Picosecond time-resolved fluorescence measurements have been performed as a function of emission wavelengths in order to investigate the possible functional differences between monomeric and trimeric Photosystem I (PS I) particles from a cyanobacterium Synechocystis. Applying global analysis, four kinetic components were found necessary to describe the fluorescecne decay for both monomers and trimers of PS I. The lifetimes and spectra of the respective components are quite similar, indicating that they can be attributed to identical processes in both the monomers and trimers. It is concluded that both forms of PS I are capable of efficient energy transfer and charge separation, in agreement with a physiological role of both forms. Small differences in the fluorescence decays are discussed in terms of a slightly higher ratio of red emitting pigments per reaction centre in trimers of PS I. Methods for the selection of transfectoma cells that express large quantities of mouse-human chimeric antibodies have been develped. SP2/0 mouse myeloma cells were transfected with pSV2-gpt and pSV2-neo based immunoglobulin expression vectors. Double transfectants were selected using the xanthine-guanine phosphoribosyl transferase (gpt)and the neomycin (neo) selection marker genes. ELISA-based screening of transfectoma clones resulted in the isolation of IgG-producing transfectomas. Introduction of the kappa light-chain 3'-enhancer into the light-chain expression vector significantly increased immunoglobulin expression, but only when the enhancer was located at its physiological site, 9 kb downstream of the kappa constant region exon. With some of the transfectomas, final yields of up to 80 mg/L of chimeric IgG were obtained in conventional flask cultures using serum-free growth medium. A pilot-scale AcuSyst Maximizer hollow fiber cell culture system was used for the production of gram amounts of chimeric IgG. Results obtained with different transfectoma clones in conventional culture were not fully predictive for yields in the hollow fiber system. In contrast, differences in productivity between individual clones in the laboratory-scale Tecnomouse cell culture unit were comparable with those in the Maximizer system. Abstract.   This review deals with those neuropeptidic hormones that are completely known in their primary structure together with the control processes that are linked directly or indirectly to development and reproduction. Actions are assessed for neuropeptides that regulate ecdysteroid and Juvenile Hormone production; oocyte growth or yolk deposition; and ecdysis and courtship behaviour. The combined data on the primary sequence, gene expression, biosynthesis, release, interaction with the receptor and mode of action provide an accurate account of the current knowledge on how these peptides function.  相似文献

Glutamine synthftase (GS) activity was investigated in a nitratt limited continuous culture of the marine diatom Chaeloccros afTinis (Lauder) Hustedt before and after the perturbation of the culture medium with 10 μM of 15 N labelled nitrate. Parallel studies were carried out on nitrate reductase(NR). nitrate uptake and assimilation, and Ievels of cellular nitrogen containing compounds with the objective to determine the validity of the GS assay as a measure of nitrate utilization. Activities in N-deficient cells, grown at steady state, correlated well with uptake and assimilation rates. In N-sufftcient celts, however, during the nitrate pertirbation period, they accounted only for about 10% of the two latter rates, when ambient nitrate concentrations were high (0. 7-10 μ). It is proposed that under these growth conditions an alternative pathway via glutamate dehydrogenase (GDH) was operative. At low ambient nitrate concentrations (0.1-0.7 μM), GS activities, uptake and assimilation rates again balanced rather well. Thus, the data support the view that GDH activity is associated with high levels and GS with low levels of external or internal nitrogen.  相似文献

Evidence for the Lower Bathonian Substage in most of the Pacific area has been lacking or controversial because index ammonoids of the Tethyan Subrealm in southern Europe and North Africa were unknown. During the Bathonian, the East Pacific margin belonged to either the Boreal Realm or the East-Pacific Subrealm of the Tethyan Realm, each with faunas distinctly different from the Eurafrican Tethyan faunas that form the basis for the chronostratigraphical standard. The first representatives of Morphoceras from South America, M. gulisanoi sp. nov., here described from a single locality in southern Mendoza Province, clearly document the Lower Bathonian Substage, probably the Zigzag Zone. Associated ammonoids are the perisphinctoidean Procerites cf. schloenbachi de Grossouvre and the oppeliid Oxycerites ( O. ) cf. aspidoides (Oppel), also typical Tethyan elements but less useful for dating.  相似文献

The pharmacokinetics of methadone were studied in beagle dogs (n = 4) following intravenous administration of the racemate (0.5 mg/kg) and of the individual (R)-(0.25 mg/kg) and (S)-enantiomers (0.25 mg/kg) using a stereospecific HPLC assay. There was no significant difference between the pharmacokinetic parameters of (R)-methadone and (S)-methadone following administration of the individual enantiomers. Stereoselective differences were evident following administration of the racemate (P values for differences in AUC and CL were 0.001 and 0.046, respectively) and the clearance of the (S)-enantiomer was increased when administered as part of the racemate (316 ± 81 vs 487 ± 128 ml/min, P = 0.04). The data suggest that stereoselective disposition including potential enantiomer–enantiomer interactions should be considered in pharmacokinetic–pharmacodynamic studies of (R,S)-methadone. © 1994 Wiley-Liss, Inc.  相似文献

Morphogenesis in pinoid mutants of Arabidopsis thaliana   总被引:6,自引:1,他引:5  
A series of mutants of Arabidopsis thaliana was selected in which the inflorescence stem elongates but loses the ability to produce flower primordia on its flanks. Mutants fell into two classes, further occurrences of pin-formed mutants and mutations at a new locus named pinoid. As well as causing inflorescence defects, pinoid mutations result in pleiotropic defects in the development of floral organs, cotyledons and leaves. Most changes involve the number of organs produced rather than their differentiation suggesting that PINOID controls an early general step in meristem development. pinoid mutant defects are similar to those seen in pin-formed mutants for inflorescences and flowers, but different for cotyledons and leaves indicating that the two genes have separate but overlapping functions. A defect in polar auxin transport is implicated in the pin-formed mutant phenotype, but in young inflorescence stems of even the strongest pinoid mutants it occurs at close to wild-type levels. It is markedly reduced only after stems have ceased elongating. Thus, it is likely that polar auxin transport is secondarily affected in pinoid mutants rather than being directly controlled by the PINOID gene product. Even so, double mutant studies indicate that the process controlled by PINOID overlaps with that specified by the AUXIN RESISTANT1 gene, suggesting that PINOID plays some role in an auxin-related process.  相似文献

We have cloned and sequenced an area of about 9.0 kb of the plastid DNA (ptDNA) from the holoparasitic flowering plant Cuscuta reflexa to investigate the evolutionary response of plastid genes to a reduced selective pressure. The region contains genes for the 16S rRNA, a subunit of a plastid NAD(P)H dehydrogenase (ndhB), three transfer RNAs (trnA, trnI, trnV) as well as the gene coding for the ribosomal protein S7 (rps7). While the other genes are strongly conserved in C. reflexa, the ndhB gene is a pseudogene due to many frameshift mutations. In addition we used heterologous gene probes to identify the other ndh genes encoded by the plastid genome in higher plants. No hybridization signals could be obtained, suggesting that these genes are either lost or strongly altered in the ptDNA of C. reflexa. Together with evidence of deleted genes in the ptDNA of C. reflexa, the plastid genome can be grouped into four classes reflecting a different evolutionary rate in each case. The phylogenetic position of Cuscuta and the significance of ndh genes in the plastid genome of higher plants are discussed.  相似文献
