Reproductive rate is a key demographic parameter of life history and population ecology. In traditional population-ecology studies of small mammals, this and other vital rates are inferred from capture-mark-recapture (CMR) data. However, CMR assumes that immigrants at first capture can be distinguished from unmarked locally born offspring, an assumption not always met. We verified CMR estimates of locally born red squirrel (Sciurus vulgaris) offspring as a measure of reproductive rate, with candidate offspring (CO)–candidate parent (mothers, CPs) assignment by CERVUS, using ten DNA microsatellite loci. Seventy-two of 122 candidate offspring (59%) were assigned to 52 of 125 CPs in six populations. Estimates of mean litter size were 1.5 young (range 1–3). The 50 CO (41%) not assigned to a reproducing female in the study site were considered immigrants. Parentage assignment also provided evidence of dispersal between two of our sites. Overall, CMR and CERVUS agreed in 77% of cases. Considering only the 55 juveniles determined as locally born by CMR, 50 (91%) were also assigned as local offspring with CERVUS. The main discrepancy between the two methods was that 22 subadult squirrels classified immigrants by CMR, were assigned by CERVUS to females which had reproduced in our sites. It is concluded that although in our study system agreement between CMR and CERVUS in determining local offspring was high, using genetic parentage assignment helped to correctly classify some subadults, considered immigrants by CMR, as locally born. Hence, in large-scale demographic studies, combining CMR with parentage assignment will allow more precise estimates of reproduction and dispersal.  相似文献

Abstract: Four different γ-(glutamyl5)amine derivatives of substance P (SP) were synthesized in vitro in the presence of purified guinea pig liver transglutaminase and Ca2+. The 1,3-diaminopropane, spermidine, spermine (Spm), and monodansylcadaverine adducts of the neuropeptide were purified by HPLC on a reversed-phase column and characterized by fast atom bombardment mass spectrometry. The γ-(glutamyl5)Spm derivative of SP (Spm-SP) was found to be able, like the parent neuropeptide, to provoke rabbit aorta relaxation, to decrease rat arterial blood pressure, and to inhibit collagen-induced platelet aggregation. Unlike SP, only a weak inflammatory response was observed when Spm-SP was injected in the rat hind limb. All these effects were found to be prevented by N ω-nitro- l -arginine methyl ester, a well-known nitric oxide synthesis inhibitor. In contrast, Spm-SP was completely ineffective in contracting guinea pig ileal segments, thus confirming our preliminary observations indicating that Spm-SP does not evoke SP-like spasmogenic effects on isolated smooth muscle preparations. The specificity of the effects due to the selective introduction of a Spm moiety at the glutamine5 level was demonstrated by the SP agonist pharmacological profile of the other γ-(glutamyl5)amine derivatives tested. These results suggest that neurokinin receptors could be differentiated by their capacity to respond to Spm-SP.  相似文献

Isolated Langendorff-perfused rat hearts after 10 minutes preperfusion, were subjected to a substrate-free anoxic perfusion (20 minutes) followed by 20 minutes reperfusion with a glucose-containing oxygen-balanced medium. Under the same perfusion conditions, the effect of exogenous 5mM fructose-1, 6-bisphosphate has been investigated. The xanthine dehydrogenase to xanthine oxidase ratio, concentrations of high-energy phosphates and the TBA-reactive material (TBARS) were determined at the end of each perfusion period in both control and fructose-1, 6-bisphosphate-treated hearts. Results indicate that anoxia induces the irreversible transformation of xanthine dehydrogenase into oxidase as a consequence of the sharp decrease of the myocardial energy metabolism. This finding is supported by the protective effect exerted by exogenous fructose-1, 6-bisphosphate which is able to maintain the correct xanthine dehydrogenase/oxidase ratio by preventing the depletion of phosphorylated compounds during anoxia. Moreover, in control hearts, the release oflactate dehydrogenase during reperfusion, is paralleled by a 50% increase in the concentration of tissue TBARS. On the contrary, in fructose-1, 6-bisphosphate-treated hearts this concentration does not significantly change after reoxygenation, while a slight but significant increase of lactate dehydrogenase activity in the perfusates is observed.

On the whole these data indicate a direct contribution of oxygen-derived free radicals to the worsening of post-anoxic hearts. A hypothesis on the mechanism of action of fructose-1, 6-bisphosphate in anoxic and reperfused rat heart and its possible application in the clinical therapy of myocardial infarction are presented.  相似文献
