Background
Chronic Lymphocytic Leukemia (CLL) pathogenesis has been linked to the prolonged survival and/or apoptotic resistance of leukemic B cells in vivo, and is thought to be due to enhanced survival signaling responses to environmental factors that protect CLL cells from spontaneous and chemotherapy-induced death. Although normally associated with cell migration, the chemokine, CXCL12, is one of the factors known to support the survival of CLL cells. Thus, the signaling pathways activated by CXCL12 and its receptor, CXCR4, were investigated as components of these pathways and may represent targets that if inhibited, could render resistant CLL cells more susceptible to chemotherapy.Methodology/Principal Findings
To determine the downstream signaling targets that contribute to the survival effects of CXCL12 in CLL, we took a phosphoproteomics approach to identify and compare phosphopeptides in unstimulated and CXCL12-stimulated primary CLL cells. While some of the survival pathways activated by CXCL12 in CLL are known, including Akt and ERK1/2, this approach enabled the identification of additional signaling targets and novel phosphoproteins that could have implications in CLL disease and therapy. In addition to the phosphoproteomics results, we provide evidence from western blot validation that the tumor suppressor, programmed cell death factor 4 (PDCD4), is a previously unidentified phosphorylation target of CXCL12 signaling in all CLL cells probed. Additionally, heat shock protein 27 (HSP27), which mediates anti-apoptotic signaling and has previously been linked to chemotherapeutic resistance, was detected in a subset (∼25%) of CLL patients cells examined.Conclusions/Significance
Since PDCD4 and HSP27 have previously been associated with cancer and regulation of cell growth and apoptosis, these proteins may have novel implications in CLL cell survival and represent potential therapeutic targets. PDCD4 also represents a previously unknown signaling target of chemokine receptors; therefore, these observations increase our understanding of alternative pathways to migration that may be activated or inhibited by chemokines in the context of cancer cell survival. 相似文献952.

Background
In many countries, the reported pertussis has increased despite high vaccination coverage. However, accurate determination of the burden of disease is hampered by reporting artifacts. The infection frequency is more reliably estimated on the basis of the prevalence of high IgG concentrations against pertussis toxin (IgG-Ptx). We determined whether the increase in reported pertussis in the last decade is associated with an increase in the number of infections.Methodology/Principal Findings
In a cross-sectional population-based serosurveillance study conducted in 2006-07, from a randomly selected age-stratified sample of 7,903 persons, serum IgG-Ptx concentrations were analyzed using a fluorescent bead-based multiplex immuno assay. In 2006-07, 9.3% (95%CI 8.5-10.1) of the population above 9 years of age had an IgG-Ptx concentration above 62.5 EU/ml (suggestive for pertussis infection in the past year), which was more than double compared to 1995-96 (4.0%; 95%CI 3.3-4.7). The reported incidence showed a similar increase as the seroprevalence between both periods.Conclusions
Although changes in the vaccination program have reduced pertussis morbidity in childhood, they have not affected the increased infection rate in adolescent and adult pertussis. Background
Due to increasing numbers of people with diabetes taking part in extreme sports (e.g., high-altitude trekking), reliable handheld blood glucose meters (BGMs) are necessary. Accurate blood glucose measurement under extreme conditions is paramount for safe recreation at altitude. Prior studies reported bias in blood glucose measurements using different BGMs at high altitude. We hypothesized that glucose-oxidase based BGMs are more influenced by the lower atmospheric oxygen pressure at altitude than glucose dehydrogenase based BGMs.Methodology/Principal Findings
Glucose measurements at simulated altitude of nine BGMs (six glucose dehydrogenase and three glucose oxidase BGMs) were compared to glucose measurement on a similar BGM at sea level and to a laboratory glucose reference method. Venous blood samples of four different glucose levels were used. Moreover, two glucose oxidase and two glucose dehydrogenase based BGMs were evaluated at different altitudes on Mount Kilimanjaro. Accuracy criteria were set at a bias <15% from reference glucose (when >6.5 mmol/L) and <1 mmol/L from reference glucose (when <6.5 mmol/L). No significant difference was observed between measurements at simulated altitude and sea level for either glucose oxidase based BGMs or glucose dehydrogenase based BGMs as a group phenomenon. Two GDH based BGMs did not meet set performance criteria. Most BGMs are generally overestimating true glucose concentration at high altitude.Conclusion
At simulated high altitude all tested BGMs, including glucose oxidase based BGMs, did not show influence of low atmospheric oxygen pressure. All BGMs, except for two GDH based BGMs, performed within predefined criteria. At true high altitude one GDH based BGM had best precision and accuracy. 相似文献954.

Novel S-aryltriazole acyclonucleosides were designed as structural analogs based on the previously identified antiviral aryltriazole acyclonucleosides in our laboratories. These S-aryltriazole nucleosides were synthesized in excellent yields via SNAr-mediated S-arylation, followed by subsequent ammonolysis. X-ray structural analysis revealed special structural feature brought by the S-linkage, which may represent an unfavorable factor contributing to the lack of anti-HCV activity for this family of triazole nucleosides. 相似文献

Aim A comparison of biodiversity patterns within Malesia in relation to surface area. Location Analysis of the patterns in species richness and endemism of vascular plants in the five major Malesian islands, i.e. Java, Sulawesi, Sumatra, Borneo and New Guinea. Methods Available data on species richness and species ranges in correlation with the surface area of the respective islands were examined in this work. Estimations of total species numbers for these islands are presented based on extrapolation of all available published Flora Malesiana information and recent checklists, all in all comprising 12,000 different species. The regression analysis of overall species richness and endemism were studied for all species together as well as for different plant families to compare the fit with the Arrhenius species–area model. Results The five islands form a series of independent areas of increasing size suited for an analysis of the species–area relationships at the regional scale. All species taken together and those of families with even distribution throughout Malesia show significant species–area relationships. Non‐significant relationships were detected in families with western or eastern‐centred Malesian distribution patterns. Relationships between number of endemic species and surface area are significant for all species and for the majority of the families with significant species–area relationships. Main conclusions Species–area relationships of families appear to be dependent on species number. Families with high numbers of species usually have a significant species–area relationship whereas small families have not. For the families that display an eastern or western Malesian centred pattern, a historical biogeographical explanation should be invoked. Island surface area appears to be a predictor for island percent endemism in Malesian vascular plants. None of the islands appears to be a hotspot of endemism nor of species diversity, as no significant departure from the Arrhenius model was noted for any of them. 相似文献

Key to mitochondrial activities is the maintenance of mitochondrial morphology, specifically cristae structures formed by the invagination of the inner membrane that are enriched in proteins of the electron transport chain. In Saccharomyces cerevisiae , these cristae folds are a result of the membrane fusion activities of Mgm1p and the membrane‐bending properties of adenosine triphosphate (ATP) synthase oligomerization. An additional protein linked to mitochondrial morphology is Pcp1p, a serine protease responsible for the proteolytic processing of Mgm1p. Here, we have used hydroxylamine‐based random mutagenesis to identify amino acids important for Pcp1p peptidase activity. Using this approach we have isolated five single amino acid mutants that exhibit respiratory growth defects that correlate with loss of mitochondrial genome stability. Reduced Pcp1p protease activity was confirmed by immunoblotting with the accumulation of improperly processed Mgm1p. Ultra‐structural analysis of mitochondrial morphology in these mutants found a varying degree of defects in cristae organization. However, not all of the mutants presented with decreased ATP synthase complex assembly as determined by blue native polyacrylamide gel electrophoresis. Together, these data suggest that there is a threshold level of processed Mgm1p required to maintain ATP synthase super‐complex assembly and mitochondrial cristae organization. 相似文献

The African cyprinid genus Labeobarbus is characterized by a high polymorphism in mouth morphology. The association between four species of this genus (two chiselmouths, one rubberlip with a well-developed fleshy mental lobe on the lower jaw, i.e., a fleshy appendage on the chin, and one usually with a poorly developed one) and 26 environmental variables was investigated in four affluents of the Luhoho River in the Albertine Highlands of the Upper Congo basin. A combination of univariate and multivariate analyses suggested that differences exist in habitat composition at localities where different Labeobarbus species were reported, and within different mouth phenotypes. The association of L. longidorsalis with the presence of cobbles and pebbles and of L. brauni with the presence of boulders suggests that the occurrence of chiselmouths phenotype is associated with stony habitats, whereas phenotypes with a mental lobe, i.e., Labeobarbus longifilis and L. paucisquamatus, mostly occupy habitats without stones. Differences in the distribution between these two species appear to be linked to physico-chemical parameters such as electrical conductivity and water temperature rather than substrate type. The findings of this study are relevant for developing guidelines that aim effective local preservation and conservation of these Labeobarbus species. 相似文献
