Over-expression of oleosin-fused IGF1 results in the formation of insoluble aggregates in Escherichia coli occupying 35% of total proteins. In this study, a method based on artificial oil body (AOB) was applied to obtain active IGF1, insulin-like growth factor 1, from its insoluble form in one step. The stability of AOB emulsions constituted with soybean oleosin was maximized in the optimal composition of TAG (97.04%, wt/wt), PL (1.14%, wt/wt), and oleosin-UbIGF1 (1.82%, wt/wt) at pH 7.5 and at 25°C. Upon sonication, the mixture comprising plant oil and the insoluble fusion protein was readily assembled into AOBs. After peptide cleavage mediated by endopeptidase, the IGF1 free of fusion tags was liberated and then recovered. Subsequently, IGF1 self-refolded on AOB was obtained with high yield of 63.2 mg/g dry cell. This on-AOB refolding can be applied to the development of bacterial expression and purification of other active recombinant proteins.  相似文献

The rejection of eggs of brood parasites in several species of hosts is based on cues only at the blunt pole (BP) and not at the sharp pole (SP) of the foreign and own eggshell. We investigated whether intraclutch variation is confined to a specific egg pole in species where the extent of intraclutch variation in the overall egg appearance is known to positively covary with either egg rejection rate or the probability of being parasitized. For the two poles separately, we analysed intraclutch variation of eggshell brightness and blue chroma. We quantified intraclutch variation as the standard deviations of these colour metrics, instead of their coefficients of variation which would represent a statistically flawed approach. Pooling measurements of brightness across the whole egg surface led to statistically non‐significant results and masked positive correlations of BP brightness with egg rejection or parasitism risk, respectively. In contrast, patterns of blue chroma were important across the whole egg. Thus, the traditional whole egg ‘averaging’ approach may mask biologically important effects of intraclutch variation when the variation and potential signalling functions of egg appearance are confined to a specific egg part (brightness). However, analyses based on only BP and SP eggshell region specific data may also lack the power to detect effects of phenotypic traits that do not vary between egg poles (blue chroma). We advocate the use of a combination of region‐specific and whole‐eggshell based colour metrics and manipulations in cognitive, perceptual, and ecological studies of foreign egg rejection.  相似文献

Background

The exceptional diversity of coloration found in avian eggshells has long fascinated biologists and inspired a broad range of adaptive hypotheses to explain its evolution. Three main impediments to understanding the variability of eggshell appearance are: (1) the reliable quantification of the variation in eggshell colours; (2) its perception by birds themselves, and (3) its relation to avian phylogeny. Here we use an extensive museum collection to address these problems directly, and to test how diversity in eggshell coloration is distributed among different phylogenetic levels of the class Aves.

Methodology and Results

Spectrophotometric data on eggshell coloration were collected from a taxonomically representative sample of 251 bird species to determine the change in reflectance across different wavelengths and the taxonomic level where the variation resides. As many hypotheses for the evolution of eggshell coloration assume that egg colours provide a communication signal for an avian receiver, we also modelled reflectance spectra of shell coloration for the avian visual system. We found that a majority of species have eggs with similar background colour (long wavelengths) but that striking differences are just as likely to occur between congeners as between members of different families. The region of greatest variability in eggshell colour among closely related species coincided with the medium-wavelength sensitive region around 500 nm.

Conclusions

The majority of bird species share similar background eggshell colours, while the greatest variability among species aligns with differences along a red-brown to blue axis that most likely corresponds with variation in the presence and concentration of two tetrapyrrole pigments responsible for eggshell coloration. Additionally, our results confirm previous findings of temporal changes in museum collections, and this will be of particular concern for studies testing intraspecific hypotheses relating temporal patterns to adaptation of eggshell colour. We suggest that future studies investigating the phylogenetic association between the composition and concentration of eggshell pigments, and between the evolutionary drivers and functional impacts of eggshell colour variability will be most rewarding.  相似文献

Background

Chicks of virulent brood parasitic birds eliminate their nestmates and avoid costly competition for foster parental care. Yet, efforts to evict nest contents by the blind and naked common cuckoo Cuculus canorus hatchling are counterintuitive as both adult parasites and large older cuckoo chicks appear to be better suited to tossing the eggs and young of the foster parents.

Methodology/Principal Findings

Here we show experimentally that egg tossing imposed a recoverable growth cost of mass gain in common cuckoo chicks during the nestling period in nests of great reed warbler Acrocephalus arundinaceus hosts. Growth rates of skeletal traits and morphological variables involved in the solicitation of foster parental care remained similar between evictor and non-evictor chicks throughout development. We also detected no increase in predation rates for evicting nests, suggesting that egg tossing behavior by common cuckoo hatchlings does not increase the conspicuousness of nests.

Conclusion

The temporary growth cost of egg eviction by common cuckoo hatchlings is the result of constraints imposed by rejecter host adults and competitive nestmates on the timing and mechanism of parasite virulence.  相似文献

Aim  We argue that 'propagule pressure', a key term in invasion biology, has been attributed at least three distinct definitions (with usage of a related term causing additional confusion). All of the definitions refer to fundamental concepts within the invasion process, with the result that the distinct importance of these different concepts has been at best diluted, and at worst lost.
Location  Global.
Methods  We reviewed pertinent literature on propagule pressure to resolve confusion about different uses of the term 'propagule pressure' and we introduced a new term for one variant, colonization pressure. We conducted a computer simulation whereby the introduction of species is represented as a simple sampling process to elucidate the relationship between propagule and colonization pressure.
Results  We defined colonization pressure as the number of species introduced or released to a single location, some of which will go on to establish a self-sustaining population and some of which will not. We subsequently argued that colonization pressure should serve as a null hypothesis for understanding temporal or spatial differences in exotic species richness, as the more species that are introduced, the more we should expect to establish. Finally, using a simple simulation, we showed that propagule pressure is related to colonization pressure, but in a non-linear manner.
Main conclusion  We suggest that the nature of the relationship between propagule pressure and colonization pressure, as well as the efficacy of various proxy measures of each, require more detailed exploration if invasion ecology is to continue to develop into a more predictive science.  相似文献

Presenilin 1 (PS1) in its active heterodimeric form is the catalytic center of the gamma-secretase complex, an enzymatic activity that cleaves amyloid precursor protein (APP) to produce amyloid beta (Abeta). Ubiquilin 1 is a recently described PS1 interacting protein, the overexpression of which increases PS1 holoprotein levels and leads to reduced levels of functionally active PS1 heterodimer. In addition, it has been suggested that splice variants of the UBQLN1 gene are associated with an increased risk of developing Alzheimer disease (AD). However, it is still unclear whether PS1 and ubiquilin 1 interact when expressed at endogenous levels under normal physiological conditions. Here, we employ three novel fluorescence resonance energy transfer-based techniques to investigate the interaction between PS1 and ubiquilin 1 in intact cells. We consistently find that the ubiquilin 1 N terminus is in close proximity to several epitopes on PS1. We show that ubiquilin 1 interacts both with PS1 holoprotein and heterodimer and that the interaction between PS1 and ubiquilin 1 takes place near the cell surface. Furthermore, we show that the PS1-ubiquilin 1 interaction can be detected between endogenous proteins in primary neurons in vitro as well as in brain tissue of healthy controls and Alzheimer disease patients, providing evidence of its physiological relevance.  相似文献

We assessed the prevalence of alien species as a driver of recent extinctions in five major taxa (plants, amphibians, reptiles, birds and mammals), using data from the IUCN Red List. Our results show that alien species are the second most common threat associated with species that have gone completely extinct from these taxa since AD 1500. Aliens are the most common threat associated with extinctions in three of the five taxa analysed, and for vertebrate extinctions overall.  相似文献

Most theoretical models of coevolution between brood parasites, whether interspecific or conspecific, and their hosts explicitly assume consistent individual behaviour in host egg‐rejection responses. Accordingly, hosts cast as acceptors always accept, whereas ejectors always reject parasitic eggs when exposed to stable ecological conditions. To date, only few studies have attempted to test this critical assumption of individual repeatability in egg‐rejection responses of hosts. Here, we studied the repeatability of egg rejection in blackbirds (Turdus merula) and song thrush (T. philomelos), species in which females are reported to reject simulated, non‐mimetic foreign eggs at intermediate frequencies at the population level. However, intermediate rates of acceptance and rejection can be consistent with either or both intra‐ and interindividual variability in rejection behaviours. Our experiments revealed generally high individual consistency in these hosts’ responses to experimentally introduced non‐mimetic and mimetic model foreign eggs. Individuals also responded faster on average to second than to first trials within the same breeding attempts, but the difference was statistically significant only in blackbirds. These results are consistent with the critical assumption of co‐evolutionary models, that statistically egg rejection is mostly individually repeatable, but also reveal that some individuals in both species change their responses even within the short time‐window of one breeding attempt. The data suggest that individuals reject foreign eggs faster when perceived parasitism risk is greater because of repeated introductions of experimental parasitic eggs. We provide methodological recommendations to facilitate experimental and meta‐analytical studies of individual egg rejection repeatability and discuss how to reduce technical constraints arising from disparate treatments and variable sample sizes for future studies.  相似文献

Current research into the evolution and adaptive function of avian eggshell pigmentation, including maculation, has focused mostly on signalling‐based and structural function hypotheses but ignored the potential consequences of shell pigmentation for the developing avian embryo, especially in moderating the embryo's interaction with its light environment. The exposure of the eggs to sunlight that frequently accompanies avian incubation behaviour is one of the major evolutionary steps setting apart birds and reptiles, and coincides with the appearance of eggshell pigmentation. This suggests that shell pigments could play a major role in ensuring the successful development of the avian embryo. We propose that the effects of shell pigments on the egg contents should be considered in addition to established hypotheses of shell pigmentation such as crypsis, egg recognition or a possible structural function. This approach has the potential to identify trade‐offs between different pigment functions and to resolve some of the long standing paradoxa in the evolution of eggshell colour, such as the occurrence of conspicuous blue eggs in passerines or the secondary evolution of white eggshells in cavity nesters. In particular, we identify seven hypotheses, which address how the interaction of eggshell pigments and the light environment may influence embryonic development. These hypotheses are the: thermo‐regulation; UV‐B protection; photo‐acceleration; lateralization; circadian rhythm; photo‐reactivation; and antimicrobial defence. We believe that the understanding of eggshell pigmentation will greatly benefit from taking these hypotheses into consideration when studying the functional significance of eggshell pigmentation and suggest a number of promising directions for future experimental and comparative research.  相似文献

The number of alien species transported as stowaways is steadily increasing and new approaches are urgently needed to tackle this emerging invasion pathway. We introduce a general framework for identifying high‐risk transport pathways and receiving sites for alien species that are unintentionally transported via goods and services. This approach combines the probability of species arrival at transport hubs with the likelihood that the environment in the new region can sustain populations of that species. We illustrate our approach using a case study of the Asian black‐spined toad Duttaphrynus melanostictus in Australia, a species that is of significant biosecurity concern in Australasia, Indonesia, and Madagascar. A correlative model fitted to occurrence data from the native geographic range of D. melanostictus predicted high environmental suitability at locations where the species has established alien populations globally. Applying the model to Australia revealed that transport hubs with the highest numbers of border interceptions and on‐shore detections of D. melanostictus were environmentally similar to locations within the species’ native range. Numbers of D. melanostictus interceptions and detections in Australia increased over time, but were unrelated to indices of air and maritime trade volume. Instead, numbers of interceptions and detections were determined by the country of origin of airplanes (Thailand) and ships (Indonesia). Thus, the common assumption that transport pressure is correlated with invasion risk does not hold in all cases. Our work builds on previous efforts to integrate transport pressure data and species distribution models, by jointly modelling the number of intercepted and detected stowaways, while incorporating imperfect detection and the environmental suitability of receiving hubs. The approach presented here can be applied to any system for which historical biosecurity data are available, and provides an efficient means to allocate quarantine and surveillance efforts to reduce the probability of alien species establishment.  相似文献
