Summary Mice deprived of B lymphocytes by the chronic administration of anti-IgM antibodies have been shown to possess a heightened natural resistance (NR) to micro-organisms, to parental bone marrow, and to natural killer (NK)-sensitive tumors in vitro and in vivo. Experiments described in this communication indicate that the latent period of primary tumors induced by the injection of methylcholanthrene (MC) is also prolonged in these mice. This observation suggests that NR mechanisms may provide protection against primary chemically induced tumors.

Streptozotocin and Alloxan-based Selection Improves Toxin Resistance of Insulin-Producing RINm Cells

The aim of our study was to develop a method for
selection of subpopulations of insulin producing
RINm cells with higher resistance to beta cell toxins.
Cells, resistant to streptozotocin (RINmS) and
alloxan (RINmA), were obtained by repeated exposure
of parental RINm cells to these two toxins,
while the defense capacity, was estimated by the
MTT colorimetric method, and [3H]-thymidine incorporation
assay. We found that RINmS and
RINmA displayed higher resistance to both streptozotocin
(STZ) and alloxan (AL) when compared to
the parental RINm cells. In contrast, no differences
in sensitivity to hydrogen peroxide were found
between toxin selected and parental cells. Partial
protection from the toxic effect of STZ and AL was
obtained only in the parental RINm cells after
preincubation of cells with the unmetabolizable 3-
O-methyl-glucose. The possibility that GLUT-2 is
involved in cell sensitivity to toxins was confirmed
by Western blot analysis, which showed higher
expression of GLUT-2 in parental RINm compared
to RINmS and RINmA cells. In addition to the
higher cell defense property evidenced in the
selected cells, we also found higher insulin content
and insulin secretion in both RINmS and RINmA
cells when compared to the parental RINm cells. In
conclusion, STZ and AL treatment can be used for
selection of cell sub-populations with higher cell defense properties and hormone production. The
different GLUT-2 expression in parental and re
sistant cells suggest involvement of GLUT-2 in

The tight junction proteins (TJPs) are major determinants of endothelial cells comprising physiological vascular barriers such as the blood–brain barrier, but little is known about their expression and role in immune cells. In this study we assessed TJP expression in human leukocyte subsets, their induction by immune activation and modulation associated with autoimmune disease states and therapies. A consistent expression of TJP complexes was detected in peripheral blood leukocytes (PBLs), predominantly in B and T lymphocytes and monocytes, whereas the in vitro application of various immune cell activators led to an increase of claudin 1 levels, yet not of claudin 5. Claudins 1 and 5 levels were elevated in PBLs of multiple sclerosis (MS) patients in relapse, relative to patients in remission, healthy controls and patients with other neurological disorders. Interestingly, claudin 1 protein levels were elevated also in PBLs of patients with type 1 diabetes (T1D). Following glucocorticoid treatment of MS patients in relapse, RNA levels of JAM3 and CLDN5 and claudin 5 protein levels in PBLs decreased. Furthermore, a correlation between CLDN5 pre‐treatment levels and clinical response phenotype to interferon‐β therapy was detected. Our findings indicate that higher levels of leukocyte claudins are associated with immune activation and specifically, increased levels of claudin 5 are associated with MS disease activity. This study highlights a potential role of leukocyte TJPs in physiological states, and autoimmunity and suggests they should be further evaluated as biomarkers for aberrant immune activity and response to therapy in immune‐mediated diseases such as MS. 相似文献

In recent years, the possible association of changes in mortality from cardiovascular disease and myocardial infarction (MI) and deaths related to violence and the suicide rate has been repeatedly discussed. This study examined the relationship between cosmic physical changes (solar, geomagnetic and other space activity parameters) and changes in the total number of in-hospital and MI-related deaths and deaths from suicide to determine if a relationship exists between the distribution of total and MI-related deaths with suicide over time; some differences in the serotonergic mechanisms involved in the pathogenesis of MI and suicide were also taken into account. All suicides (n=2359) registered in the State of Israel from 1981 to 1989 (108 months) were analysed and compared with the total number of deaths (n=15601) and deaths from MI (n=1573) in a large university hospital over 180 months (1974–1989). The following were the main features of the Results. (1) Monthly suicide rate was correlated with space proton flux (r=0.42,P=0.0001) and with geomagnetic activity (r=–0.22,P=0.03). (2) Total hospital and MI-related deaths were correlated with solar activity parameters (r=0.35,P<0.001) and radiowave propagation (r=0.52-0.44,P<0.001), an with proton flux (r=–0.3 to –0.26,P<0.01). (3) Monthly suicide distribution over 108 months was correlated with MI (r=–0.33,P=0.0005) and total hospital mortality (r=–0.22,P=0.024). (4) Gender differences were prominent. We conclude that the monthly distributions of suicides and deaths from MI are adversely related to many environmental physical parameters and negatively correlated with each other. 相似文献
