With the depletion of oil resources in more accessible areas, those of remote regions are being considered or indeed are now being exploited. In many of these regions, especially the polar ones, little is known of the effects such exploitation will have on the environment. But it is known that the ecosystems are often subject to great stress by natural climatic conditions and additional burdens imposed by man may have catastrophic environmental effects. South Georgia, a sub-Antarctic island, has a history of industrial activity mainly concerned with whaling operations that peaked around 1925–1935 but has since declined to virtually nothing. Studies of the ecology of the area provided a unique opportunity to assess the long-term effects that such activities had on the ecosystem. Off the whaling stations a considerable amount of waste material, including fuel oil, was released into the bays and inevitably some of this material was deposited in the sediments. Chemical evidence in the form of both paraffinic and aromatic hydrocarbons still persists in the sediments. The implications of this persistence in relation to the possible influence of the low temperature conditions are discussed. The superficial sediments, marine biota and terrestrial plants, which since 1965 have returned virtually to a pristine state, contain hydrocarbons essentially similar to unpolluted areas around the coast of Britain. Relatively high levels of carcinogenic/mutagenic polynuclear aromatic hydrocarbons in surface sediments suggests a world-wide background of abiogenic hydrocarbons probably disseminated by airborne transport. This appears to indicate that contamination reaches even remote parts of the world in relatively undiminished quantities.  相似文献

Vaccines play a vital role in modern medicine. The development of novel vaccines for emerging and resistant pathogens has been aided in recent years by the use of novel adjuvants in subunit vaccines. A deeper understanding of the molecular pathways behind adjuvanticity is required to better select immunostimulatory molecules for use in individual vaccines. To this end, we have undertaken a study of the essential signaling pathways involved in the innate and adaptive immune responses to the Neisseria meningitidis outer membrane protein Porin B (PorB). We have previously demonstrated that PorB is an agonist of Toll-Like Receptor 2 (TLR2) and acts as an adjuvant in vaccines for protein, carbohydrate and lipopolysaccharide antigens using murine models. Here we demonstrate NFκB translocation following stimulation with PorB only occurs in the presence of TLR2. IL-6 and TNF-α secretion was shown to be MAPK dependent. Surface expression of activation markers on macrophages, including CD40, CD69, and CD86, was increased following PorB stimulation in vitro. Interestingly, some upregulation of CD54 and CD69 was still observed in macrophages obtained from TLR2 KO mice, indicating a possible non-TLR2 mediated activation pathway induced by PorB. In a murine vaccination model, using ovalbumin as the antigen and PorB as the adjuvant, a decreased antigen-specific IgG production was observed in TLR2 KO mice; adjuvant-dependent increased IgG production was entirely ablated in MyD88 KO mice. These observations demonstrate the importance of the above pathways to the adjuvant activity of PorB. The potential TLR2 independent effect is currently being explored.  相似文献

Animal models have been used for decades in the Alzheimer's disease (AD) research field and have been crucial for the advancement of our understanding of the disease. Most models are based on familial AD mutations of genes involved in the amyloidogenic process, such as the amyloid precursor protein (APP) and presenilin 1 (PS1). Some models also incorporate mutations in tau (MAPT) known to cause frontotemporal dementia, a neurodegenerative disease that shares some elements of neuropathology with AD. While these models are complex, they fail to display pathology that perfectly recapitulates that of the human disease. Unfortunately, this level of pre-existing complexity creates a barrier to the further modification and improvement of these models. However, as the efficacy and safety of viral vectors improves, their use as an alternative to germline genetic modification is becoming a widely used research tool. In this review we discuss how this approach can be used to better utilize common mouse models in AD research. This article is part of a Special Issue entitled: Animal Models of Disease.  相似文献
