Gene expression in autumn leaves   总被引:36,自引:0,他引:36  
Two cDNA libraries were prepared, one from leaves of a field-grown aspen (Populus tremula) tree, harvested just before any visible sign of leaf senescence in the autumn, and one from young but fully expanded leaves of greenhouse-grown aspen (Populus tremula x tremuloides). Expressed sequence tags (ESTs; 5,128 and 4,841, respectively) were obtained from the two libraries. A semiautomatic method of annotation and functional classification of the ESTs, according to a modified Munich Institute of Protein Sequences classification scheme, was developed, utilizing information from three different databases. The patterns of gene expression in the two libraries were strikingly different. In the autumn leaf library, ESTs encoding metallothionein, early light-inducible proteins, and cysteine proteases were most abundant. Clones encoding other proteases and proteins involved in respiration and breakdown of lipids and pigments, as well as stress-related genes, were also well represented. We identified homologs to many known senescence-associated genes, as well as seven different genes encoding cysteine proteases, two encoding aspartic proteases, five encoding metallothioneins, and 35 additional genes that were up-regulated in autumn leaves. We also indirectly estimated the rate of plastid protein synthesis in the autumn leaves to be less that 10% of that in young leaves.  相似文献

Natural clones and clones from laboratory crossings of Daphnia pulex have been tested in their response to stimuli for male and ephippia formation. The clones are from ponds in Illinois, USA, an area with both obligate and cyclical clones of D. pulex. The progeny from two types of crosses were studied: 1) crosses between an obligate clone producing males and a cyclical clone producing only females, and 2) self-fertilization within a cyclical clone producing both sexes. Both the natural and the artificial clones showed great variation in response to environmental stimuli for males. Ephippia response seems more equal among the clones, but two of the artificial clones could hardly be stimulated to produce ephippia in the induction experiments. In crowded cultures, however, they produced a few. Crossings between cyclicals and obligates yielded mostly cyclical progeny. Progeny from the selfed clone had low survival and fecundity.  相似文献

During storage of moist animal feed, growth of detrimental fungi causing spoilage, or being mycotoxigenic or pathogenic, is a severe problem. Addition of biopreservative yeasts or lactic acid bacteria can significantly reduce this problem. However, their use requires several careful considerations. One is the safety to the animal, humans and the environment, tightly connected to legal aspects and the need for pre-market authorisation when supplementing feed with microorganisms. Although both yeasts and lactic acid bacteria are considered comparatively safe organisms due to low production of toxic metabolites, it is of great importance to understand the mechanisms behind the biopreservative abilities. Another important issue concerns practical aspects, such as the economic production of large amounts of the organisms and the development of a suitable formulation giving the organisms a long shelf life. These aspects are discussed and a recommendation of this review is that both safety and formulation aspects of a specific microbe should be considered at an early stage in the selection of new organisms with biopreservation potential.  相似文献

Terrestrial organisms of the Arctic are faced with strong climatic fluctuations. Predictable seasonality with cold/long winters and short/cool summers are combined with unpredictable between and within year variation. This indicates that various selection pressures act on the reproductive strategies of the populations. The arctic collembolan Hypogastrura tullbergi reproduces in a short period following snow melt. Hatching occurs in late summer, the animals grow to adult size within their second summer and reproduce for the first time in the beginning of their third summer. We performed several experiments to determine the reproductive investment and proximate mechanism that regulate timing and duration of reproduction. We found that H. tullbergi entered a reproductive diapause when reared at constant temperature, a diapause that was terminated by a cold exposure (winter). Surprisingly, cold exposure of small juveniles may also prevent development of a reproductive diapause in adults. Thus, the life-cycle normally spanning 2 yr can potentially be reduced to one year if the overwintering juveniles reach maturity before the end of the reproductive period in the field. After termination of the diapause, the animals reproduced up to 3 times during a period of 6 weeks at 15°C. This reproductive period was considerably longer (measured in degree days) than the one observed in the field. Our results suggested that temperature quiescence, i.e. the inability to reproduce under a certain temperature threshold, may adjust the termination of the reproductive period with current temperature before a new diapause occurs in late summer. The cost of reproduction was low and suggests that it can be adaptive to spread reproduction over more than one year. The results are discussed in relation to the arctic climate and strategies favoured by unpredictable and predictable (seasonal) variations in the environment. The present study forms part of a larger investigation on population dynamics and life history strategies of H. tullbergi from the Arctic.  相似文献

The mutations of the Abruptex locus in Drosophila melanogaster fall into three categories. There are recessive lethal alleles and viable alleles. The latter can be divided into suppressors and nonsuppressors of Notch mutations. The recessive lethals are lethal in heterozygous combination with Notch. As a rule the recessive lethals are lethal also in heterozygous combination with the viable alleles. Heterozygous combinations of certain viable alleles are also lethal. In such heterozygotes, one heteroallele is a suppressor of Notch and the other is a nonsuppressor. Other heterozygous combinations of viable alleles are viable and have an Abruptex phenotype. The insertion of the wild allele of the Abruptex locus as an extra dose (carried by a duplication) into the chromosomal complement of the fly fully restores the viability of the otherwise lethal heterozygotes if two viable alleles are involved. The extra wild allele also restores the viability of heterozygotes in which a lethal and a suppressor allele are present. If, however, a lethal and a nonsuppressor are involved, the wild allele only partly restores the viability, and the effect of the wild allele is weakest if two lethal alleles are involved. It seems likely that of the viable alleles the suppressors of Notch are hypermorphic and the nonsuppressors are hypomorphic. The lethal alleles share properties of both types, and are possibly antimorphic mutations. It is suggested that the locus is responsible for a single function which, however, consists of two components. The hypermorphic mutations are defects of the one component and the hypomorphic mutations of the other. In heterozygotes their cumulative action leads to decreased viability. The lethal alleles are supposed to be defects of the function as a whole. The function controlled by the locus might be a regulative function.  相似文献

Autoantibodies to components of chromatin, which include double-stranded DNA (dsDNA), histones and nucleosomes, are central in the pathogenesis of lupus nephritis. How anti-chromatin autoantibodies exert their nephritogenic activity, however, is controversial. One model assumes that autoantibodies initiate inflammation when they cross-react with intrinsic glomerular structures such as components of membranes, matrices or exposed nonchromatin ligands released from cells. Another model suggests glomerular deposition of autoantibodies in complex with chromatin, thereby inducing classic immune complex–mediated tissue damage. Recent data suggest acquired error of renal chromatin degradation due to the loss of renal DNaseI enzyme activity is an important contributing factor to the development of lupus nephritis in lupus-prone (NZBxNZW)F1 mice and in patients with lupus nephritis. Down-regulation of DNaseI expression results in reduced chromatin fragmentation and in deposition of extracellular chromatin–IgG complexes in glomerular basement membranes in individuals who produce IgG anti-chromatin autoantibodies. The main focus of the present review is to discuss whether exposed chromatin fragments in glomeruli are targeted by potentially nephritogenic anti-dsDNA autoantibodies or if the nephritogenic activity of these autoantibodies is explained by cross-reaction with intrinsic glomerular constituents or if both models coexist in diseased kidneys. In addition, the role of silencing of the renal DNaseI gene and the biological consequences of reduced chromatin fragmentation in nephritic kidneys are discussed.  相似文献

The analyses of R- and C-banding patterns of chromosomes of Arvicanthis niloticus originating from two different localities (Egypt and Central African Republic) revealed karyotypic differences caused by one pericentric inversion and three translocations, one being reciprocal and the others Robertsonian. There were also some differences in centromeric heterochromatin patterns.The data indicate that these two forms are distinct species, cytogenetically isolated, and that a revision of the taxonomic status of the genus Arvicanthis is needed.  相似文献

MOTIVATION: The vastness and complexity of the biochemical networks that have been mapped out by modern genomics calls for decomposition into subnetworks. Such networks can have inherent non-local features that require the global structure to be taken into account in the decomposition procedure. Furthermore, basic questions such as to what extent the network (graph theoretically) can be said to be built by distinct subnetworks are little studied. RESULTS: We present a method to decompose biochemical networks into subnetworks based on the global geometry of the network. This method enables us to analyze the full hierarchical organization of biochemical networks and is applied to 43 organisms from the WIT database. Two types of biochemical networks are considered: metabolic networks and whole-cellular networks (also including for example information processes). Conceptual and quantitative ways of describing the hierarchical ordering are discussed. The general picture of the metabolic networks arising from our study is that of a few core-clusters centred around the most highly connected substances enclosed by other substances in outer shells, and a few other well-defined subnetworks. Both chlorophyll a and b and polypeptides of the photosynthetic apparatus are found in gymnosperm seedlings. germinated and grown in absolute darkness. The photosystem II (PSII) activity is, however, limited, probably due to an inactive oxygen evolving system. In the present study dark-grown seedlings of Scots pine ( Pinus sylvestris L.) were transferred to light and changes in antenna size and the activation process of PSII were investigated using fluorescence measurements and quantitative western blotting. It was found that the activation process is rapid, requires very little light and that strong light inhibits the process. It takes place without any changes in the primary reactions of PSII. Furthermore, all polypeptides except the major light-harvesting chlorophyll a/b -binding protein complex of PSII (LHCII) were present in dark-grown seedlings in amounts comparable to the light treated control. The dark-grown seedlings had the same LHCII polypeptide composition as light treated seedlings, and the LHCII present seemed to be fully connected to the reaction centre. The results indicate that activation of PSII in dark-grown conifer seedlings resembles the photoactivation process of angiosperms. This implies that the fundamental processes in the assembly of the photosystem II complex is the same in all plants, but that the regulation differs between different taxa.  相似文献
