p27Xic1, a Cdk inhibitor, promotes the determination of glial cells in Xenopus retina 总被引:4,自引:0,他引:4
p27Xic1, a member of the Cip/Kip family of Cdk inhibitors, besides its known function of inhibiting cell division, induces Müller glia from retinoblasts. This novel gliogenic function of p27Xic1 is mediated by part of the N-terminal domain near but distinct from the region that inhibits cyclin-dependent kinases. Cotransfections with dominant-negative and constitutively active Delta and Notch constructs indicate that the gliogenic effects of p27Xic1 work within the context of an active Notch pathway. The gradual increase of p27Xic1 in the developing retina thus not only limits the number of retinal cells but also increasingly favors the fate of the last cell type to be born in the retina, the Müller glia. 相似文献

Nod1 and Nod2 are cytosolic proteins involved in intracellular recognition of microbes and their products. Recently, it was shown that these proteins recognize different moieties of bacterial peptidoglycan (PGN) mediating non-specific pathogen resistance and possibly generating signals for the adaptive immune response. The laminar arrays of distinct cell types in the vertebrate retina are built by a histogenic process in which cell fate is correlated with birth order. To explore this co-ordination mechanistically, we altered the relative timing of cell cycle exit in the developing Xenopus retina and asked whether this affected the activity of neural determinants. We found that Xath5, a bHLH proneural gene that promotes retinal ganglion cell (RGC) fate, ( Kanekar, S., Perron, M., Dorsky, R., Harris, W. A., Jan, L. Y., Jan, Y. N. and Vetter, M. L. (1997) Neuron 19, 981-994), does not cause these cells to be born prematurely. To drive cells out of the cell cycle early, therefore, we misexpressed the cyclin kinase inhibitor, p27Xic1. We found that early cell cycle exit potentiates the ability of Xath5 to promote RGC fate. Conversely, the cell cycle activator, cyclin E1, which inhibits cell cycle exit, biases Xath5-expressing cells toward later neuronal fates. We found that Notch activation in this system caused cells to exit the cell cycle prematurely, and when it is misexpressed with Xath5, it also potentiates the induction of RGCs. The potentiation is counteracted by co-expression of cyclin E1. These results suggest a model of histogenesis in which the activity of factors that promote early cell cycle exit enhances the activity of factors that promote early cellular fates. 相似文献

One of the diseases of veterinary and public health importance affecting the Kafue lechwe (Kobus leche kafuensis) on the Kafue flats is brucellosis, for which only scant information is available. During the 2003 (October), 2004 (December), and 2008 (July-December) hunting seasons in the Kafue flats, we conducted a study to determine the seroprevalence of Brucella spp. in the Kafue lechwe and to evaluate serologic tests for detection of Brucella spp. antibodies in lechwe. The Rose Bengal Test (RBT), competitive enzyme-linked immunosorbent assay (cELISA), and fluorescence polarization assay (FPA) were used. A total of 121 Kafue lechwe were hunted for disease investigations in 2003, 2004, and 2008 in the Kafue Flat Game Management Area. Of these, 21.6%, (95% confidence interval [CI]: 14.2-29.1%) had detectable antibodies to Brucella spp. The Kafue lechwe in Lochnivar National Park had higher antibody results than those in Blue Lagoon National Park (odds ratio=3.0; 95% CI: 0.94-9.4). Infection levels were similar in females (21.6%) and males (21.7%). Results were similar among RBT, FPA, cELISA tests, suggesting that these could effectively be used in diagnosing brucellosis in the Kafue lechwe. Our study demonstrates the presence of Brucella infections in the Kafue lechwe in two national parks located in the Kafue flats and further highlights the suitability of serologic assays for testing the Kafue lechwe. Because the Kafue lechwe is the most hunted wildlife species in Zambia, hunters need to be informed of the public health risk of Brucella spp. infection. 相似文献

One commonly studied driver of community assembly is the effect of dominant species on subordinate species. Dominant species may impact community assembly during competitive sorting, or recruitment. For ants, important and abundant species in the tropics, several factors may drive community assembly including competition, dispersal, priority effects, and environmental conditions. Although competition is a hallmark of ant ecology, few have examined the influence of patchily distributed dominant ants on other ant species and diversity, especially at the recruitment stage. Here, I consider the impacts of a canopy dominant ant species, Azteca instabilis, and changes in vegetation on twig‐nesting ant colony founding and ant community assembly in a coffee agroecosystem. I added artificial nests to coffee plants in areas with and without A. instabilis four times over a year, and then examined the occupation rate and identity of species colonizing nests. I also examined vegetation characteristics of sites where nests were added. The presence of A. instabilis on coffee plants drastically lowered colonization rates, but nest occupation increased with tree density, and with decreasing proportion of Inga spp. trees in the canopy. The presence of A. instabilis limited the number of nests occupied by six of the ten most common species; most rare species, however, were not affected by A. instabilis presence. Richness of colonizing ants in areas with A. instabilis was lower, but these effects did not significantly affect richness across broader scales. Despite large effects on individual species, species composition did not differ greatly in areas with and without A. instabilis, but some vegetation characteristics (basal area and tree richness) were predictive of ant composition. These results suggest that A. instabilis strongly affects founding events especially for common twig‐nesting species and that both vegetation and influences from this dominant species affect community assembly of twig‐nesting ants at the local scale. 相似文献
