The subsynaptic distribution of kainate receptors is still a matter of much debate given its importance to understand the way they influence neuronal communication. Here, we show that, in synapses of the rat hippocampus, presynaptic kainate receptors are localized within the presynaptic active zone close to neurotransmitter release sites. The activation of these receptors with low concentrations of agonists induces the release of [(3)H]glutamate in the absence of a depolarizing stimulus. Furthermore, this modulation of [(3)H]glutamate release by kainate is more efficient when compared with a KCl-evoked depolarization that causes a more than two-fold increase in the intra-terminal calcium concentration but no apparent release of [(3)H]glutamate, suggesting a direct receptor-mediated process. Using a selective synaptic fractionation technique that allows for a highly efficient separation of presynaptic, postsynaptic and non-synaptic proteins we confirmed that, presynaptically, kainate receptors are mainly localized within the active zone of hippocampal synapses where they are expected to be in a privileged position to modulate synaptic phenomena. 相似文献

The golden mussel, Limnoperna fortunei a highly invasive species in Brazil, has generated productive, economical, and biological impacts. To evaluate genetic structure and variability of L. fortunei populations present in fish farms in the reservoirs of Canoas I (CANFF), Rosana (ROSFF), and Capivara (CAPFF) (Paranapanema River, Paraná, Brazil), eight microsatellite loci were amplified. Five of those eight loci resulted in 38 alleles. The observed heterozygosity (Ho) was lower than the expected heterozygosity (He) in all populations, with a deviation from the Hardy–Weinberg equilibrium (HWE). The average value for the inbreeding coefficient (Fis) was positive and significative for all populations. There was higher genetic variability within populations than among them. The fixation index (Fst) showed a small genetic variability among these populations. The occurrence of gene flow was identified in all populations, along with the lack of a recent bottleneck effect. The clustering analysis yielded K = 2, with genetic similarity between the three populations. The results demonstrate low genetic structure and suggest a founding population with greater genetic variability (ROSFF). Our data point to the possible dispersal of L. fortunei aided by anthropic factors in the upstream direction. It was concluded that the three populations presented a unique genetic pool for Paranapanema River, with occurrence of gene flow. 相似文献

Dengue fever vectored by the mosquito Aedes aegypti is one of the most rapidly spreading insect-borne diseases, stimulating the search for alternatives to current control methods. Screening assays using a range of Metarhizium anisopliae and Beauveria bassiana isolates were performed against adult female Ae. aegypti. Four virulent isolates were selected for detailed study. Adult female mosquitoes were exposed to supports previously inoculated with fungal suspensions. Fungal isolates were suspended in Tween 80+8% vegetable oil. The isolates caused between 70 and 89% mortality as a result of fungal infection over the 7-day test period. Mean survival times varied between 3 and 5 days for treated insects, whilst control survival exceeded 40 days. The most promising isolate, M. anisopliae LPP133, based not only on virulence but facility for mass production, was used for lethal exposure time determinations. An exposure time of only 3.5 h was necessary to cause 50% mortality. Large cage trails were also carried out and mean survival time of insects exposed to fungus impregnated black cloths was significantly reduced. These results show that entomopathogenic fungi could be promising biological control agents for use against adult Ae. aegypti, by inoculating fungi onto surfaces on which the mosquitoes tend to rest. The subsequent mortality caused by the fungi could potentially reduce the populations of this insect thus reducing the incidence of Dengue. 相似文献

Coffee is native to shady environments, but often grows and yields better without shade. Thus, it may be reasoned that coffee leaves should display enough plasticity to acclimate themselves to contrasting light environments. However, little is known about mechanisms associated with such plasticity in coffee. This work aimed, therefore, to explore differences in leaf photoprotective mechanisms. Plants were grown in the field and received either 48 or 100% natural light. Evaluations were made using outer leaves from the sun-facing sides of the coffee hedgerow in Viçosa (Brazil) in August and October, when growth and photosynthetic rates are expected to be minimal and maximal, respectively, and in December, when temporary depressions in those variables are common. Regardless of light treatments, coffee leaves showed: (1) very low photosynthetic rates (generally below 2.5 μmol m?2 s?1), (2) chronic photoinhibition in August (dry, cool season) that was accompanied by strong loss of pigment concentration, and (3) discrete, dynamic photoinhibition in October and December (rainy, warm season). Compared with shaded leaves, sunlit leaves generally exhibited lower pigment concentration, lower quantum yield of electron transport, steeper inclinations and similar electron transport rate. Total ascorbate pool tended to be larger in sunlit than in shaded leaves (but with similar redox state), whereas activities of key antioxidant enzymes, as well as malondialdehyde accumulation and electrolyte leakage, were similar between those leaf types. As a whole, the photosynthetic apparatus of the coffee tree showed a low phenotypic plasticity to varying irradiance. 相似文献

Efflux pumps are important defense mechanisms against antimicrobial drugs and maintenance of Burkholderia pseudomallei biofilms. This study evaluated the effect of the efflux pump inhibitor promethazine on the structure and antimicrobial susceptibility of B. pseudomallei biofilms. Susceptibility of planktonic cells and biofilms to promethazine alone and combined with antimicrobials was assessed by the broth microdilution test and biofilm metabolic activity was determined with resazurin. The effect of promethazine on 48 h-grown biofilms was also evaluated through confocal and electronic microscopy. The minimum inhibitory concentration (MIC) of promethazine was 780 mg l?1, while the minimum biofilm elimination concentration (MBEC) was 780–3,120 mg l?1. Promethazine reduced the MIC values for erythromycin, trimethoprim/sulfamethoxazole, gentamicin and ciprofloxacin and reduced the MBEC values for all tested drugs (p<0.05). Microscopic analyses demonstrated that promethazine altered the biofilm structure of B. pseudomallei, even at subinhibitory concentrations, possibly facilitating antibiotic penetration. This study compared the structure of metazoan parasite communities in two populations of Leporinus friderici from two rivers of the Amazon River system in Brazil. Jainus leporini, Urocleidoides paradoxus, Urocleidoides sp., Tereancistrum parvus, Tereancistrum sp., Clinostomum marginatum, Procamallanus (Spirocamallanus) inopinatus, Contracaecum sp., Octospiniferoides incognita and Ergasilus sp. were found in both hosts. There were differences in species richness of parasites, Shannon index and the evenness between both host populations, because the parasite community showed a similarity of only 33%. Parasites of both host populations had an aggregated dispersion. Host size also contributes to the differences between the populations of hosts investigated, and the feeding habit contributes to the occurrence of endoparasites in L. friderici, an intermediate host for these parasites. The existence of variation in infracommunity and community of parasites for the same host species from different localities indicates the presence of an uneven distribution in terms of species and density of parasites, which are intermediate hosts, and some of them constitute food items for L. friderici in the localities surveyed. 相似文献

Hepatitis C infection is a cause of chronic liver diseases such as cirrhosis and carcinoma. The current therapy for hepatitis C has limited efficacy and low tolerance. The HCV encodes a serine protease which is critical for viral replication, and few protease inhibitors are currently on the market. In this paper, we describe the synthesis and screening of novel isosorbide-based peptidomimetic inhibitors, in which the compounds 1d, 1e, and 1i showed significant inhibition of the protease activity in vitro at 100 µM. The compound 1e also showed dose-response (IC50 = 36 ± 3 µM) and inhibited the protease mutants D168A and V170A at 100 µM, indicating it as a promising inhibitor of the HCV NS3/4A protease. Our molecular modeling studies suggest that the activity of 1e is associated with a change in the interactions of S2 and S4 subsites, since that the increased flexibility favors a decrease in activity against D168A, whereas the appearance of a hydrophobic cavity in the S4 subsite increase the inhibition against V170A strain. 相似文献

Males of solitary bees usually spend the night out of the nests. In the middle or late afternoon, they stop the patrolling behavior and move on to their sleeping places. Usually, they hang with the mandibles on small branches of the vegetation or stay inside flowers until the next day. We report the sleeping places of males of four Tapinotaspidini species on flowers of six plant species of four families. Flowers of three Iridaceae species were the most sought by males, especially flowers of Sisyrinchium scariosum which show high synchrony between anthesis and activity period of Lanthanomelissa discrepans males. Moreover, S. scariosum flowers are the most visited by females of L. discrepans which are the main pollinators; however, the role of the males as pollinators is unclear. Similar situation is evident for the interaction between males of Arhysoceble picta and Cuphea glutinosa (Lythraceae), where the males take nectar and may act as pollinators, like their females. We believe the plants are indirectly benefited by these interactions through the maintenance of the male populations of the pollinator bee species. 相似文献
