Chloride intracellular channel proteins (CLICs) are distinct from most ion channels in that they have both soluble and integral membrane forms. CLICs are highly conserved in chordates, with six vertebrate paralogues. CLIC-like proteins are found in other metazoans. CLICs form channels in artificial bilayers in a process favoured by oxidising conditions and low pH. They are structurally plastic, with CLIC1 adopting two distinct soluble conformations. Phylogenetic and structural data indicate that CLICs are likely to have enzymatic function. The physiological role of CLICs appears to be maintenance of intracellular membranes, which is associated with tubulogenesis but may involve other substructures.  相似文献

This is a scale-down study of a 500-m3 methane recovery test plant for anaerobic treatment of palm oil mill effluent (POME) where biomass washout has become one of the problems because of the continuous mixing of effluent during anaerobic treatment of POME. Therefore, in this study, anaerobic POME treatment using a scaled down 50-l bioreactor which mimicked the 500-m3 bioreactor was carried out to improve biogas production with and without biomass sedimentation. Three sets of experiments were conducted under different conditions in terms of biomass sedimentation applied to the system. The first experiment was operated under semi-continuous mode whereas the second and third experiments were operated based on mix and settle mode. As expected, biomass retention improved the anaerobic process as the POME treatment incorporated with mix and settle system were able to operate at an organic loading rate (OLR) of 3.5 and 6.0 kg COD/m3/day respectively, while the semi-continuous operated anaerobic treatment only achieved OLR of 3.0 kg COD/m3/day. The highest biogas and methane production rates achieved were 2.42 m3/m3 of reactor/day and 0.992 m3/m3 of reactor/day, respectively at OLR 6.0 kg COD/m3/day. The biomass or solids retention in the reactors was represented by the total solids measured in this study.  相似文献

SecA, the ATPase of Sec translocase, mediates the post-translational translocation of preprotein through the protein-conducting channel SecYEG in the bacterial inner membrane. Here we report the structures of Escherichia coli Sec intermediates during preprotein translocation as visualized by electron microscopy to probe the oligomeric states of SecA during this process. We found that the translocase holoenzyme is symmetrically assembled by SecA and SecYEG on proteoliposomes, whereas the translocation intermediate 31 (I31) becomes asymmetric because of the presence of preprotein. Moreover, SecA is a dimer in these two translocation complexes. This work also shows surface topological changes in the components of translocation intermediates by immunogold labeling. The channel entry for preprotein translocation was found at the center of the I31 structures. Our results indicate that the presence of preprotein introduces asymmetry into translocation intermediates, while SecA remains dimeric during the translocation process.  相似文献

Pseudomonas aeruginosa is a ubiquitous Gram-negative bacterium which secretes a wide range of hydrolytic enzymes, toxins, and virulence factors into the extracellular medium. Although P. aeruginosa possesses numerous specific systems for the export of proteins across its double-membrane envelopes, the Sec system is still the major and essential mechanism. However, very little is known about its molecular basis. We constructed, cloned, and expressed the N-terminal 236 amino acids of PaSecA domain (PaSecAN236), and SecAL43P mutants of P. aeruginosa in Escherichia coli BL21.19 (secA(ts)). Here, we describe the purification of PaSecAN236 by using osmotic shock as the first step to efficiently release targeted protein from cells, followed by cation-exchange and size exclusion columns to obtain homogeneous PaSecAN236. The purified PaSecA N-terminal domain was functional in stimulating the ATPase activity of mutant SecAL43P protein of P. aeruginosa.  相似文献

We asked whethercrystalloid administration improves tissue oxygen extraction inendotoxicosis. Four groups of anesthetized pigs(n = 8/group) received either normalsaline infusion or no saline and either endotoxin or no endotoxin. Wemeasured whole body (WB) and gut oxygen delivery and consumption duringhemorrhage to determine the critical oxygen extraction ratio(ERO2 crit). Just after onset of ischemia (critical oxygen delivery rate), gut was removed for determination of area fraction of interstitial edema and capillary hematocrit. Radiolabeled microspheres were used todetermine erythrocyte transit time for the gut. Endotoxin decreased WBERO2 crit(0.82 ± 0.06 to 0.55 ± 0.08, P < 0.05) and gutERO2 crit(0.77 ± 0.07 to 0.52 ± 0.06, P < 0.05). Unexpectedly, saline administration also decreased WBERO2 crit (0.82 ± 0.06 to 0.62 ± 0.08, P < 0.05) and gutERO2 crit (0.77 ± 0.07 to 0.67 ± 0.06, P < 0.05) in nonendotoxin pigs. Saline administration increased thearea fraction of interstitial space (P < 0.05) and resulted in arterial hemodilution(P < 0.05) but not capillaryhemodilution (P > 0.05). Salineincreased the relative dispersion of erythrocyte transit times from0.33 ± 0.08 to 0.72 ± 0.53 (P < 0.05). Thus saline administration impairs tissue oxygen extractionpossibly by increasing interstitial edema or increasing heterogeneityof microvascular erythrocyte transit times.

Checklists of the marine algae of Malaysia have been published. The last checklist included the marine algae of Singapore. The checklist recorded 212 taxa for the region. This paper lists an additional 7 taxa of Chlorophyta, 18 taxa of Rhodophyta and 5 taxa of Phaeophyta, which had been left out of the last checklist. These include marine algae from Pulau Pinang, Pulau Redang and Pulau Sibu. In addition, new records of Cyanophyta (2 species), Chlorophyta (4 species), Rhodophyta (10 species) and Phaeophyta (3 species) are reported.  相似文献

Profound changes in calcium metabolism occur during pregnancy. The mother has to make available extra calcium for fetal requirements while ensuring that her plasma and bone calcium concentrations are satisfactorily maintained. In a cross-sectional study plasma concentrations of the major calcium-regulating hormones--namely, calcitonin, parathyroid hormone, 25-hydroxyvitamin D (25-OHD), and 1,25-dihydroxyvitamin D (1,25-(OH)2D)--were measured to establish their interrelations during normal pregnancy. The major changes observed were increases in the circulating concentrations of 1,25-(OH)2D and calcitonin. Concentrations of parathyroid hormone and 25-OHD remained within the normal range. The increased concentrations of 1,25-(OH)2D enable the increased physiological need for calcium to be met by enhancing intestinal absorption of this element. The simultaneous rise in calcitonin opposes the bone-resorbing activities of 1,25-(OH)2D, thereby protecting the integrity of the maternal skeleton. Maternal calcium homeostasis is thus maintained yet the requirements of the fetus are fulfilled.  相似文献

We previously reported that pyrroline-5-carboxylate (PC), the intermediate in the interconversions of proline, ornithine and glutamate markedly stimulates hexosemonophosphate-pentose pathway activity in human erythrocytes. The stimulation is mediated by pyrroline-5-carboxylate reductase which generates NADP+ accompanying the conversion of pyrroline-5-carboxylate to proline. We now report that the previously demonstrated effect of pyrroline-5-carboxylate on glucose oxidation through the hexose-monophosphate-pentose pathway is accompanied by increased phosphoribosyl-pyrophosphate production and increased formation of nucleotides via the salvage pathway. The demonstrated effect of pyrroline-5-carboxylate on purine processing may provide a regulatory link between amino acid and nucleotide metabolism.  相似文献
