Short-term weightlessness produced by parabolic flight maneuvers altered gene expression patterns in human endothelial cells

This study focused on the effects of short-term microgravity (22 s) on the gene expression and morphology of endothelial cells (ECs) and evaluated gravisensitive signaling elements. ECs were investigated during four German Space Agency (Deutsches Zentrum für Luft- und Raumfahrt) parabolic flight campaigns. Hoechst 33342 and acridine orange/ethidium bromide staining showed no signs of cell death in ECs after 31 parabolas (P31). Gene array analysis revealed 320 significantly regulated genes after the first parabola (P1) and P31. COL4A5, COL8A1, ITGA6, ITGA10, and ITGB3 mRNAs were down-regulated after P1. EDN1 and TNFRSF12A mRNAs were up-regulated. ADAM19, CARD8, CD40, GSN, PRKCA (all down-regulated after P1), and PRKAA1 (AMPKα1) mRNAs (up-regulated) provide a very early protective mechanism of cell survival induced by 22 s microgravity. The ABL2 gene was significantly up-regulated after P1 and P31, TUBB was slightly induced, but ACTA2 and VIM mRNAs were not changed. β-Tubulin immunofluorescence revealed a cytoplasmic rearrangement. Vibration had no effect. Hypergravity reduced CARD8, NOS3, VASH1, SERPINH1 (all P1), CAV2, ADAM19, TNFRSF12A, CD40, and ITGA6 (P31) mRNAs. These data suggest that microgravity alters the gene expression patterns and the cytoskeleton of ECs very early. Several gravisensitive signaling elements, such as AMPKα1 and integrins, are involved in the reaction of ECs to altered gravity.     

Eleven microsatellite markers in Nasonia,Ashmead 1904 (Hymenoptera; Pteromalidae)

We designed primer sequences for 11 microsatellite markers in the jewel wasp Nasonia vitripennis. Most loci could be cross‐amplified in Nasonia longicornis and Nasonia giraulti, which make them amenable for linkage analysis in hybrid crosses. Eight loci were assigned to specific chromosomes. Additionally, 10 loci showed allelic variation in a Nasonia vitripennis field population. The observed number of alleles in this population ranged from two to seven, with observed heterozygosities from 0.0750 to 0.4750.     

Incidence of Klebsiella species in surface waters and their expression of virulence factors.

To investigate the occurrence of different Klebsiella spp. in aquatic environments, a total of 208 samples of natural surface waters was examined. From half (53%) of these samples, 123 Klebsiella strains were isolated, the most common species being Klebsiella pneumoniae. A comparison of these isolates to a group of 207 clinical K. pneumoniae isolates demonstrated that water isolates of K. pneumoniae, unlike those of K. oxytoca and K. planticola, are as capable as clinical isolates of expressing putative virulence factors such as serum resistance and capsular polysaccharides, pili, and siderophores.     

Hemodynamics of the lower extremity with pneumatic foot compression. Effect on leg position]

External pneumatic compression of the foot is being used more and more to increase the venous blood flow in the lower limbs and thus reduce the risk of postoperative deep venous thrombosis. We have investigated the efficacy of the foot pump pneumatic compression device (A-V Impulse, Novamedix, Andover, England) in 10 healthy subjects and in 10 patients undergoing total hip arthroplasty. The velocity of venous blood flow in the common femoral artery was measured in the horizontal, Trendelenburg (head-down, foot-up) and reverse Trendelenburg (head-up and foot-down) positions using a duplex ultrasound unit (Sonoline Elegra, Siemens, Erlangen, Germany) with a 5 MHz linear array probe. Application of the foot pump produced an increase in venous blood flow velocity in all healthy subjects and in all patients. In healthy subjects the mean increase in the horizontal position was 31.18% (SD = 15.86%), and in the Trendelenburg position 20.72 (SD = 15.69%) (right limb). In the reverse Trendelenburg position, the foot pump produced a mean increase of 94.08% (SD = 55.00%). The difference is statistically significant (p < 0.005). In patients with a total hip arthroplasty, the mean increase in the horizontal position was 28.67% (SD = 9.95%), and in the Trendelenburg position 20.34% (SD = 17.85%) (operated limb). In the reverse Trendelenburg, the foot pump produced a mean increase of 91.55% (SD = 42.38%). The difference is statistically significant (p < 0.005). There was no noteworthy difference in results between the controls and patients. Pneumatic compression devices designed to reduce venous stasis are effective in decreasing the rate of deep venous thrombosis after surgery on the lower limb. The results of the present study show that the efficiency of the foot pump in increasing venous return is improved by adopting the reverse Trendelenburg position. This may increase its thromboprophylactic effect.     

Amiloride does not alter NaCl avoidance in Fischer-344 rats

Fischer-344 (F-344) rats differ from other common rat strains in that they fail to show any preference for NaCl at any concentration in two- bottle preference tests. Because 100 microM amiloride partially blocks the NaCl-evoked chorda tympani (CT) response in electrophysiological studies, we tested NaCl preference (0.068-0.273 M) in F-344 rats with and without 100 microM amiloride solution as the solvent. A third group was tested with unadulterated NaCl solutions following CT transection. Amiloride had no significant effect on the NaCl preference-aversion function, whereas CT transection significantly reduced NaCl avoidance. These results suggest that the amiloride-sensitive component of the NaCl response is not necessary for F-344 rats to display avoidance of NaCl, but the entire CT input is.      

Trends in migrant and ethnic minority voting in Australia: findings from the Australian election study

A review of the research literature on migrant voting in Australia shows that the “ethnic vote” has almost disappeared now that migrants tend to vote in a similar way to the rest of the population according to traditional class cleavages. In addition, it is argued that migrants in Australia predominantly reside in safe Labor seats that are represented by cabinet or shadow cabinet ministers. As such their group-based interests are often neglected by the major parties. Using findings from the ABS census and the 1993–2013 AES, this article re-examines whether there is a migrant vote, and if so, the extent to which migrant voting patterns have changed since the 1990s when the migrant and ethnic vote reached its peak. This study reveals patterns of migrant voting among certain birthplace subgroups that are more volatile than in previous decades but at the same time distinctive.
