An unidentified spirochete, referred to as the 277F agent, was isolated from Haemaphysalis leporispalustris ticks from two cottontail rabbits by inoculation of the tick suspension into embryonated chicken eggs. Because of its minute width (0.1 mu), the organism was difficult to see when stained by the Giemsa method, but was readily demonstrated by silver impregnation or fluorescent-antibody procedures. In dark-field microscopy, the spirochetes appeared uniformly and rather tightly coiled, and exhibited typical corkscrewlike motility. After yolk-sac inoculation, the agent was highly lethal for chick embryos and was recovered in large quantity from several embryonic tissues and fluids. It could also be maintained in nonfertile eggs and in an enriched liquid medium. This previously undescribed spirochete could pass through Berkefeld N but not Seitz EK filters. It was relatively resistant to heat and to penicillin or sulfadiazine, but was markedly inhibited by streptomycin, broad-spectrum antibiotics, and homologous neutralizing antiserum. Of several species of animals tested for susceptibility to this spirochete, only the snowshoe hare gave evidence of infection.  相似文献

Two outdoor shiitake (Lentinula edodes) cultivation experiments, established in Missouri USA in 1999 and 2000, produced mushrooms in 2000–2005. We examined shiitake production in response to substrate species, inoculum form, inoculum strain, and inoculation timing, using total mushroom weight per log as the primary response variable with log characteristics as covariates. The significantly greater mushroom weight produced by sugar maple logs compared with white or northern red oak was attributable to the higher proportion of undiscolored wood volume in the maple logs, rather than to bark thickness or log diameter. The “wide temperature range” shiitake strain produced significantly greater yield compared with the “warm” or “cold” weather strains. Both the wide-range and warm-weather strains were stimulated to fruit by significant rain events, while the cold-weather strain was responsive to temperature. Inoculation with sawdust spawn gave significantly greater yield than colonized wooden dowels or pre-packaged “thimble” plug inoculum. The second and third full years following inoculation were the most productive.  相似文献

棕色固氮菌(Azotobacter vinelandii Lipmann)突变种UW3能在无Mo的无氨培养基中固氮生长,低浓度的Mn对UW3突变种生长有促进作用,从在Mn中生长的UW3菌体中分离得到的部分纯固氮酶组分Ⅰ蛋白含量有Mn和Fe原子(Fe/Mo/Mn为10.41:0.19:1.00)并有OP MoFe蛋白一半的还原乙炔和质子的活性。这种蛋白的吸收光谱和圆二色谱与MoFe蛋白存在明显的差异,含Mn蛋白的亚基分子量都与MoFe蛋白的α亚基相近。初步结果表明,这种含Mn蛋白可胡是一种固氮酶组分Ⅰ蛋白。  相似文献

在合适的结晶条件下 ,从含Cr无氨培养基中生长的固氮菌 (AzotobactervinelandiiLipmann)突变种UW3 中纯化出的CrFe蛋白可从溶液中析出深棕色斜四棱柱晶体 ,晶体最大的两条对角线长度分别可达 0 .2 5mm和 0 .12mm。PEG 80 0 0、MgCl2 、NaCl、Tris和Hepes缓冲液的浓度及结晶方法等对该蛋白的出晶率、晶核数目、晶体大小和质量都有明显影响。CrFe蛋白结晶所需的上述化合物的最适浓度与在Mn中生长的固氮菌突变种UW3 的MnFe蛋白和缺失nifZ固氮菌突变种的ΔnifZMoFe蛋白结晶所需的最适浓度有所不同。结果表明 ,该蛋白晶体可能为CrFe蛋白的晶体  相似文献

Extracts of B. subtilis contain somatostatin-like immunoactivity (1-20 pg per g wet weight cells). Two major forms were detected, one with reactivity in both N- and C-terminal immunoassays similar to somatostatin-28 and a second form reactive only in the C-terminal specific immunoassay similar to somatostatin-14. Both forms were active in a bioassay and the bioactivity was neutralized in the presence of antibody to the central, biologically active part of somatostatin-14. Preconditioned medium contained no detectable somatostatin whereas conditioned medium had 80-380 pg per liter.  相似文献

With the aim of developing high-affinity mono and multivalent antagonists of cholera toxin (CT) and Escherichia coli heat-labile enterotoxin (LT) we are using the galactose portion of the natural receptor ganglioside GM1 as an anchoring fragment in structure-based inhibitor design efforts. In order to establish a better structure-activity relationship for guiding these studies, we designed and prepared a small focused library of twenty 3,5-substituted phenylgalactosides based on two previous leads. The compounds were tested for their ability to block CTB(5) binding to immobilized ganglioside receptor and compared to the two previous leads. The crystal structures of the most promising compounds bound to either CTB(5) or LTB(5) were then determined in order to understand the basis for affinity differences. The most potent new compound yielded a six-fold improvement over our benchmark lead m-nitrophenyl-alpha-d-galactopyranoside (MNPG), and a two-fold improvement in IC(50) over a newer MNPG derivative. These results support the notion that the m-nitrophenyl moiety of MNPG and its derivatives is an important element to retain in future optimization efforts. Additionally, a consensus binding-pocket for the alkylmorpholine or piperazine moiety present in all of the designed antagonists was established as an important area of the GM1 binding site to target in future work.  相似文献
