Quantitative analyses of indol-3yl-acetic acid (I aa ) in Zea mays L. (cv. LG 11) root segments cultured in vitro were performed by gas chromatography-mass spectrometry with selected ion monitoring. The root extracts were first purified by highperformance liquid chromatography. Root primordia initiation in intact and decapitated roots showed different patterns: decapitation strongly enhanced primordia initiation in their first 10 mm. During the culture (5 days), I aa content decreased in both intact and decapitated roots. No correlation was found between the level of endogenous auxin and the numher of root primordia initiated from either intact or decapitated maize root segments.  相似文献

Changes in mechanical properties and chemical nature of the cell walls of the different zones along elongating maize ( Zea mays L. cv. LG 11) roots were analyzed and the following results were obtained. (1) The apical region 2 to 5 mm from the tip of 15 mm long roots showed rapid elongation whereas the region 8–10 mm from the tip showed very little growth. (2) The minimum stress-relaxation time (To) and the mean stress-relaxation rate (R) of the cell wall were small whereas the maximum stress-relaxation time (Tm) was large in the region where cell elongation was optimum. The To and R increased and the Tm decreased gradually towards the base of the root. (3) The amounts of non-cellulosic polysaccharides of the cell wall were highest in the region 1.5–2.5 mm from the tip, decreasing until 5 mm from the tip, and then increasing towards the base. However, the proportion of this fraction in the total cell wall polysaccharides was highest in the extreme tip (cap and meristem, 0–1 mm) and decreased towards the base. (4) Major neutral sugars constituting the non-cellulosic polysaccharides of the cell wall were xylose, arabinose, galactose and glucose, with minor amounts of rhamnosc. mannose and fucose. The 1–15 mm region was on the whole rich in glucose and xylose and contained arabinose to a lesser extent. However, the chemical nature in the apical region, (0–2 mm, was rather special, being rich in galactose and fucose. (5) The cell wall of maize roots contained, as a whole, only little pectic substances but was high in hemicellulose 1 (rich in xylose, arabinose and glucose) and hemicellulose 2 (rich in glucose and xylose). (6) It appeared that in the elongating region (apical 2 to 5 mm) the cell elongation rate (CET) showed a rather good correlation with the parameters of mechanical properties (To, Tm and R) and with neutral sugar compositions in the non-cellulosic polysaccharides.  相似文献

Apical segments of roots of Zea mays L. cv. Orla and cv. Anjou show a strong georeaction during 7 hours geostimulation. This is abolished by detipping the segments and restored by replacing the tips upon the apical cut surfaces. After exodiffusion of endogenous indoleacetic acid (IAA) the retipped segments showed a significantly lower geocurvature. Application of low concentrations of IAA to the basal cut surface of root segments from which endogenous IAA had not been allowed to exodiffuse increased the geocurvature of retipped Orla segments but decreased geocurvature of Anjou segments. At appropriate concentration basally applied IAA restored the georeaction capacity of root segments from which the endogenous auxin had exodiffused. The implications of the interaction between exogenous and endogenous IAA in the control of root georeaction are discussed with special reference to the normal role of endogenous IAA in the regulation of root georeaction and the variation in endogenous IAA content of roots of different cultivars of maize. The probability that the normal control of root growth and georeaction involves concomitant actions in the elongation zone of IAA moving preferentially in the acropetal direction and basipetally transported growth inhibitors (such as abscisic acid) produced in the cap cells is stressed.  相似文献

Auxin binding onto membrane fractions of primary roots of maize seedlings has been demonstrated using naphth-1yl-acetic acid (NAA) and indol-3yl-acetic acid (IAA) as ligands. This binding is compared with the already well characterized interaction between auxins and coleoptile membranes. The results indicate that while kinetic parameters are of the same order for root and coleoptile binding, a number of differences occur with respect to location in cells and relative affinity. Variations of the Level of the Nitrogen and Thiol Groups in Tissues Culture.—Distribution of nitrogen and thiol compounds was analysed in the fragments of carrot roots cultivated in vitro. A typical gradient of –SH groups was observed: high level in the callus, low level in the base of the fragment (in contact with the medium). In discontinuous light, callus—which have a great amount of chlorophylls—contain less sulfhydric compounds than in continuous dark. With increasing age of the tissues, the concentration of the endogenous –SH groups was decreasing. The probable biochemical relations between –SH level, auxin content and auxin-oxidase activity are discussed.  相似文献

In stem sections of lentil seedlings, there is a typical polar movement of IAA labelled with 14C. The degree of polarity, expressed as the ratio of basipetal to acropetal transport, was (25°C) 7.6. A decrease (from 25° to 15°C) and an increase (from 25° to 30°C) of temperature cause a reduction of the IAA uptake by the sections and a decrease of both the basipetal and the acropetal translocation of IAA. Results suggest that the basipetal as well as the acropetal movement of auxin, are dependent of a metabolical component which is discussed.  相似文献

Under water stress conditions, induced by mannitol solutions (0 to 0.66 M ) applied to the apical 12 mm of intact roots of Zea mays L. (cv. LG 11), a growth inhibition, a decrease in the osmotic potential of the cell sap and a significant accumulation of abscisic acid (ABA) were observed. When the roots were placed in a humid atmosphere after the stress, the growth rate increased again, even if elongation had been totally inhibited. Under a stress corresponding to an osmotic potential of -1.09 MPa in the solution, growth was totally inhibited, which means that the root cell turgor pressure was reduced to the yield threshold. These conditions led to the largest accumulation of ABA. The effect of water stress on the level of ABA was studied for three parts of the root. The greatest increase in ABA (about 10 fold) was obtained in the growth zone and this increase was apparently independent of the hydrolysis of the conjugated form. With a mannitol treatment of 1 h equivalent to a stress level of -1.39 MPa, a 4-fold increase in ABA efflux into the medium was obtained. These results suggest that there are interactions between water stress, root growth, osmotic potential and the ABA level. The growth under conditions of stress and the role of endogenous ABA in the control of plant metabolism, specially in the growth zone, are discussed.  相似文献

The use of stable indole-3yl-acetic acid (IAA) labeled by 6 atoms of 13C allowed, after [13C]IAA treatment, simultaneous gas chromatography-mass spectrometry quantifications of both endogenous [12C]IAA and applied [13C]IAA levels in Zea mays L. roots. Root material was immersed for 1 hour in a buffered (pH 6.0) solution without or with [13C]IAA at 10−7 molar. Both applied and endogenous IAA were thus measured for three zones of the roots (apical, elongating, differentiating) directly after treatment and also 2 hours later. Growth was followed over a 4 hour period. Roots not immersed elongated more than control roots (immersed in buffer), which grew more than IAA-treated roots. Immersion in buffer induced a large decrease (−68%) of [12C]IAA in the apical part of control roots, whereas immersion in [13C]IAA prevented most of it. No significant difference between control and treated roots occurred in the two other zones. Two hours after treatment, [13C]IAA had completely disappeared from the elongating zone even though [12C]IAA level was essentially stable. A direct relationship occurred between the level of IAA in the elongating zone and the growth of the root. This relationship was strongly disturbed if unmetabolized [13C]IAA was present. However, the relationship returned to its initial state when significant amounts of free [13C]IAA were no longer detectable. These results are discussed in terms of the stability of both types of compounds and the utility of the method of using stable isotopes of hormones, for the understanding of hormonal regulation of plant growth.  相似文献

Aims: To investigate the antimicrobial spectrum of Lactococcus piscium CNCM I‐4031 and its protective effect in cooked and peeled shrimp against Brochothrix thermosphacta. Methods and Results: Sixteen pathogenic and spoiling bacteria were inhibited in Elliker, but not in shrimp juice agar plates. In shrimp packed under modified atmosphere and stored at 8°C, B. thermosphacta (103 CFU g?1) was inhibited by 4·1 log CFU g?1 when co‐inoculated with L. piscium (106 CFU g?1). Brochothrix thermosphacta spoiled the product after 11 days, with the emission of strong butter/caramel off‐odours. In co‐culture with L. piscium, sensory shelf‐life was extended by at least 10 days. The inhibition was partially explained by a drop in pH from 6·6 to 5·6. The physicochemical composition of shrimp and shrimp juice was established to identify the inhibition mechanisms involved. Conclusion: Lactococcus piscium CNCM I‐4031 has a wide antimicrobial spectrum. The strain inhibits B. thermosphacta in shrimp and significantly prolongs sensory shelf‐life. Significance and Impact of the Study: Lactococcus piscium CNCM I‐4031 is shown to be a promising agent for improving shrimp quality and may be tested against pathogens and in other food matrices. Knowledge of the physicochemical composition of shrimp and shrimp juice will allow the development of a chemically defined model medium for determining the inhibition mechanisms involved.  相似文献

Aphanomyces euteiches is a polyphagous, homothallic soilborne pathogen producing asexual (zoospores) and sexual (oospores) spores. Even if oospores are essential for disease development and survival, to date, no study has focused on the production rates of oospores or the quality of the offspring produced by oospores. In this study, a nonabrasive oospore extraction method from infected roots of leguminous species (pea, faba bean and vetch) was developed. This methodology includes steps of grinding and filtration. The quality of oospores (viable, dormant and dead) was assessed with tetrazolium bromide staining, and germination of oospores was tested using exudates of peas, faba bean and vetch. The average yield of the extraction method was approximately 21%. Staining revealed some differences between strains and between leguminous species. The germination percentage of oospores extracted from pea, faba bean and vetch was 25%, 62% and 70%, respectively, and a significant difference was observed according to the origin of A. euteiches‐inoculated strains. Application of exudates seems to stimulate the germination of oospores (2% for the control, 18% for pea exudates and 1% for vetch exudates). Differences observed between A. euteiches strains and leguminous species indicate that more knowledge concerning the biology of oospores is needed. This will help to better estimate evolution process of the pathogen and manage resistance and crop successions.  相似文献
