【目的】水产养殖过程中蜡状芽孢杆菌(Bacillus cereus)作为益生菌被广泛应用。本研究旨在深入了解分离于养殖水体中的一株蜡状芽孢杆菌S458-1对养殖水体以及养殖对象的影响,以期为菌株S458-1在水产养殖生产上的应用提供理论依据。【方法】根据控制变量法,各试验组鲫鱼养殖模式设置相同温度、盐度、溶氧和pH,利用分光光度法测定水体的水化学指标;利用试剂盒法测定鲫鱼的血清生理生化指标。【结果】添加菌株S458-1处理组(终浓度分别为10~6、10~7、10~8CFU/mL)与对照组(未加S458-1菌)相比:水体活性磷浓度显著性增加(P0.05);同时具有把NH_4~+-N和NO_2~–-N转化为NO_3~–-N的趋势,但无显著性差异(P0.05);养殖鲫鱼血清检测结果显示谷草转氨酶(GOT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)以及丙二醛(MDA)含量均显著降低(P0.05)。【结论】蜡状芽孢杆菌S458-1具有脱氮解磷、调节水质的作用,并可增强养殖对象的生理健康状态,可作为益生菌应用于水产养殖中,具有较高的应用价值。  相似文献

The monocyte locomotion inhibitory factor (MLIF) is an anti-inflammatory oligopeptide produced by Entamoeba histolytica. Among its different effects, it inhibits locomotion of human monocytes, hence its original name. The carboxyl-terminal end group Cys-Asn-Ser is the pharmacophore of anti-inflammatory peptide Met-Gln-Cys-Asn-Ser. In this study, the N-terminal of Cys-Asn-Ser was modified. With the aim to enhance the antioxidant ability and penetrability of Cys-Asn-Ser, we designed and synthesized two tetrapeptides Tyr-Cys-Asn-Ser and His-Cys-Asn-Ser. The neuroprotective effects of Tyr-Cys-Asn-Ser and His-Cys-Asn-Ser on focal ischemia reperfusion were investigated, and their pharmacological activities compared with Cys-Asn-Ser were studied. In order to study the mechanism of neuroprotective effect of these peptides, the level of oxidative stress markers malondialdehyde (MDA) and superoxide dismutase (SOD) and pro-inflammatory factors interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), and myeloperoxidase (MPO) were detected in brain tissue homogenate.  相似文献

In the brains of Alzheimer's disease patients, the tau protein dissociates from the axonal microtubule and abnormally aggregates to form a paired helical filament (PHF). One of the priorities in Alzheimer research is to clarify the mechanism of PHF formation. Although several reports on the regulation of tau assembly have been published, it is not yet clear whether in vivo PHFs are composed of beta-structures or alpha-helices. Since the four-repeat microtubule-binding domain (4RMBD) of the tau protein has been considered to play an essential role in PHF formation, its heparin-induced assembly propensity was investigated by the thioflavin fluorescence method to clarify what conformation is most preferred for the assembly. We analyzed the assembly propensity of 4RMBD in Tris-HCl buffer with different trifluoroethanol (TFE) contents, because TFE reversibly induces the transition of the random structure to the alpha-helical structure in an aqueous solution. Consequently, it was observed that the 4RMBD assembly is most significantly favored to proceed in the 10-30% TFE solution, the concentration of which corresponds to the activated transition state of 4RMBD from a random structure to an alpha-helical structure, as determined from the circular dichroism (CD) spectral changes. Since such an assembly does not occur in a buffer containing TFE of < 10% or > 40%, the intermediate conformation between the random and alpha-helical structures could be most responsible for the PHF formation of 4RMBD. This is the first report to clarify that the non-native alpha-helical intermediate in transition from random coil is directly associated with filament formation at the start of PHF formation.  相似文献

High mobility group box-1 protein (HMGB1) had been proved to induce maturation and activation of dendritic cell (DC), however, the endogenous changes and mechanisms underlying are unknown. Since endoplasmic reticulum stress (ERS) activates an adaptive unfolded protein response (UPR) that facilitates cellular survival and repair, we hypothesized that HMGB1 may regulate the function of DC by modulating ERS. In our study, HMGB1 stimulation induced significant ERS responses in DCs in a time- and dose-dependent manner, demonstrated by the up-regulation of a number of ERS markers. Gene silence of XBP-1 in splenic DCs decreased the levels of CD80, CD86 as well as major histocompatibility complex (MHC)-II expression and cytokine secretion after HMGB1 treatment, when compared with untransfected or nontargeting-transfected DCs (all P<0.05). Moreover, XBP-1 silenced DCs after treatment with HMGB1 failed to stimulate notable proliferation and differentiation of T cells, unlike normal DCs or nontargeting-transfected DCs (all P<0.05). Gene silence of XBP-1 resulted in down-regulation of the receptor for advanced glycation end products (RAGE) expression on the surface of splenic DCs induced by HMGB1 stimulation (P<0.05). These findings demonstrate an important role for ERS and its regulator XBP-1 in HMGB1-induced maturation and activation of DCs.  相似文献

Three new compounds, 1 – 3 , and 20 known compounds were isolated from the AcOEt and BuOH extract of edible Opuntia Milpa Alta. The petroleum ether extract was examined by GC and MS. A total of 26 compounds were identified, representing 95.6% of the total extract, phytosterol (36.03%) being the most abundant component, and polyunsaturated fatty acids (18.57%) represented the second largest group, followed by phytol (12.28%), palmitic acid, palmitate (13.54%), vitamin E (4.51%), and other compounds (7.47%). The effects of various extracts from edible Opuntia Milpa Alta (petroleum ether extract, AcOEt extract, BuOH extract, aqueous extract, H2O parts) and the positive control (received dimethylbiguanide) were tested on streptozotocin (STZ)‐induced diabetic mice. The results indicated that all the treatment groups could significantly decrease blood glucose levels in STZ‐induced diabetic mice compared to the model control group (P<0.01), except the aqueous extract group (P<0.05). Especially, the petroleum ether extract group and the positive control group showed remarkable decrease of blood glucose levels. Taken together, the results indicate that the petroleum ether extract is the major hypoglycemic part in edible Opuntia Milpa Alta, which may be developed to a potential natural hypoglycemic functional ingredient.  相似文献

Organic matter addition is thought to be an important regulator of nitrous oxide (N2O) emissions from croplands. Contradictory effects, however, were reported in previous studies. To investigate the effects of crop residue management on N2O emissions from rice-wheat rotation ecosystems, we conducted field experiments at three sites (Suzhou, Wuxi and Jiangdu) in the Yangtze River Delta, using static chamber and gas chromatography methods. Our data show that N2O emissions throughout the rice season from plots treated with wheat straw application at a high rate (WS) prior to rice transplanting (1.1–2.0 kg N ha?1) were significantly lower (P?<?0.05) than those from the control plots without organic matter addition or added with wheat straw at a moderate rate (1.6–2.9 kg N ha?1). Furthermore, the WS treatments had a residual inhibitory effect on N2O emissions in the following non-rice season, which consistently resulted in significantly lower emissions (P?<?0.05) compared to the control treatments (2.2–3.1 vs. 3.9–5.6 kg N ha?1). In comparison to the control treatments, the WS treatments reduced both the seasonal and annual direct emission factors of the applied nitrogen (EFd) by 50–68% (mean: 57%). The addition of compost (aerobically composted rice or wheat straw harvested in the last rotation) reduced the seasonal and annual EFds by 29–32%. Over the entire rice-wheat rotation cycle, annual N2O emissions from the fertilized fields at the three sites ranged from 3.3?±?0.3 to 16.8?±?0.6 kg N ha?1, with a coefficient of variation (CV) of 61%. Similarly, the EFds during the rice-wheat rotation cycle ranged from 0.4% to 2.5%, with a CV of 67%. These high spatial variations might have been related to: variations in soil properties, such as texture and soil organic carbon; management practices, such as straw treatments (i.e., compost versus fresh straw) and weather conditions, such as precipitation and rainfall distribution. Our results indicate that the incorporation of fresh wheat straw at a high rate during the rice season is an effective management practice for the mitigation of N2O emissions in rice-wheat rotation systems. Whether this practice is also effective in reducing the overall global warming potential of net N2O, CH4 and CO2 emissions needs to be seen through further studies.  相似文献

Platelet gels (PG) are new topical single-donor blood products which are attracting great interest in regenerative medicine. They are obtained by mixing a platelet-rich plasma fraction with thrombin to generate a fibrin gel enriched in platelet growth factors (GF). The type of thrombin preparation may affect PG reproducibility. We have determined the impact of 14.6% (v/v) ethanol-stabilized thrombin (EHT) on the release of GF by platelets. Various ratios of EHT and platelet concentrates were mixed to obtain from 2.43 to 7.96% ethanol concentration. Platelet-derived growth factor-AB (PDGF-AB), transforming growth factor-ß1 (TGF-ß1), vascular endothelium growth factor (VEGF), epidermal growth factor (EGF), and insulin-like growth factor-1 (IGF-1) were assessed at 5, 120, and 300 min after PG formation. Protein profiles of thrombin and PG releasates were analyzed by SDS-PAGE. The amount of PDGF-AB, TGF-ß1, and VEGF released per platelet decreased significantly (p < 0.05) with increasing ethanol concentrations but, however, not that of EGF. IGF-1 content was stable, consistent with its presence mostly in plasma. SDS-PAGE indicated that ethanol did not affect fibrin formation. In conclusion, ethanol has a significant impact on the amount of GF released by platelets and should be strictly controlled to standardize PG and optimize clinical benefits.  相似文献

A computational method independent of experimental protein structure information is proposed to recognize key residues in protein folding, from the study of hydration water dynamics. Based on all-atom molecular dynamics simulation, two key residues are recognized with distinct water dynamical behavior in a folding process of the Trp-cage protein. The identified key residues are shown to play an essential role in both 3D structure and hydrophobic-induced collapse. With observations on hydration water dynamics around key residues, a dynamical pathway of folding can be interpreted.  相似文献
