stiff fibre caps and the soft parenchymatous tissue may serve as a model system for avoiding stress discontinuities in inhomogeneous and anisotropic fibre-reinforced composite materials. We investigated the mechanical, structural and biochemical properties of the fibre caps of the palm Washingtonia robusta at different levels of hierarchy with high spatial resolution. A gradual decrease in stiffness across the fibre cap towards the surrounding parenchymatous tissue was observed. Structural adaptations at the tissue level were found in terms of changes in cell cross sections and cell wall thickness. At the cell wall level, gradients across the fibre cap were found in the degree of orientation of the microfibrils and in the lignin level and composition. The impact of these structural variations in the local material stiffness distribution is discussed.  相似文献

To enable use of Empididae s.l. (Diptera) as a tool in nature conservation, a Red Data Book of this taxonomical group was generated for Flanders, Belgium. All distribution data on species in Belgium between 1887 and 1999 were gathered from collections as well as personal sampling efforts by the first two authors. This resulted in about 21,000 records of Empididae, Hybotidae, Microphoridae and Atelestidae with 16,119 records for Flanders (northern Belgium) and 4776 for Wallony (southern Belgium). All species were assigned to Red Data Book categories which are based on a combination of a rarity and a trend criterion. Rarity is expressed as the proportion of the total number of UTM 5km squares sampled in which the species have been found since 1981. The trend criterion is interpreted as the change of the species rarity between 1887–1980 and 1981–1999. A comparable number of UTM 5km squares was investigated during the two time periods. A total of 259 species were recorded in Flanders. Twenty-seven or 10% of them are considered as 'extinct in Flanders', 10 (4%) as 'critically endangered', 12 (5%) as 'endangered', 11 (4%) as 'vulnerable', 99 species (38%) as 'susceptible' or 'rare', 65 species (25%) as 'safe' or 'at low risk' and 34 species (13%) are assigned to the category 'data deficient' due to taxonomic problems or a lack of ecological data. Only one of the common species shows a recent decrease of more than 50% and is classified as 'nearly threatened'. Current threats in most species are related to the alteration or destruction of their favoured habitats. The results are discussed in the light of recent criticism of the use of Red Data Books in nature conservation.  相似文献
