The mechanism by which the rate of cell proliferation is regulated in different regions of the root apical meristem is unknown. The cell populations comprising the root cap and meristem cycle at different rates, proliferation being particularly slow in the quiescent centre. In an attempt to detect the control points in the cell cycle of the root apical meristem of Zea mays L. (cv. LG 11), quiescent-centre cells were stimulated to synthesise DNA and to enter mitosis either by decapping or by immersing intact roots in an aqueous 3,3-dimethyl-glutaric acid buffer solution. From microdensitometric and flow-cytometric data, we conclude that, upon immersion, the G2 phase of the cell cycle of intact roots was shortened. However, when 50 M abscisic acid (ABA) was added to the immersion buffer, parameters of the cell cycle were restored to those characteristic of intact roots held in a moist atmosphere. On the other hand, decapping of primary roots preferentially shortened the G1 phase of the cell cycle in the quiescent centre. When supplied to decapped roots, ABA reversed this effect. Therefore, in our model, applied ABA retarded the completion of the cell cycle and acted upon the exit from either the G1 or the G2 phase. An explanation is sought for the inhibition of maize root growth and gravireaction brought about by treatment with 3,5-diiodo-4-hydroxybenzoic acid (DIHB). The effects of DIHB and 2,3,5-triiodobenzoic acid (TIBA) on the uptake and efflux of [3H]-indol-3yl-acetic acid (IAA) were tested using segments prepared from the elongation zone (2 to 7 mm region) of maize (Zea mays L. cv. LG11) roots. The uptake of [3H]-IAA (21 nM) by root segments incubated in buffered solutions (pH 5.0) was measured over a 5-min time-course. No significant effect of DIHB at 100 μM was observed, whereas TIBA at 10 μM slightly stimulated the uptake of [3H]-IAA. This experiment was repeated with the addition of non-radioactive IAA (total IAA concentration 1.0 μM). Up to 3 min DIHB (100 μM) had no significant effect, but thereafter a slight stimulation of IAA net uptake was observed. Treatment with TIBA (10 μM) stimulated the accumulation of IAA in the segments. The effects of DIHB (10, 50, 100 μM) and TIBA (10 and 50 μM) on the efflux of [3H]-IAA from segments that had been pretreated in [3H]-IAA (22 nM) were then tested. Treatment with DIHB or TIBA at pH 5.0 inhibited IAA efflux; the inhibition by TIBA was more marked than that produced by DIHB. This experiment was repeated using DIHB (10, 50, 100 μM) buffered at pH 6.0, and an inhibition of IAA efflux was again observed. Both DIHB (10 μM) and TIBA (10 μM) inhibited the binding of [3H]-NPA to a 5000–48000 g membrane fraction prepared from whole maize roots. The effects of the two substances were similar: 40% inhibition of specific binding by DIHB and 41% inhibition by TIBA. This indicates that DIHB, like TIBA, binds to the N-1-naphthyl-phthalamic acid-sensitive carrier for IAA efflux. It is concluded that DIHB, like TIBA, inhibits IAA transport at the level of efflux. The similarity between DIHB and TIBA as regards chemical structure and their inhibitory effects on IAA efflux and NPA binding strongly suggest that they act on the same carrier for IAA efflux across the plasmalemma.  相似文献

The uptake of [5-3H]indoleacetic acid (IAA) by Zea mays L. root segments involves nonsaturable and saturable processes. The pH optimum of the saturable component was found to be 5.0. The proton ionophore carbonylcyanide p-trifluoromethoxyphenylhydrazone inhibited at 100 micromolar the saturable component of IAA uptake but had no effect on non-saturable uptake. This indicates that the saturable component of IAA uptake is dependent on the proton gradient across the plasmalemma. The high level of proton extrusion in the elongation zone of the root will stimulate nonsaturable and saturable uptake of IAA in that zone.  相似文献

A brief account is given of the discovery of abscisic acid (ABA) in roots and root caps of higher plants as well as the techniques by which ABA may be demonstrated in these tissues. The remainder of the review is concerned with examining the rôle of ABA in the regulation of root growth. In this regard, it is well established that when ABA is supplied to roots their elongation is usually inhibited, although at low external concentrations a stimulation of growth may also be found. Fewer observations have been directed at exploring the connection between root growth and the level of naturally occurring, endogenous ABA. Nevertheless, the evidence here also suggests that ABA is an inhibitory regulator of root growth. Moreover, ABA appears to be involved in the differential growth that arises in response to a gravitational stimulus. Recent reports that deny a rôle for ABA in root gravitropism are considered inconclusive. The response of roots to osmotic stress and the changes in ABA levels which ensue, are summarised; so are the interrelations between ABA and other hormones, particularly auxin (e.g. indoleacetic acid); both are considered in the context of the root growth and development. Quantitative changes in auxin and ABA levels may together provide the root with a flexible means of regulating its growth.  相似文献

The mechanisms linking fat intake to bone loss remain unclear. By demonstrating the expression of the free fatty acid receptor G-coupled protein receptor 40 (GPR40) in bone cells, we hypothesized that this receptor may play a role in mediating the effects of fatty acids on bone remodeling. Using micro-CT analysis, we showed that GPR40−/− mice exhibit osteoporotic features suggesting a positive role of GPR40 on bone density. In primary cultures of bone marrow, we showed that GW9508, a GRP40 agonist, abolished bone-resorbing cell differentiation. This alteration of the receptor activator of NF-κB ligand (RANKL)-induced osteoclast differentiation occurred via the inhibition of the nuclear factor κB (NF-κB) signaling pathway as demonstrated by decrease in gene reporter activity, inhibitor of κB kinase (IKKα/β) activation, inhibitor of κB (IkBα) phosphorylation, and nuclear factor of activated T cells 1 (NFATc1) expression. The GPR40-dependent effect of GW9508 was confirmed using shRNA interference in osteoclast precursors and GPR40−/− primary cell cultures. In addition, in vivo administration of GW9508 counteracted ovariectomy-induced bone loss in wild-type but not GPR40−/− mice, enlightening the obligatory role of the GPR40 receptor. Then, in a context of growing prevalence of metabolic and age-related bone disorders, our results demonstrate for the first time in translational approaches that GPR40 is a relevant target for the design of new nutritional and therapeutic strategies to counter bone complications.  相似文献

Intraperitoneal administration of polar glycopeptidolipids extracted from Mycobacterium chelonae (GPLp-Mc) has led to reversal of Doxorubicin-induced leucopenia in a manner comparable to that effected by GM-CSF administered in a dose of 100 IU (2.5 micrograms/kg). The mode of action and the toxicity of this product are being studied. Results obtained on the mouse indicate that it would be worthwhile to undertake tests in man aimed at studying the effect of GPLp-Mc on chemotherapy- and radiotherapy-induced leukopenias, once toxicological studies have been carried out.  相似文献

Following different reports on the stoichiometry and configuration of NO binding to mammalian and bacterial reduced cytochrome c oxidase aa3 (CcO), we investigated NO binding and dynamics in the active site of beef heart CcO as a function of NO concentration, using ultrafast transient absorption and EPR spectroscopy. We find that in the physiological range only one NO molecule binds to heme a3, and time-resolved experiments indicate that even transient binding to CuB does not occur. Only at very high (∼ 2 mM) concentrations a second NO is accommodated in the active site, although in a different configuration than previously observed for CcO from Paracoccus denitrificans [E. Pilet, W. Nitschke, F. Rappaport, T. Soulimane, J.-C. Lambry, U. Liebl and M.H. Vos. Biochemistry 43 (2004) 14118-14127], where we proposed that a second NO does bind to CuB. In addition, in the bacterial enzyme two NO molecules can bind already at NO concentrations of ∼ 1 μM. The unexpected differences highlighted in this study may relate to differences in the physiological relevance of the CcO-NO interactions in both species.  相似文献
