The dentate gyrus is a site of continued neurogenesis in the adult brain. The CA3 region of the hippocampus is the major projection area from the dentate gyrus. CA3 sends reciprocal projections back to the dentate gyrus. Does this imply that CA3 exerts some control over neurogenesis? We studied the effects of lesions of CA3 on neurogenesis in the dentate gyrus, and on the ability of fluoxetine to stimulate mitotic activity in the progenitor cells. Unilateral ibotenic-acid generated lesions were made in CA3. Four days later there was no change on the number of either BrdU or Ki67-positive progenitor cells in the dentate gyrus. However, after 15 or 28 days, there was a marked reduction in surviving BrdU-labelled cells on the lesioned side (but no change in Ki-67+ cells). pCREB or Wnt3a did not co-localise with Ki-67 but with NeuN, a marker of mature neurons. Lesions had no effect on the basal expression of either pCREB or Wnt3a. Subcutaneous fluoxetine (10 mg/kg/day) for 14 days increased the number of Ki67+ cells as expected on the control (non-lesioned) side but not on that with a CA3 lesion. Nevertheless, the expected increase in BDNF, pCREB and Wnt3a still occurred on the lesioned side following fluoxetine treatment. Fluoxetine has been reported to decrease the number of “mature” calbindin-positive cells in the dentate gyrus; we found this still occurred on the side of a CA3 lesion. We then showed that the expression GAP-43 was reduced in the dentate gyrus on the lesioned side, confirming the existence of a synaptic connection between CA3 and the dentate gyrus. These results show that CA3 has a hitherto unsuspected role in regulating neurogenesis in the dentate gyrus of the adult rat. 相似文献

The effect of leaves of Quercus agrifolia, Cercis occidentalis, Eucalyptus globulus, and Juglans regia on the initial deposit and subsequent rate of decay of viable spores of Bacillus thuringiensis was investigated. Significant differences in the size of the initial viable spore deposit were found between the various species, with E. globulus markedly lower than the others. The viable spore decay patterns of the various species were analyzed using a segmented linear model and significant differences in initial rates of decay were also measured. Thus it appears that the characteristics of the leaves of the treated plants may have an effect on the coverage and field persistence of viable B. thuringiensis spores. Forty-eight Burkholderia isolates from different land use systems in the Amazon region were compared to type strains of Burkholderia species for phenotypic and functional characteristics that can be used to promote plant growth. Most of these isolates (n=46) were obtained by using siratro (Macroptilium atropurpureum - 44) and common bean (Phaseolus vulgaris - 2) as the trap plant species; two isolates were obtained from nodules collected in the field from Indigofera suffruticosa and Pithecellobium sp. The evaluated characteristics were the following: colony characterisation on "79" medium, assimilation of different carbon sources, enzymatic activities, solubilisation of phosphates, nitrogenase activity and antifungal activity against Fusarium oxysporium f. sp. phaseoli. Whole cell protein profiles, 16S rRNA, gyrB, and recA gene sequencing and multilocus sequence typing were used to identify the isolates. The isolates showed different cultural and biochemical characteristics depending on the legume species from which they were obtained. Except for one isolate from I. suffruticosa, all isolates were able to solubilise calcium phosphate and present nitrogenase activity under free-living conditions. Only one isolate from common beans, showed antifungal activity. The forty four isolates from siratro nodules were identified as B. fungorum; isolates UFLA02-27 and UFLA02-28, obtained from common bean plants, were identified as B. contaminans; isolate INPA89A, isolated from Indigofera suffruticosa, was a close relative of B. caribensis but could not be assigned to an established species; isolate INPA42B, isolated from Pithecellobium sp., was identified as B. lata. Aims Quantification of the effects and interactions of natural and anthropogenic factors, including climate, canopy structure, land use and management conditions, on vegetation burning. The study of these relationships is fundamental to predict regional fire patterns and develop sound management and regulation policies for biomass burning at national and global levels. Location Southern South America, including Argentina, Brazil, Paraguay, Uruguay, Bolivia and Chile. Methods Based on National Oceanic and Atmosphere Administration–Advance Very High Resolution Radiometer (NOAA–AVHRR) satellite images, we identified fires in southern South America with a daily frequency for two periods (1999/2000 and 2000/01) using a contextual fire detection algorithm and integrating the density of these fires at a monthly scale into a 0.5 × 0.5° grid. We combined vegetation and climate global databases and land use information from national census data to explore the relationship of these factors with fires across the region. Results The whole study region had a mean fire density of 0.10 and 0.05 fires km?2 year?1 in 1999/2000 and 2000/01, respectively, with extreme values as high as 1.37 in fires km?2 year?1 in Para State, Brazil. Water deficit estimates, derived from a climatic water balance, showed the better correlation with fire density (r = 0.28; P < 0.001; n = 4467), interacting strongly with land use. In areas with low agricultural use fire density increased with water deficit, whereas in highly agricultural areas this relationship was not observed. Agriculture significantly reduced fire density in prairies and savannas but increased its frequency in rain forests. Main conclusions These results suggest that agriculture prevents biomass burning in semiarid areas but enhances it in humid environments, where biomass accumulates at faster rates. 相似文献
