A dynamic view of cellular processes by in vivo fluorescence auto- and cross-correlation spectroscopy 总被引:6,自引:0,他引:6
Fluorescence correlation spectroscopy (FCS) is becoming increasingly popular as a technique that aims at complementing live cell images with biophysical information. This article provides both a short overview over recent intracellular FCS applications and a practical guide for investigators, who are seeking to integrate FCS into live cell imaging to obtain information on particle mobility, local concentrations, and molecular interactions. A brief introduction to the principles of FCS is provided, particularly emphasizing practical aspects such as the choice of appropriate dyes and positioning of the measurement volume in the sample. Possibilities and limitations in extracting parameters from autocorrelation curves are discussed, and attention is drawn to potential artifacts, such as photobleaching and probe aggregation. The principle of dual-color cross-correlation is reviewed along with considerations for proper setup and adjustment. Practical implications of nonideal conditions including incomplete focus overlap and spectral cross-talk are considered. Recent examples of both auto- and cross-correlation applications demonstrate the potential of FCS for cell biology. 相似文献

The composition of molecular species of phosphatidylethanolamines (PEs) and phosphatidylcholines (PCs) was analysed in fat body and muscle tissues of Chymomyza costata larvae of different physiological states that markedly differed in their level of freeze-tolerance. Actively moving and feeding 3rd instar larvae had low (zero) capacity of freeze-tolerance and similar phospholipid (PL) compositions irrespective of their developmental destiny (non-diapause vs. diapause). Extensive remodelling of PL composition was found in these larvae in response to: (a) chilling of non-diapause larvae at 5 degrees C for 1 month; (b) developmental transition to diapause; and (c) chilling of diapause larvae. Transition to diapause and chilling led to an increase in freeze-tolerance. The increase in molar proportion of molecular species containing palmitic/linoleic (16:0/18:2) fatty acyls (FAs) esterified to sn-1/sn-2 positions of glycerol was the most prominent change, which was tightly statistically correlated with increasing freeze-tolerance. The increase of PLs with combination of 16:0/18:2 FAs was registered consistently in PEs and PCs in fat body and muscle tissues in response to chilling and to diapause onset. This increase was countered by a decreases of various molecular species, depending on tissue and lipid class. Most decreasing species shared one common theme: they had a saturated FA (palmityl, margaryl, stearyl) esterified at sn-1 position and a monounsaturate (palmitoleyl, oleyl) esterified at sn-2 position of glycerol. Human adipose tissue expresses all components necessary for the local production of angiotensin II, which has multiple functions in adipose tissue, ranging from regulation of local blood flow to complex influences on tissue homeostasis. Still the mechanisms controlling human adipose tissue angiotensin II concentrations are not yet known. We investigated whether angiotensin II is degraded by human primary cultured preadipocytes and adipocytes and which enzymes are responsible for its metabolism. Distinct but transient angiotensin II production was limited by degradation due to consecutive proteolytic cleavage by endopeptidase and aminopeptidase activities. The endopeptidase could be identified as neprilysin expressed on the surface of both preadipocytes and adipocytes. Degradation of angiotensin II was preceded by a lag phase that was considerably longer in preadipocytes. This time span could not be explained by an induction of neprilysin nor by an increase in its surface localization. Following the lag phase, adipocytes showed a higher degradation activity than preadipocytes as mirrored by increased neprilysin levels and activity measured in their membrane fractions. Our findings demonstrate that human preadipocytes and adipocytes differentially express functional neprilysin and aminopeptidase activity involved in the regulation of angiotensin II concentrations in human adipose tissue. 相似文献

Caenorhabditis elegans has previously been used as an alternative to mammalian models of infection with bacterial pathogens. We have developed a liquid-based assay to measure the effect of bacteria on the feeding ability of C. elegans. Using this assay we have shown that Pseudomonas aeruginosa strain PA14, Burkholderia pseudomallei and Yersinia pestis were able to inhibit feeding of C. elegans strain N2. An increase in sensitivity of the assay was achieved by using C. elegans mutant phm-2, in place of the wild-type strain. Using this assay,P. aeruginosa PA01 inhibited the feeding of C. elegans mutant phm-2. Such liquid-based feeding assays are ideally suited to the high-throughput screening of mutants of bacterial pathogens. 相似文献

The present study evaluated whether the previously reported alterations in core temperature circadian rhythm associated with bed rest might be attributable to increased heat loss from the skin. Infra-red thermograms were obtained at weekly intervals during 5 weeks of bed rest and after 4 weeks of active recovery. Tympanic temperature (Tty) was measured at hourly intervals from 0800 to 2300 hrs on similar occasions during bed rest. There were no significant changes in mean tympanic temperature or amplitude of Tty circadian rhythm during the 5 week bed rest period. Skin temperature decreased progressively during bed rest (P<0.005), with distal regions being the most affected. 相似文献

Extracts of apple fruits (Malus domestica cv. "Brettacher") were analysed by HPLC with photodiode array detection. An unknown peak was monitored displaying the same retention time as isorhamnetin 3-O-glucoside. Preliminary identification of the isorhamnetin aglycone was performed by comparison of UV spectral data of the unknown compound with a reference substance. Using atmospheric pressure chemical ionisation mass spectrometry in the negative ion mode, the presence of an isorhamnetin glycoside was supported by loss of 162 amu from the pseudomolecular ion (m/z 477). MS2 product ion analysis of the parent ion m/z 477 provided a fragmentation pattern identical to the reference. Collision-induced dissociation of the aglycone (m/z 315) in the MS3 product ion analysis allowed the differentiation of rhamnetin and isorhamnetin, and unambiguous assignment by comparison with standard compounds. A second isorhamnetin glycoside eluting prior to the glucoside was tentatively identified as isorhamnetin 3-O-galactoside. To the best of our knowledge, this is the first report of isorhamnetin glycosides in apple fruit extracts. Results are discussed with respect to chemotaxonomic relevance within the genera Malus and Pyrus, and especially in consideration of the control of the authenticity of apple products. 相似文献

The Collaborative Research Center (CRC) 436 'Metal-Mediated Reactions Modeled after Nature' was founded for the express purpose of analyzing the catalytic principles of metallo-enzymes in order to construct efficient catalysts on a chemical basis. The structure of the active center and neighboring chemical environment in enzymes serves as a focal point for developing reactivity models for the chemical redesign of catalysts. Instead of simply copying enzyme construction, we strive to achieve new chemical intuition based on the results of long-lasting natural evolution. We hope for success, since nature uses a limited set of building blocks, whereas we can apply the full repertoire of chemistry. Key substrates in this approach are small molecules, such as CO2, O2 NO3- and N2. Nature complexes these substrates, activates them and performs chemical transformations--all within the active center of a metalloenzyme. In this article, we report on some aspects and first results of the Collaborative Research Center (CRC) 436, such as nitrate reductase, sphingolipid desaturase, carbonic anhydrase, leucine aminopeptidase and dopamine beta-monooxygenase. 相似文献
