Two pectin esterase cDNA clones representing different isozymes with ca. 95% homology were isolated from an early ripening tomato fruit cDNA library. Both clones were longer than previously published sequences, and the encoded proteins possessed extended (229–233 amino acid) putative N-terminal extensions. In addition, the mRNA species corresponding to the two clones showed differential levels of expression in fruit.  相似文献

The G-protein-coupled receptor, CXCR4, is highly expressed on a number of cell types, and together with its ligand, CXCL12, plays an important role in immune development and trafficking of cells. CXCR4 promotes tumor growth, angiogenesis and metastasis, and is a prognostic marker in a number of different types of tumors. Additionally, CXCR4 is utilized, together with CD4, for entry of T-tropic HIV viruses. Ethnic differences in incidence and mortality of various cancers, and in the response to highly active antiretroviral treatment (HAART) of HIV-1 infected individuals have been reported. The aim of this study was to establish if differences in the CXCR4–CXCL12 axis exist between ethnically divergent uninfected South Africans. CXCR4 density was significantly higher on CD4+ and CD8+ T cells, B cells and CD56dim NK cells, and CXCL12 levels lower in Black compared with Caucasian individuals. Furthermore, an inverse correlation was observed between CXCR4 density on CD56+ and CD3+ cells and age, only in Black individuals. CXCL12–3′A heterozygosity (AG) found in 28% of Caucasians did not explain the higher plasma levels of CXCL12 compared to Black individuals who were all GG genotypes, suggesting that other factors influence homeostatic levels of CXCL12. In conclusion, this study demonstrates that ethnically divergent populations show clear differences in both CXCR4 density and CXCL12 plasma levels which may influence the course of cancer and HIV-1 infection.  相似文献

Imprinted genes are expressed predominantly or exclusively from one allele only. This mode of gene expression makes the regulation of imprinted genes susceptible to epigenetic insults, which may in turn lead to disease. There is compelling experimental evidence that certain aspects of assisted reproductive technology (ART) such as in vitro cell culture may have adverse effects on the regulation of epigenetic information in mammalian embryos, including the disruption of imprinted genes and epigenetic regulators. Moreover, in humans, disorders of genomic imprinting have been reported in children conceived by ART. The derivation and in vitro culture of embryonic stem (ES) cells are potential points of origin for epigenetic abnormalities. There is evidence that defects of genomic imprinting occur in mouse embryonic stem cells, with similar data now emerging in related studies in non-human primate and human ES cells. It is therefore pertinent to rigorously assess the epigenetic status of all stem cells and their derivatives prior to their therapeutic use in humans. Focusing on the stability of genomic imprinting, this review discusses the current evidence for epigenetic disruption in mammalian embryonic stem cells in light of the epigenetic disruption observed in ART-derived mammalian embryos.  相似文献

Previous experiments have demonstrated differences in various follicular characteristics between the prolific Chinese Meishan (MS) pig and European Large-White (LW) hybrids and the present experiment was designed to compare the cAMP response to LH by granulosa and theca cells in vitro between the two breeds. Ovaries were recovered from MS (n = 7) and LW hybrid (n = 8) gilts on the day before predicted oestrus and the 12 largest follicles dissected. Quadruplicate aliquots of granulosa cells or minced theca tissue were incubated for 1 h in the presence of 0, 0.1, 1.0, 10, 100 or 1000 ng/ml LH and cAMP production measured. Follicles from MS gilts were smaller (5.9 vs. 7.7 mm; P < 0.001), contained less fluid (81.5 vs. 177.4 microl; P < 0.001), had fewer granulosa cells (3.8 vs. 5.3 x 10(6); P < 0.01) and less theca tissue (30.3 vs. 50.5 mg; P < 0.05) than those from LW hybrid animals. Mean follicular fluid oestradiol concentration was > or = 149 ng/ml in all animals and tended to be higher in the MS follicles (P = 0.07). LH stimulated cAMP production by granulosa and theca cells from both breeds (P < 0.001). Although there was no overall breed effect for the granulosa cells, there was a significant (P < 0.001) interaction between LH dose and breed in the granulosa cells, whether cAMP production was expressed per 10(6) cells or per follicle. In the theca incubations, cAMP production by MS tissue was higher (P < 0.01) when results were expressed per mg tissue and again there was an interaction (P < 0.001) between LH dose and breed whether cAMP production was expressed per mg tissue or per follicle. For both tissue types, MS follicles produced more cAMP at the higher LH doses. In conclusion, this study has shown that MS granulosa and theca tissue respond differently to increasing doses of LH in terms of cAMP production in vitro compared to LW hybrid tissue and this supports previous suggestions of enhanced maturity of MS follicles in the late follicular phase.  相似文献

Background

Heterogeneity in B-type natriuretic peptide (BNP) levels, especially among individuals with acute heart failure with normal left ventricular ejection fraction (HFNEF), can cause confusion in interpreting results. We investigated the characteristics of cases of acute HFNEF with only modestly elevated BNP.

Methods

One hundred forty-two patients with acute or acute exacerbation of chronic HFNEF were divided into two groups by BNP level: BNP < 100 pg/ml (NB group, n = 45) and BNP ≥ 100 pg/ml (B group, n = 97). We compared clinical findings, echocardiography results, and neurohormonal factors between these two groups.

Results

In the NB group, a history of open-heart surgery (OHS) was more frequent (71% vs. 22%, p < 0.0001) and hypertension was less frequent (p = 0.0005). Left atrial diameter (LAd) was higher (p = 0.0026), while interventricular septal thickness, posterior wall thickness, relative wall thickness, left ventricular mass index were lower (p = 0.0005, p = 0.0225, p = 0.0114, p = 0.0051, respectively) in the NB group. In patients with HFNEF, a history of OHS remained an independent predictor of BNP level (< 100 pg/ml) after adjustment for hypertension, age, LAd, and interventricular septal thickness (odds ratio 3.6, p = 0.0252).

Conclusion

We found associations between acute HFNEF with less elevated BNP and a history of OHS. In a patient suspected of HFNEF, a history of OHS is considered diagnostic evidence of presence of diastolic heart failure when plasma levels of BNP are less elevated.  相似文献

Diaz-Perez, S. V., Crouch, V. W., and Orbach, M. J. 1996. Construction and characterization of a Magnaporthe grisea bacterial artificial chromosome library. Fungal Genet. Biol. 20, 280-288. A bacterial artificial chromosome (BAC) library of Magnaporthe grisea containing 4128 clones with an average insert size of 66-kb has been constructed. This library represents seven genome equivalents of M. grisea and has been demonstrated to be representative of the genome by screening for the presence of several single-copy genes and DNA markers. The utility of the library for use in map-based cloning projects was shown by the spanning of a nine-cosmid, 207-kb DNA contig with only 3 BAC clones. In addition, using a lys1-3 auxotroph, we have shown that BAC clones at least 113 kb can be transformed into M. grisea to screen for complementation of mutations. Thus, BACs isolated in chromosome walks can be rapidly screened for the presence of the sought after gene. The ease of construction of BAC libraries and of isolation and manipulation of BAC clones makes the BAC system an ideal one for physical analyses of fungal genomes.  相似文献

Assisted reproduction technologies for clinical and research purposes rely on a brief in vitro embryo culture which, despite decades of progress, remain suboptimal in comparison to the physiological environment. One promising tool to improve this technique is the development of bespoke microfluidic chambers. Here we present and validate a new microfluidic device in polydimethylsiloxane (PDMS) for the culture of early mouse embryos. Device material and design resulted embryo compatible and elicit minimal stress. Blastocyst formation, hatching, attachment and outgrowth formation on fibronectin-coated devices were similar to traditional microdrop methods. Total blastocyst cell number and allocation to the trophectoderm and inner cell mass lineages were unaffected. The devices were designed for culture of 10–12 embryos. Development rates, mitochondrial polarization and metabolic turnover of key energy substrates glucose, pyruvate and lactate were consistent with groups of 10 embryos in microdrop controls. Increasing group size to 40 embryos per device was associated with increased variation in development rates and altered metabolism. Device culture did not perturb blastocyst gene expression but did elicit changes in embryo metabolome, which can be ascribed to substrate leaching from PDMS and warrant further investigation.  相似文献
