The aim of this study was to analyse the load-velocity and load-power relationships in the free-weight back-squat (BSQ) and hexagonal bar deadlift (HBD) exercises. Twenty-five (n = 25) resistance-trained men (age = 23.7 ± 2.8 years) performed a progressive load test at maximal intended velocity to determine their BSQ and HBD one-repetition maximum (1RM). Mean propulsive velocity (MPV) during the concentric phase of the lift was recorded through a linear encoder. Load-velocity and load-power relationships were analysed by fitting linear regression and the second-order polynomial, respectively, to the data. Maximum strength (1RM), MPV (30–80% 1RM), and power output (30–90% 1RM) were higher for HBD compared to BSQ exercise (p < 0.05). A very strong relationship between MPV and relative intensity was found for both BSQ (R2 = 0.963) and HBD (R2 = 0.967) exercises. The load that maximizes power output (Pmax) was 64.6 ± 2.9% (BSQ) and 59.6 ± 1.1% (HBD) 1RM. There was a range of loads at which power output was not different than Pmax (BSQ: 40–80% 1RM; HBD: 50–70% 1RM). In conclusion, the load-velocity and load-power relationships might assist strength and conditioning coaches to monitor and prescribe exercise intensity in the BSQ and HBD exercises using the velocity-based training approach. 相似文献

Phospholipases A2 (PLA2) are a group of enzymes that hydrolyze phospholipids at the sn-2 position, being present in all nature. In venomous animals, these proteins assume a special role, being able to exert diverse pharmacological effects. In this work, authors identified a new isoform of PLA2 in the venom of Porthidium hyoprora, which was isolated through sequential chromatographic steps and named PhTX-III. The enzyme was characterized biochemically and structurally. Structural studies using mass spectrometry confirmed an acidic secretory PLA2, family IIA, with molecular mass of 13,620.9 Da and identification of 86% of its primary sequence. PhTX-III did not exhibit myotoxic, anticoagulant or antibacterial effects, often present in this class of enzymes. Although, it was capable of initiate inflammatory response, with local edema and release of cytokines IL-1α, IL-6 and TNF-α, probably due to mast cell degranulation. 相似文献

The interest upon products obtained from fungi has increased during the recent years. Among the most noticeable, nutraceuticals, enzymes, and natural drugs occupy a privileged position. Fungal biomass for the obtainment of those products can be produced either by solid-state fermentation (SSF) or submersed fermentation. SSF has been employed for the production of spawn on pretreated wheat grains with the objective of increasing the fungal polysaccharide (glucomannans) contents. Among the important factors for the production of spawn, time of cooking, time of resting after grain cooking, consequently grain moisture, substrate pH, temperature of incubation, and initial inoculum amount are among the most significant. For wheat grains, cooking time of 21 min followed by a 24-min resting time has been shown as optimal for the production of glucomannans by the fungus Agaricus subrufescens (=Agaricus brasiliensis). Amendments of CaSO4 (up to 3 %) and CaCO3 (up to 1 %) had an important influence on the substrate pH. In general, better results for glucomannan production were obtained when no supplement was added or when up to 0.25 % CaCO3 (pH 6.6) has been added to the mix. Our results demonstrate that the inoculum amount necessary for the best polysaccharide levels is around 10.3 %, while the best temperature is around 27.2 °C. Besides using the spawn for its main purpose, it could potentially and alternatively be used as nutraceutical due to the high levels of glucomannan observed (6.89 %), a compound technically proven to be a potent immunostimulatory and antitumoral agent. 相似文献

Background
The error threshold puts a limit on the amount of information maintainable in Darwinian evolution. The error threshold was first formulated in terms of genotypes. However, if a genotype-phenotype map involves redundancy ("mutational neutrality"), the error threshold should be formulated in terms of phenotypes since there is no unique fittest genotype. yeast Yarrowia lipolytica. Both single- and multi-copy transformants were constructed, and the secretion of the enzyme was evaluated as an in-frame
fusion with the LIP2 secretion signal, as well as with its natural secretion signal. In shake-flask analysis, the highest volumetric enzyme activity
(13,073 nkat/ml) and specific enzyme activity (1,020 nkat/(mg dcw)) were obtained with a multi-copy integrant utilizing β-mannanase’s
