The binding of Bacteroides fragilis to plasmatic fibronectin was investigated using strains isolated from healthy subjects and from patients with bacteremia. They were cultivated in a synthetic media in which variations in cysteine concentrations determined alterations in the oxidation–reduction potential (Eh). All the strains assayed were capable of adhering to plasmatic fibronectin when cultivated under oxidizing and reducing conditions. Bacteroides fragilis 1405 showed the greatest difference when the results under these conditions were compared and it was selected for further investigations. Chemical treatments suggested the involvement of a protein in the interaction between B. fragilis and plasmatic fibronectin. Sodium dodecyl sulfate polyacrylamide gel electrophoresis analysis of outer membrane proteins (OMPs) revealed differences between the extracts obtained from cultures grown under the two conditions. Protein bands of c . 102, 100, 77, 73, 50 and 40 kDa were more highly expressed under oxidizing than reducing conditions. Dot blot analysis showed a stronger recognition of plasmatic fibronectin by OMPs obtained from cultures grown under higher Eh, and Western blot assays confirmed a band of c . 102 kDa as fibronectin-binding protein. This protein was sequenced and revealed to be a putative TonB-dependent OMPs. PCR analysis confirmed the presence of this gene in all the studied strains. 相似文献

Anti-human immunodeficiency virus type 1 (HIV-1) "binding antibodies" (antibodies capable of binding to synthetic peptides or proteins) occur throughout HIV-1 infection, are high-titered and highly cross-reactive, as confirmed in this study by analyzing plasma from B and F genotype HIV-1 infected individuals. Plasma from individuals infected with clade F HIV-1 displayed the most frequent cross-reactivity, in high titers, while Bbr plasma showed much higher specificity. Similarly, neutralization of a reference HIV-1 isolate (HIV-1 MN) was more frequently observed by plasma from F than B genotype infected individuals. No significant difference was seen in neutralization susceptibility of primary B, Bbr or F clade HIV-1 by plasma from individuals infected with the classical B (GPGR) or F HIV-1, but Bbr (GWGR) plasma were less likely to neutralize the F genotype primary HIV-1 isolates. The data indicate that both B and F genotype derived vaccines would be equally effective against B and F HIV-1 infection, with a slightly more probable effectiveness for F than B genotype. Although the Bbr variant appears to induce a much more specific humoral immune response, the susceptibility in neutralizing the Brazilian HIV-1 B genotype Bbr variant is similar to that observed with the classical B genotype HIV-1. 相似文献

Background
Older adults are often excluded from clinical trials. Decision making for administration of statins to older patients with diabetes mellitus (DM) is under debate, particularly in frail older patients with comorbidity and high mortality risk. We tested the hypothesis that statin treatment in older patients with DM was differentially effective across strata of mortality risk assessed by the Multidimensional Prognostic Index (MPI), based on information collected with the Standardized Multidimensional Assessment Schedule for Adults and Aged Persons (SVaMA).Methods
In this retrospective observational study, we estimated the mortality risk in 1712 community-dwelling subjects with DM ≥ 65 years who underwent a SVaMA evaluation to establish accessibility to homecare services/nursing home admission from 2005 to 2013 in the Padova Health District, Italy. Mild (MPI-SVaMA-1), moderate (MPI-SVaMA-2), and high (MPI-SVaMA-3) risk of mortality at baseline and propensity score-adjusted hazard ratios (HR) of three-year mortality were calculated according to statin treatment.Results
Higher MPI-SVaMA scores were associated with lower rates of statin treatment (MPI-SVaMA-1 = 39% vs MPI-SVaMA-2 = 36% vs MPI-SVaMA-3 = 24.9%. p<0.001) and higher three-year mortality (MPI-SVaMA-1 = 12.9% vs MPI-SVaMA-2 = 24% vs MPI-SVaMA-3 = 34.4%, p<0.001). After adjustment for propensity score quintiles, statin treatment was significantly associated with lower three-year mortality irrespective of MPI-SVaMA group (interaction test p = 0.303). HRs [95% confidence interval (CI)] were 0.19 (0.14–0.27), 0.28 (0.21–0.36), and 0.26 (0.20–0.34) in the MPI-SVaMA-1, MPI-SVaMA-2, and MPI-SVaMA-3 groups, respectively. Subgroup analyses showed that statin treatment was also beneficial irrespective of age. HRs (95% CI) were 0.21 (0.15–0.31), 0.26 (0.20–0.33), and 0.26 (0.20–0.35) among patients aged 65–74, 75–84, and ≥ 85 years, respectively (interaction test p=0.812).Conclusions
Statin treatment was significantly associated with reduced three-year mortality independently of age and multidimensional impairment in community-dwelling frail older patients with DM. 相似文献14.

While there has been increasing interest in how taxonomic diversity is changing over time, less is known about how long‐term taxonomic changes may affect ecosystem functioning and resilience. Exploring long‐term patterns of functional diversity can provide key insights into the capacity of a community to carry out ecological processes and the redundancy of species’ roles. We focus on a protected freshwater system located in a national park in southeast Germany. We use a high‐resolution benthic macroinvertebrate dataset spanning 32 years (1983–2014) and test whether changes in functional diversity are reflected in taxonomic diversity using a multidimensional trait‐based approach and regression analyses. Specifically, we asked: (i) How has functional diversity changed over time? (ii) How functionally distinct are the community''s taxa? (iii) Are changes in functional diversity concurrent with taxonomic diversity? And (iv) what is the extent of community functional redundancy? Resultant from acidification mitigation, macroinvertebrate taxonomic diversity increased over the study period. Recovery of functional diversity was less pronounced, lagging behind responses of taxonomic diversity. Over multidecadal timescales, the macroinvertebrate community has become more homogenous with a high degree of functional redundancy, despite being isolated from direct anthropogenic activity. While taxonomic diversity increased over time, functional diversity has yet to catch up. These results demonstrate that anthropogenic pressures can remain a threat to biotic communities even in protected areas. The differences in taxonomic and functional recovery processes highlight the need to incorporate functional traits in assessments of biodiversity responses to global change. 相似文献

The oxidation-reduction (redox) of the environment characterizes the Bacteroides fragilis pathogenic potential. Previously, using 3D confocal laser scanning microscopy, the bacteria prepared from cultures grown under oxidizing conditions (Eh(7)ca. + 100 mV) were able to penetrate into Hela cell monolayers. In contrast, when grown under reducing conditions (Eh(7)ca. - 60 mV), there were no bacteria evident within Hela cells. The influence of the anaerobe/aerobe environmental flux during the process of the anaerobe infection could be significant. In B. fragilis peritonitis, this may depend on the occurrence of aerobiosis as opposed to anaerobiosis. To this end, three clinical B. fragilis strains, two infectious and one non-infectious, were grown under oxidizing and reducing conditions; then, the outer membrane protein expressions derived from these strains were assessed, following sarcosyl extraction and SDS-PAGE. The differences between the protein profiles from these strains when cultured under oxidizing and reducing conditions were found to be statistically significant for the two infectious strains, but not for the non-infectious strain. OMP profiles under aerobic conditions compared to anaerobic conditions exhibited products with a range of apparent molecular weights suggestive of unique participation in the interaction with the host cell. 相似文献
