Aim

This study presents a micrometre-scale map of the elemental distribution within roots and surrounding sediment of Halimione portulacoides of a contaminated salt marsh in the Tagus estuary.

Methods

Microprobe particle induced X-ray emission analysis was performed in sediment slices containing roots with tubular rhizoconcretions attached to host sediments.

Results

Strong concentration gradients were found particularly in the inner part of rhizoconcretions adjacent to the root wall. Local enrichment was observed in sediment interstices with Fe precipitates and other associated elements. A maximum of 55 % of Fe was measured near the concretion–root interface, with a decrease to <5 % in the host sediment. Maximum concentrations of P (3 %), As (1,200 μg g?1) and Zn (3,000 μg g?1) were registered in concretions, one order of magnitude above the values of the host sediment. The elemental concentration profiles across roots showed that the epidermis was an efficient selective barrier to the entrance of elements. Fe and As were retained in the epidermis. The highest Cu and Zn concentrations were also observed in the epidermis. However, the concentrations of Mn, Cu and Zn increased in the inner root.

Conclusions

As and Fe were mostly retained in the concretion, whereas P, Mn, Cu and Zn were mobilised by the root.  相似文献

Neospora caninum (Apicomplexa; Sarcocystidae) is a protozoan that causes abortion in cattle, horses, sheep, and dogs as well as neurological and dermatological diseases in dogs. In the central nervous system of dogs infected with N. caninum, cysts were detected that exhibited gliosis and meningitis. Flavonoids are polyphenolic compounds that exhibit antibacterial, antiparasitic, antifungal, and antiviral properties. In this study, we investigated the effects of flavonoids in a well-established in vitro model of N. caninum infection in glial cell cultures. Glial cells were treated individually with 10 different flavonoids, and a subset of cultures was also infected with the NC-1 strain of N. caninum. All of the flavonoids tested induced an increase in the metabolism of glial cells and many of them increased nitrite levels in cultures infected with NC-1 compared to controls and uninfected cultures. Among the flavonoids tested, 3'',4''-dihydroxyflavone, 3'',4'',5,7-tetrahydroxyflavone (luteolin), and 3,3'',4'',5,6-pentahydroxyflavone (quercetin), also inhibited parasitophorous vacuole formation. Taken together, our findings show that flavonoids modulate glial cell responses, increase NO secretion, and interfere with N. caninum infection and proliferation.  相似文献

Mass flowering is a widespread blooming strategy among Neotropical trees that has been frequently suggested to increase geitonogamous pollination. We investigated the pollination ecology of the mass‐flowering tree Handroanthus impetiginosus, addressing its breeding system, the role in pollination of different visitors, the impact of nectar robbers on fruit set and the function of colour changes in nectar guides. This xenogamous species is mainly pollinated by Centris and Euglossa bees (Apidae) seeking nectar, which are known to fly long distances. The flowers favour these bees by having: (1) a closed entrance in newly opened flowers which provides access only to strong bees capable of deforming the flower tube; and (2) a nectar chamber that is accessible only to long‐tongued bees. Only first‐day flowers with yellow nectar guides produce nectar. Pollinators prefer these flowers over second‐ and third‐day flowers with orange and red nectar guides, respectively. Nectar robbers damage two‐thirds of the flowers and this robbing activity decreases fruit set by half. We attribute the low fruit set of H. impetiginosus to the intense nectar robbing and hypothesize that visual signalling of nectar presence in newly opened (receptive) flowers reduces geitonogamy by minimizing bee visits to unrewarding (non‐receptive) flowers. © 2014 The Linnean Society of London, Botanical Journal of the Linnean Society, 2014, 176 , 396–407.  相似文献

Objectives

To quantify resting myocardial blood flow (MBF) in the left ventricular (LV) wall of HCM patients and to determine the relationship to important parameters of disease: LV wall thickness, late gadolinium enhancement (LGE), T2-signal abnormalities (dark and bright signal), LV outflow tract obstruction and age.

Materials and Methods

Seventy patients with proven HCM underwent cardiac MRI. Absolute and relative resting MBF were calculated from cardiac perfusion MRI by using the Fermi function model. The relationship between relative MBF and LV wall thickness, T2-signal abnormalities (T2 dark and T2 bright signal), LGE, age and LV outflow gradient as determined by echocardiography was determined using simple and multiple linear regression analysis. Categories of reduced and elevated perfusion in relation to non- or mildly affected reference segments were defined, and T2-signal characteristics and extent as well as pattern of LGE were examined. Statistical testing included linear and logistic regression analysis, unpaired t-test, odds ratios, and Fisher’s exact test.

Results

804 segments in 70 patients were included in the analysis. In a simple linear regression model LV wall thickness (p<0.001), extent of LGE (p<0.001), presence of edema, defined as focal T2 bright signal (p<0.001), T2 dark signal (p<0.001) and age (p = 0.032) correlated inversely with relative resting MBF. The LV outflow gradient did not show any effect on resting perfusion (p = 0.901). Multiple linear regression analysis revealed that LGE (p<0.001), edema (p = 0.026) and T2 dark signal (p = 0.019) were independent predictors of relative resting MBF. Segments with reduced resting perfusion demonstrated different LGE patterns compared to segments with elevated resting perfusion.

Conclusion

In HCM resting MBF is significantly reduced depending on LV wall thickness, extent of LGE, focal T2 signal abnormalities and age. Furthermore, different patterns of perfusion in HCM patients have been defined, which may represent different stages of disease.  相似文献

Using 2-DE of total cell protein extracts, we compared soluble proteins from murine melanoma lines Tm1 and Tm5 with proteins from the nontumoral cell melan-a from which they were derived. Seventy-one of the 452 spots (average) detected with CBB were differentially accumulated, i.e., increased or decreased twofold. Forty-four spots were identified by PMF/MALDI-TOF, 15 with increased and 29 with decreased protein levels. SAGE showed that 17/34 (50%) of the differentially accumulated proteins, pI range 4-7, presented similar differences at the mRNA level. Major reductions in protein were observed in tumor cells of proteins that degrade reactive oxygen species (ROS). Decreases of > or = twofold in GST, superoxide dismutase, aldehyde dehydrogenase, thioredoxin, peroxiredoxin 2, and peroxiredoxin 6 protein were observed. SAGE indicated the reduction of other proteins involved in ROS degradation. As expected, the accumulation of exogenous peroxides was significantly higher in the tumor cells while the levels of glutathionylation were two times lower in the tumor cells compared to melan-a. The differential accumulation of proteins involved in oncogene/tumor suppressor pathways was observed. Melanoma cells can favor survival pathways activated by ROS by inhibiting p53 pathways and activation of Ras and c-myc pathways.  相似文献

Efflux pumps are important defense mechanisms against antimicrobial drugs and maintenance of Burkholderia pseudomallei biofilms. This study evaluated the effect of the efflux pump inhibitor promethazine on the structure and antimicrobial susceptibility of B. pseudomallei biofilms. Susceptibility of planktonic cells and biofilms to promethazine alone and combined with antimicrobials was assessed by the broth microdilution test and biofilm metabolic activity was determined with resazurin. The effect of promethazine on 48 h-grown biofilms was also evaluated through confocal and electronic microscopy. The minimum inhibitory concentration (MIC) of promethazine was 780 mg l?1, while the minimum biofilm elimination concentration (MBEC) was 780–3,120 mg l?1. Promethazine reduced the MIC values for erythromycin, trimethoprim/sulfamethoxazole, gentamicin and ciprofloxacin and reduced the MBEC values for all tested drugs (p<0.05). Microscopic analyses demonstrated that promethazine altered the biofilm structure of B. pseudomallei, even at subinhibitory concentrations, possibly facilitating antibiotic penetration. Promethazine improves antibiotics efficacy against B. pseudomallei biofilms, by disrupting biofilm structure.  相似文献

Hepatitis C infection is a cause of chronic liver diseases such as cirrhosis and carcinoma. The current therapy for hepatitis C has limited efficacy and low tolerance. The HCV encodes a serine protease which is critical for viral replication, and few protease inhibitors are currently on the market. In this paper, we describe the synthesis and screening of novel isosorbide-based peptidomimetic inhibitors, in which the compounds 1d, 1e, and 1i showed significant inhibition of the protease activity in vitro at 100 µM. The compound 1e also showed dose-response (IC50 = 36 ± 3 µM) and inhibited the protease mutants D168A and V170A at 100 µM, indicating it as a promising inhibitor of the HCV NS3/4A protease. Our molecular modeling studies suggest that the activity of 1e is associated with a change in the interactions of S2 and S4 subsites, since that the increased flexibility favors a decrease in activity against D168A, whereas the appearance of a hydrophobic cavity in the S4 subsite increase the inhibition against V170A strain.  相似文献

This study compared the structure of metazoan parasite communities in two populations of Leporinus friderici from two rivers of the Amazon River system in Brazil. Jainus leporini, Urocleidoides paradoxus, Urocleidoides sp., Tereancistrum parvus, Tereancistrum sp., Clinostomum marginatum, Procamallanus (Spirocamallanus) inopinatus, Contracaecum sp., Octospiniferoides incognita and Ergasilus sp. were found in both hosts. There were differences in species richness of parasites, Shannon index and the evenness between both host populations, because the parasite community showed a similarity of only 33%. Parasites of both host populations had an aggregated dispersion. Host size also contributes to the differences between the populations of hosts investigated, and the feeding habit contributes to the occurrence of endoparasites in L. friderici, an intermediate host for these parasites. The existence of variation in infracommunity and community of parasites for the same host species from different localities indicates the presence of an uneven distribution in terms of species and density of parasites, which are intermediate hosts, and some of them constitute food items for L. friderici in the localities surveyed.  相似文献

Males of solitary bees usually spend the night out of the nests. In the middle or late afternoon, they stop the patrolling behavior and move on to their sleeping places. Usually, they hang with the mandibles on small branches of the vegetation or stay inside flowers until the next day. We report the sleeping places of males of four Tapinotaspidini species on flowers of six plant species of four families. Flowers of three Iridaceae species were the most sought by males, especially flowers of Sisyrinchium scariosum which show high synchrony between anthesis and activity period of Lanthanomelissa discrepans males. Moreover, S. scariosum flowers are the most visited by females of L. discrepans which are the main pollinators; however, the role of the males as pollinators is unclear. Similar situation is evident for the interaction between males of Arhysoceble picta and Cuphea glutinosa (Lythraceae), where the males take nectar and may act as pollinators, like their females. We believe the plants are indirectly benefited by these interactions through the maintenance of the male populations of the pollinator bee species.  相似文献
