The postembryonic antennal development and life cycle of a member of the insect order Mantophasmatodea (Lobatophasma redelinghuysense) was investigated using a series of annulus counts and a time sequence of head capsule measurements. The life cycle comprised six instars. Females achieved significantly larger head capsules from instar 2 onwards, resulting in adult females having a larger mean head capsule diameter (2.58 mm) than males (2.27 mm). Antennae of first instar larvae comprised a smooth four-segmented basiflagellum and a seven-segmented, sensilla-rich distiflagellum. Lengthening of the basiflagellum was achieved by the addition of two annuli per moult, generated by division of the basal annulus (meriston). Annulus number and the unique annulation pattern of the distiflagellum remained constant until adulthood. The segmentation pattern of adult antennae (comprising a basiflagellum and a distiflagellum of 14 and seven annuli respectively) and mode of antennal elongation was consistent for all 11 species examined. Subdivisions in basiflagellar annuli were observed in adults of all species examined, although they are not considered to be true annular divisions. The structure of the mantophasmatodean antenna appears to be autapomorphic within Insecta, bearing little resemblance to that of Grylloblattodea, Dictyoptera or Phasmatodea, all putative sister groups of the Mantophasmatodea. However, the mode of flagellar elongation most closely resembles that of Isoptera, Blattaria and Dermaptera. 相似文献

During the past years, major advances have been made in understanding the sequential events involved in neural plate patterning. Positional information is already conferred to cells of the neural plate at the time of its induction in the ectoderm. The interplay between the BMP- and the Fgf- signaling pathways leads to the induction of neural cell fates. Thus, neural induction and neural plate patterning are overlapping processes. Later, at the end of gastrulation, positional cell identities within the neural plate are refined and maintained by the action of several neural plate organizers. By locally emitting signaling molecules, they influence the fate of the developing nervous system with high regional specificity. Recent advances have been made both in understanding the mechanisms that dictate the relative position of these organizers and in how signaling molecules spread from them with high spatial and temporal resolution. 相似文献

The goal of an AIDS vaccine regimen designed to induce cellular immune responses should be to reduce the viral set point and preserve memory CD4 lymphocytes. Here we investigated whether vaccine-induced cellular immunity in the absence of any Env-specific antibodies can control viral replication following multiple low-dose challenges with the highly pathogenic SIVmac239 isolate. Eight Mamu-A*01-positive Indian rhesus macaques were vaccinated with simian immunodeficiency virus (SIV) gag, tat, rev, and nef using a DNA prime-adenovirus boost strategy. Peak viremia (P = 0.007) and the chronic phase set point (P = 0.0192) were significantly decreased in the vaccinated cohort, out to 1 year postinfection. Loss of CD4(+) memory populations was also ameliorated in vaccinated animals. Interestingly, only one of the eight vaccinees developed Env-specific neutralizing antibodies after infection. The control observed was significantly improved over that observed in animals vaccinated with SIV gag only. Vaccine-induced cellular immune responses can, therefore, exert a measure of control over replication of the AIDS virus in the complete absence of neutralizing antibody and give us hope that a vaccine designed to induce cellular immune responses might control viral replication. 相似文献

A mouse genomic clone containing a lactate dehydrogenase-A (LDH-A)
processed pseudogene and a B1 repetitive element was isolated, and a
nucleotide sequence of approximately 3 kb was determined. The pseudogene
and B1 element are flanked by perfect 13-bp repeats, and the B1 sequence
starts at 14 nucleotides 3' to the presumptive polyadenylation signal of
the pseudogene. The nucleotide sequences of the LDH-A genes and processed
pseudogenes from mouse, rat, and human were compared, and a phylogenetic
tree was constructed. The rate and pattern of nucleotide substitutions in
the LDH-A pseudogenes are similar to previously reported results (Li et al.
1984). The average rate of nucleotide substitutions in the LDH-A
pseudogenes is 4.3 X 10(- 9)/site/year. The substitutions of C----T and
G----A are most frequent, and A----G substitutions are relatively high. The
rate of synonymous substitutions in the LDH-A genes is 5.3 X 10(-9), which
is not significantly higher than the average rate of 4.7 X 10(-9) for 35
mammalian genes. The rate of nonsynonymous substitutions in the LDH-A genes
is 0.20 X 10(-9), which is considerably lower than the average rate of 0.88
X 10(-9) for 35 mammalian genes. Thus, the mammalian LDH-A gene appears to
be highly conserved in evolution.

Summary Examination of eroded and intact earth mounds in the Clanwilliam district, South Africa, indicates that they are well-established active termitaria of the harvester termite Microhodotermes viator. Unoccupied lower portions of the mounds contain ubiquitous trace-fossil evidence of earlier inhabitation by the same species. Previous studies indicating that fossorial molerats played a major role in the formation of the mounds are not supported by the observations presented here. Calcretization of the basal parts of the earth mounds has been caused by groundwater interaction with the more alkaline mound soil. 14C dating of this calcrete indicates that the earth mounds have been in existence for at least 4000 years, an order of magnitude greater than any previously recorded longevity for termitarium inhabitation. 相似文献

Summary The P type conjugative plasmid RP4 has been shown to be able to promote the transfer of the Agrobacterium Ti-plasmid. The results provide additional evidence that agrocin 84 sensitivity, exclusion of phage AP1, ability to catabolize the guanidine derivatives octopine and nopaline and tumor inducing ability, are Ti-plasmid determined properties. Furthermore, the results strongly support the notion that at least part of the Ti-plasmid is transferred from the bacterium to the target plant cells, since it was demonstrated that Ti-plasmid linked genes specify the synthesis of octopine or nopaline by crown-gall tumor cells. 相似文献

Herceptin failure is a major clinical problem in breast cancer. A subset of breast cancer patients with high HER-2/neu levels eventually experience metastatic disease progression when treated with Herceptin as a single agent. Mechanistic details of development of this aggressive disease are not clear. Therefore, there is a dire need to better understand the mechanisms by which drug resistance develops and to design new combined treatments that benefit patients with aggressive breast cancer and have minimal toxicity. We hypothesized that 3, 3′-diindolylmethane (DIM), a non-toxic agent can be combined with Herceptin to treat breast cancers with high levels of HER-2/neu. Here, we evaluated the effects of Herceptin alone and in combination with DIM on cell viability, apoptosis and clonogenic assays in SKBR3 (HER-2/neu-expressing) and MDA-MB-468 (HER-2/neu negative) breast cancer cells. We found that DIM could enhance the effectiveness of Herceptin by significantly reducing cell viability, which was associated with apoptosis-induction and significant inhibition of colony formation, compared with single agent treatment. These results were consistent with the down-regulation of Akt and NF-kB p65. Mechanistic investigations revealed a significant upregulation of miR-200 and reduction of FoxM1 expression in DIM and Herceptin-treated breast cancer cells. We, therefore, transfected cells with pre-miR-200 or silenced FoxM1 in these cells for understanding the molecular mechanism involved. These results provide experimental evidence, for the first time, that DIM plus Herceptin therapy could be translated to the clinic as a therapeutic modality to improve treatment outcome of patients with breast cancer, particularly for the patients whose tumors express high levels of HER-2/neu. 相似文献
