The effects of ascorbic acid on dopaminergic 3H-agonist receptor binding were studied in membrane homogenates of bovine anterior pituitary and caudate, and rat striatum. In all tissues virtually no stereospecific binding (defined using luM (+)butaclamol) of the 3H-agonists N-propylnorapomorphine (NPA), apomorphine, or dopamine could be demonstrated in the absence of ascorbic acid. Although levels of total 3H-agonist binding were three to five times greater in the absence than in the presence of 0.1% ascorbic acid, the increased binding was entirely non-stereospecific. Greater amounts of dopamine-inhibitable 3H-NPA binding could be demonstrated in the absence of 0.1% ascorbic acid, but this measure of “specific binding” was demonstrated not to represent dopamine receptor binding since several other catecholamines and catechol were equipotent with dopamine and more potent than the dopamine agonist (±)amino-6,7-dihydroxy-1,2,3,4-tetrahydronapthalene (ADTN) in inhibiting this binding. High levels of dopamine-displaceable 3H-agonist binding were detected in fresh and boiled homogenates of cerebellum, an area of brain which receives no dopaminergic innervation, further demonstrating the non-specific nature of 3H-agonist binding in the absence of ascorbic acid. These studies emphasize that under typical assay conditions ascorbic acid is required in order to demonstrate reversible and specific 3H-agonist binding to dopamine receptors.  相似文献

A naturally occurring antibody that reacts with the keratinised tissue of animal oesophagus was found in the serum of 75 out of 129 patients (58%) with classical or definite rheumatoid arthritis (RA) but not in sera from 105 healthy people. Detection of the antibody, which is unrelated to rheumatoid factor, is more specific for RA than the reaction in the sheep-cell agglutination test but less sensitive.  相似文献

We have examined the role of rapidly turning over proteins in the T3 regulation of multiple rat hepatic genes. T3 induction of the rapidly responsive mRNA-S14 was markedly inhibited by cycloheximide (1 mg/100 g BW) or emetine (3 mg/100 g) injected ip 30 min before T3 (mRNA-S14 concentration was only 35% of that in T3-treated controls 8.5 h after administration of either protein synthesis inhibitor, P less than 0.01). Cycloheximide exhibited a similar effect on each of five other more slowly responsive T3 regulated genes. When cycloheximide was given 10 h after T3, the expected T3-induced rise of mRNA-S7 activity was completely prevented, and for mRNA-S4 activity the anticipated rise was blunted to 40% of T3-treated control (P less than 0.05). Cycloheximide caused sharp declines in the activity of two other mRNAs, S6 and S8, which because of shorter lag times of response to T3, had already risen when the drug was given. Values for both these mRNAs returned to the baseline hypothyroid level within 6 h of injection of the drug and remained low for a further 8 h (P less than 0.05). The expected deinduction of mRNA-S10 by T3 was also markedly modified. T3 lowered this mRNA to 11% of the hypothyroid control after 8 h, whereas cycloheximide given 30 min before the hormone blunted this fall to only 72% of control (P less than 0.01). Thus there appeared to be a 70% reduction in the rate of T3 induced fall of mRNA-S10. Seasonal content of total soluble carbohydrate, total soluble protein, free fatty acids and alkaloids were examined in a population of Ceratophyllum demersum growing in a pond on the southwestern edge of the Canadian Shield. Total soluble carbohydrate and total soluble protein were inversely correlated. Free fatty acids were dominated by (16:0 + 16:1), 18:2 and 18:3. Relative proportions of the different free fatty acids remained roughly constant throughout the season. Alkaloid concentrations showed large fluctuations, with the highest values found during the first half of the season. Carbohydrate and protein contents of associated periphyton were not correlated with host macrophyte metabolism.  相似文献

Photodynamic therapy (PDT) employs non-toxic dyes called photosensitizers (PSs), which absorb visible light to give the excited singlet state, followed by the long-lived triplet state that can undergo photochemistry. In the presence of ambient oxygen, reactive oxygen species (ROS), such as singlet oxygen and hydroxyl radicals are formed that are able to kill cancer cells, inactivate microbial pathogens and destroy unwanted tissue. Although there are already several clinically approved PSs for various disease indications, many studies around the world are using animal models to investigate the further utility of PDT. The present review will cover the main groups of animal models that have been described in the literature. Cancer comprises the single biggest group of models including syngeneic mouse/rat tumours that can either be subcutaneous or orthotopic and allow the study of anti-tumour immune response; human tumours that need to be implanted in immunosuppressed hosts; carcinogen-induced tumours; and mice that have been genetically engineered to develop cancer (often by pathways similar to those in patients). Infections are the second biggest class of animal models and the anatomical sites include wounds, burns, oral cavity, ears, eyes, nose etc. Responsible pathogens can include Gram-positive and Gram-negative bacteria, fungi, viruses and parasites. A smaller and diverse group of miscellaneous animal models have been reported that allow PDT to be tested in ophthalmology, atherosclerosis, atrial fibrillation, dermatology and wound healing. Successful studies using animal models of PDT are blazing the trail for tomorrow''s clinical approvals.  相似文献

As part of a study to assess the prevalence of common viral agents in African wildlife, nearly 3,300 sera from 44 different wild species, from eight African countries, have been examined for neutralising antibodies to parainfluenza 3 (PI3) virus. Antibody was demonstrated in 20 of the 44 species examined, including seven species not previously reported as sero-positive. Sera were collected between 1963 and 1977 and results indicated that infection has been widespread for a considerable time. The high prevalence of antibody, and the range of titres, to PI3 virus found in free-living populations of buffalo suggest that this species is particularly important as a reservoir of infection in the wild.  相似文献

Spring wheat cv. ‘Gutha’ was grown in continuous wheat (W/W) and narrow-leafed lupin (L. angustifolius L. cv. Yandee)-wheat (L/W) rotation on a yellow earth over mottled clay (Arenic Fragiudult) in a mediterranean climate for two years. The first year had a higher than average rainfall with adequate soil water until anthesis. The second year was very dry (only 232 mm total rainfall) and soil water contents were low throughout the growing season. Nitrogen fertilizer (+N) treatments were included in both years. In the first year an adjacent experiment compared the effects of loosening a pronounced traffic pan which existed on the site (LS)versus unloosened (US). In the first year roots contained more dry matter than tops in the early vegetative stage in all crops and then declined exponentially to a ratio of 0.1 in the US and LS treatments. In the second year however, the decline was both linear and much less, so that root:shoot ratios at harvest were still between 0.4 and 0.8. There was a consistent trend in root:shoot ratios from the most favourable (LS) to least favourable (W/W-N) treatments over the combined two years’ data, and this was also found in grain yield, with a higher yield in year one from the LS than US, and the lowest yield in year two from the W/W-N treatment. The proportion of total biomass recovered from below ground was substantially higher than is commonly reported from studies carried out in temperate, high fertility soils, but probably still under-estimates of the true amount of dry matter in roots because of inadequacies of sampling, washing and storage techniques. Root length densities were much greater in the drier year, especially in the surface 0.1-m, and based on theoretical considerations, much greater than required for extraction of available water. The effect of environmental conditions on the relative size of cereal crop carbon sinks are discussed in relation to these results.  相似文献
