We hypothesized that reactive oxygen species (ROS) may be involved in the pathogenesis of silicosis. To investigate ROS' dependent pathophysiological processes during silicosis we studied the kinetic clearance of instilled stable nitroxide radicals (TEMPO). Antioxidant enzymes' superoxide dismutase (SOD) and glutathione peroxidase (GPx), and lipid peroxidation were also studied in whole lungs of rats exposed to crystalline silica (quartz) and sham exposed controls. Low frequency L-band electron spin resonance spectroscopy was used to measure the clearance of TEMPO in whole-rat lungs directly. The clearance of TEMPO followed first order kinetics showing significant differences in the rate for clearance between the diseased and sham exposed control lungs. Comparison of TEMPO clearance rates in the sham exposed controls and silicotic rats showed an oxidative stress in the rats exposed to quartz. Studies on the antioxidant enzymes SOD and GPx in the lungs of silicotic and sham exposed animals supported the oxidative stress and accelerated clearance of TEMPO by up regulated levels of enzymes in quartz exposed animals. Increased lipid peroxidation potential in the silicotics also supported a role for enhanced generation of ROS in the pathogenesis of silica-induced lung injury. These in vivo experiments directly demonstrate, for the first time, that silicotic lungs are in a state of oxidative stress and that increased generation of ROS is associated with enhanced levels of oxidative enzymes and lipid peroxidation. This technique offers great promise for the elucidation of ROS induced lung injury and development of therapeutic strategies for the prevention of damage.  相似文献

The bauxite mine at Cornet near Oradea in northwestern Romania produced thousands of bones in an excavation in 1978, mainly from ornithopod dinosaurs and rarer pterosaurs. Bird specimens reported previously from this fauna are equivocal. The fossils are disarticulated bones in good condition which occur highly concentrated in lenses within bauxite clays, which are dated as Berriasian (earliest Cretaceous). The bauxite represents detrital material washed into deep fissures and caves formed within a karst of uplifted Tithonian (latest Jurassic) marine limestones. The bones are generally uniform in size and shape, and they are abraded, evidence for considerable transport and for winnowing of the deposit. The area was one of several islands on the northern shore of Tethys, and it was inundated by the sea later in the Early Cretaceous. There is evidence for insular adaptations in the dinosaur faunas. The ornithopod dinosaurs may include several taxa, but they are smaller on average than an assemblage of typical Wealden ornithopods, perhaps because of dwarfing on the island. In addition, sauropods are absent and theropods are barely represented in the fauna. The fauna is geographically significant since it shows relationships with western Europe and with Asia.  相似文献

An Arabidopsis thaliana cDNA encoding a new RNA-binding protein (RBP37) was cloned from a silique cDNA library. The predicted amino acid sequence corresponds to a RBP containing two RNA recognition motifs (RRM) and a basic domain. An affinity for nucleic acids was confirmed in binding assays using in vitro synthesised AtRBP37 protein. In situ hybridisation experiments on sections of flowers and siliques showed expression only in growing organs: gynoecium, petals, filaments and during early-embryogenesis expression is located in the embryo proper and the suspensor up to late heart stage. Expression is not detected in the embryo during maturation.This results suggests an expression pattern correlated with dividing cells.  相似文献

Since 1991, Envirovet Summer Institutes have provided an intensive educational program in wildlife and ecosystem health to approximately 225 animal health professionals. Envirovet provides knowledge, skills, and multiple mentors to catalyze ecosystem health-oriented careers for young veterinarians. It is a total immersion experience, with the students engaged in 60–70 hours a week of instruction, 6–7 days a week, for 6 weeks. The course is comprised of lecture, laboratory, and field experiences organized into three sessions: 1) terrestrial wildlife and ecosystem health; 2) aquatic wildlife and ecosystem health; and 3) an ecosystem health approach to international development. Sessions 1 and 2 take place in Florida and Georgia; Session 3 takes place in a developing country (e.g., Kenya, Brazil, South Africa). Ultimately, the goal of Envirovet is to increase the numbers and effectiveness of veterinarians in productive ecosystem health research and application teams around the world.  相似文献

Matrilins are putative adaptor proteins of the extracellular matrix (ECM) which can form both collagen-dependent and collagen-independent filamentous networks. While all known matrilins (matrilin-1, -2, -3, and -4) are expressed in cartilage, only matrilin-2 and matrilin-4 are abundant in non-skeletal tissues. To clarify the biological role of matrilin-2, we have developed a matrilin-2-deficient mouse strain. Matrilin-2 null mice show no gross abnormalities during embryonic or adult development, are fertile, and have a normal lifespan. Histological and ultrastructural analyses indicate apparently normal structure of all organs and tissues where matrilin-2 is expressed. Although matrilin-2 co-localizes with matrilin-4 in many tissues, Northern hybridization, semiquantitative RT-PCR, immunohistochemistry and biochemical analysis reveal no significant alteration in the steady-state level of matrilin-4 expression in homozygous mutant mice. Immunostaining of wild-type and mutant skin samples indicate no detectable differences in the expression and deposition of matrilin-2 binding partners including collagen I, laminin-nidogen complexes, fibrillin-2 and fibronectin. In addition, electron microscopy reveals an intact basement membrane at the epidermal-dermal junction and normal organization of the dermal collagen fibrils in mutant skin. These data suggest that either matrilin-2 and matrilin-2-mediated matrix-matrix interactions are dispensable for proper ECM assembly and function, or that they are efficiently compensated by other matrix components including wild-type levels of matrilin-4.  相似文献

Himatanthus sucuuba is a tree species that occurs in Central Amazonian white water floodplains and in non-flooded uplands. The objective of this paper was to verify whether flooding causes differences in germination percentages, seedling mortality as well as alterations of leaf number and biomass increments, related to the habitat of origin of the seeds (várzea floodplain versus terra firme upland, in the following V and TF, respectively). To test this effect, seeds from populations of both habitats were collected, and germination percentages were calculated under different experimental conditions. One hundred seeds collected in three sites of the floodplain and one hundred seeds from three sites of the upland were placed in trays for germination in a greenhouse in the Amazon Research Institute (INPA) in Manaus, Brazil. Seedlings with a height of 7 cm were then subjected to a period of 120 days of flooding in tanks with three treatments: control, waterlogging (root system and the base of the stem), and complete submersion. Ground water from the INPA was used in the tanks and was changed at weekly intervals. Germination percentages were high in most treatments. Seedling mortality was significantly higher in seedlings originating from upland, reaching 100% in submerged seedlings. On the other hand, várzea-originated seedlings showed no mortality under waterlogging or submergence.

Height growth of waterlogged seedlings from floodplains exceeded that of the control. Submerged seedlings did not grow, independently of the origin, and those originating from uplands died after about 100 days. Number of leaves and leaf area were significantly lower under partial submersion than in the control, independently of the origin of the seeds. Under total submersion abscission of all leaves was verified 30 days after inundation for seedlings of both origins. In the V population, all seedlings survived the waterlogged period, whereas in the TF population, 30% of the seedlings died when subjected to waterlogging. With complete submergence, in the V population 30% of the seedlings had died after 120 days, in the TF population after 90 days mortality was 100%. The results show that germination, seedling growth and seedling survival in H. sucuuba differ among populations of floodplain and upland sites: V plants performed better than TF plants under waterlogging and under submersion. The results strongly indicate that ecotypic differentiation exists between these two populations of H. sucuuba individuals in the two habitat types, which may be enforced by a strong selective pressure due to the regular occurring flood pulse.  相似文献

Question: Is the diverse mosaic of forest/grassland (Campos) vegetation on the hills in the Porto Alegre region natural or of anthropogenic origin? What are the best approaches to management and conservation of forest/grassland mosaics in southern Brazil? Location: 280 m a.s.l., Rio Grande do Sul State (30°04′32″S; 51°06′05″W, southern Brazil. Methods: A 50-cm long radiocarbon dated sediment core from a swamp on Morro Santana was analysed for pollen and charcoal, and multivariate data analysis was used to reconstruct past vegetation and fire dynamics. Results: The formation of swamp deposits is related to a change to wetter climatic conditions since 1230 cal yr BP. The diverse forest/grassland mosaic existed already at that time and can be seen as natural in origin as it has been also shown from other studies in southern Brazil. Since 580 cal yr BP, forests expanded continuously. The marked higher occurrence of the pioneer Myrsine during the last 70 years, indicates a change in the disturbance regime. In the past, vegetation has been influenced by mostly anthropogenic fire, set first by Amerindians and later by European settlers. Conclusions: Management for conservation of forest/grassland mosaics should take into account, first, that grasslands are remnants of earlier drier Holocene periods and not a result of deforestation and, second, the history of disturbance by grazing and fire. Suppression of grazing and burning has likely resulted in a trend towards more woody vegetation under modern wet climatic conditions. If management for conservation excludes fire, the present grassland patches will tend to disappear due to forest expansion under the modern humid climate. Maintaining or reintroducing cattle grazing in conservation areas could be an alternative to fire.  相似文献

(Dibenzoylmethanato)boron difluoride (DBMBF2) was found to form with benzene (B) and its methylated derivatives (MB) in cyclohexane (cHex) two types of ground state complexes. The first complexes with low stoichiometries 2 : 1, 1 : 1 and 1 : 2 do not fluoresce when they are excited. On the other hand, the ground state complexes with high stoichiometry, DBMBF2-(B)(n) or DBMBF2-(MB)(n) (with n> 2), exhibit a strong fluorescence in their excited states. These findings differ from the previous results, where the strongly fluorescing complexes have been argued to be the 1 : 1 and 1 : 2 exciplexes, complexes of the singlet excited state of DBMBF2 with one or two B or MB molecules. These differences are discussed in terms of the solute-solute and solute-solvent interactions when DBMBF2 and MB are solutes in cyclohexane or when MB is a co-solvent of cyclohexane in binary mixtures of DBMBF2. We also argue that the use of well-suited analytical methods is important for the determination of the nature of the various complexes. Furthermore, to understand the nature of the interactions between benzene and DBMBF2 molecules, we attempt to predict the sites of interaction between DBMBF2 and benzene molecules by determining theoretically the structure of the 1 : 1 complex.  相似文献

We have analyzed 61 German breast and breast/ovarian cancer families for BRCA1 mutations using single-strand conformation polymorphism analysis (SSCP) followed by sequencing. Forty-seven of the families had at least three cases (at least two under 60 years) and 14 families had only two cases of breast/ovarian cancer (at least one under 50 years). Twenty-eight families were breast/ovarian and 33 were breast cancer-only families. Eighteen mutations in BRCA1 were detected in 11/28 breast/ovarian cancer families and 7/33 breast cancer families and none in the families with only two cases. We identified 17 truncation mutations (8 frameshift, 7 nonsense and 2 splice variants) and one missense mutation. Seven of these are novel and two, the 5382insC and 5622C→T mutations, occurred in two apparently unrelated families. The genotype of the two families with the 5382insC mutation is compatible with the rare haplotype segregating with the 5382insC mutation in different populations, further supporting its European origin. One unclassified missense alteration, R841W, was found in one family but did not segregate with the disease, suggesting that it is more likely a polymorphism. We also report and discuss the sequence of several new unclassified single-nucleotide changes first identified by SSCP. Of the 18 mutations, 13 occurred in the 3′ third of the gene (end of exon 11–24) and ovarian cancers were found in eight of these families. Received: 5 February 1998 / Accepted: 7 April 1998  相似文献

A novel complex mutation consisting of a small deletion/insertion (3958del5ins4) was found in the breast cancer-1 gene (BRCA-1) in three unrelated French breast and/or ovarian cancer families. These mutations occurred at the same nucleotide position of the 3′ end of exon 11. The wild-type sequence, CTCAG, was deleted and replaced by AGGC in the three families. The consequence is the generation of a stop codon, TAG, resulting in a truncated protein. We propose two different mechanisms to explain the generation of this complex mutation: (i) the simultaneous occurrence of a deletion and an insertion in a stem-loop structure and (ii) the abortive integration of a human transposable element (Tigger 1) that deleted 5 nucleotides and inserted a 4-nucleotide “scar”, corresponding to the 5′ extremity of the transposon.
