Developing taste buds in the anterior mandibular floor of perihatching
chicks were studied by high voltage electron microscopic autoradiography in
order to identify proliferating gemmal cell types. Montaged profiles of 29
taste buds in five cases euthanized between embryonic day 21 and
posthatching day 2 were analyzed after a single [3H]thymidine injection
administered on embryonic day 16, 17 or 18. Results showed that dark cells
comprised 55% of identified (n = 900 cells) and 62% of labeled (n = 568
cells) gemmal cells as compared with light, intermediate, basal or
perigemmal bud cells. Dark cells had both a greater (P < 0.05) number of
labeled cells and a greater amount of label (grains/nucleus) than the other
four bud cell types, irrespective of injection day. The nuclear area
(micron 2) of dark cells was not significantly larger (P > 0.05) than
that of the other gemmal cell types and therefore cannot account for the
greater amount for label in the dark cells. Interestingly, only dark cells
showed a positive correlation (P < 0.003) between amount of label and
nuclear area. Results suggest that, during the perihatching period of
robust cell proliferation, dividing dark cells may give rise primarily, but
not exclusively, to dark cell progeny.

Drosophila oocytes develop together with 15 sister germline nurse cells (NCs), which pass products to the oocyte through intercellular bridges. The NCs are completely eliminated during stages 12–14, but we discovered that at stage 10B, two specific NCs fuse with the oocyte and extrude their nuclei through a channel that opens in the anterior face of the oocyte. These nuclei extinguish in the ooplasm, leaving 2 enucleated and 13 nucleated NCs. At stage 11, the cell boundaries of the oocyte are mostly restored. Oocytes in egg chambers that fail to eliminate NC nuclei at stage 10B develop with abnormal morphology. These findings show that stage 10B NCs are distinguished by position and identity, and that NC elimination proceeds in two stages: first at stage 10B and later at stages 12–14. 相似文献

The fate of benthic bacterial biomass in benthic food webs is a topic of major importance but poorly described. This paper describes an alternative method for evaluation of bacterial grazing rate by meiofauna and macrofauna using bacteria pre-enriched with stable isotopes. Natural bacteria from the sediment of an intertidal mudflat were cultured in a liquid medium enriched with 15NH4Cl. Cultured bacteria contained 2.9% of 15N and were enriched sufficiently to be used as tracers during grazing experiments. Cultured bacteria presented a biovolume (0.21 μm3) and a percentage of actively respiring bacteria (10%) similar to those found in natural communities. The number of Operational Taxon Units found in cultures fluctuated between 56 and 75% of that found in natural sediment. Despite this change in community composition, the bacterial consortium used for grazing experiments exhibited characteristics of size, activity and diversity more representative of the natural community than usually noticed in many other grazing studies. The bacterial ingestion rates of three different grazers were in the range of literature values resulting from other methods: 1149 ngC ind− 1h− 1 for the mud snail Hydrobia ulvae, 0.027 ngC ind− 1 h− 1 for the nematode community, and 0.067 ngC ind− 1 h− 1 for the foraminifera Ammonia tepida. Wolbachia are widespread endosymbionts that affect arthropod reproduction and fitness. Mostly maternally inherited, Wolbachia are occasionally transferred horizontally. Previously, two Wolbachia strains were reported at low prevalence and titres across seven Australian tephritid species, possibly indicative of frequent horizontal transfer. Here, we performed whole-genome sequencing of field-caught Wolbachia-positive flies. Unexpectedly, we found complete mitogenomes of an endoparasitic strepsipteran, Dipterophagus daci, suggesting that Wolbachia in the flies are linked to concealed parasitization. We performed the first genetic characterization of D. daci and detected D. daci in Wolbachia-positive flies not visibly parasitized, and most but not all Wolbachia-negative flies were D. daci-negative, presumably reflecting polymorphism for the Wolbachia infections in D. daci. We dissected D. daci from stylopized flies and confirmed that Wolbachia infects D. daci, but also found Wolbachia in stylopized fly tissues, likely somatic, horizontally transferred, non-heritable infections. Furthermore, no Wolbachia cif and wmk genes were detected and very low mitogenomic variation in D. daci across its distribution. Therefore, Wolbachia may influence host fitness without reproductive manipulation. Our study of 13 tephritid species highlights that concealed early stages of strepsipteran parasitization led to the previous incorrect assignment of Wolbachia co-infections to tephritid species, obscuring ecological studies of this common endosymbiont and its horizontal transmission by parasitoids. 相似文献

We examined the calling behaviour of O. nubilalis virgin females, held at three different constant conditions of relative humidity from the time of emergence through their sixth night of calling, as well as the incidence of mating over time at low or high relative humidity conditions. The mean age at which females initiated calling in their life was independent of relative humidity conditions, although more variable at low humidity. On successive nights of calling, fewer virgin European corn borer females called under low compared with high humidity conditions, and those that did began later in scotophase, had fewer calling bouts, and spent less time calling. These changes in the female prereproductive behaviour were reflected on the incidence of mating which, in the first two days following emergence, was significantly less at low than at high humidity conditions. The ecological significance of these results is discussed in relation with the susceptibility of O. nubilalis to desiccation.
Résumé Nous avons étudié le comportement d'appel des femelles vierges d'Ostrinia nubilalis maintenues sous trois conditions constantes d'humidité relative, depuis l'émergence jusqu'à la sixième nuit d'appel, ainsi que l'incidence d'accouplement d'individus placés en milieu sec ou humide pour diverses périodes de temps. Les conditions d'humidité n'ont pas significativement affecté l'âge moyen auquel les femelles ont commencé à appeler au cours de leur vie, bien qu'il ait été plus variable à 53% h.r. Les nuits subséquentes, le comportement d'appel des femelles maintenues à basse humidité a débuté plus tard dans la nuit, s'est exprimé pour une période de temps plus courte avec moins de séquences d'appel que celui de leurs congénères placés en milieu humide, et la proportion de femelles en appel a toujours été plus faible à 53% h.r. Ces changements du comportement préreproducteur des femelles se sont répercutés sur l'incidence d'accouplement qui, les deux premiers jours suivant lémergence, a été significativement plus faible à basse humidité qu'elle ne l'a été en milieu humide. Ces résultats seront discutés en relation avec l'écologie de O. nubilalis et associés à sa susceptibilité à la dessication.相似文献

The equilibrium interactions between histone H2A-H2B and H3/H4 subunits with 200 base pair chicken erythrocyte DNA have been studied by monitoring the fluorescence polarization of a long-lived fluorescence probe covalently bound to the histone subunits. These studies have brought to light the formation of highly asymmetric complexes exhibiting very high histone/DNA stoichiometries as well as very high apparent affinities. The stoichiometries observed for these non-nucleosome complexes depended both upon the concentration of the histones and the concentration of the DNA 200mer. The observed stoichiometries varied approximately between 4 and 16 histone octamers/DNA 200mer and the affinities were in the nanomolar range. These results are discussed in terms of their in vitro as well as their possible in vivo significance. 相似文献
