Two years of physical/chemical and algal abundance data obtained from national Stream-Quality Accounting Network records of 10 river sites in Kentucky USA (4 Ohio River sites and 6 Ohio River tributaries) were analyzed to determine how seasonal changes in river phytoplankton related to changes in physical and chemical parameters. Phytoplankton assemblages differed among rivers as a function of drainage basin characteristics, but exhibited common seasonal changes related to temporal variation in the physical/chemical environment. Distinct shifts in algal dominance were identified between spring (March, April), late summer (July, August, September), and transitional (May, June, November) periods in the 10 systems. Nine common algal genera were found to differ in their response to changes in physical or chemical parameters. Abundances of Anacystis, Oscillatoria, Scenedesmus, and Melosira were strongly positively correlated with temperature while Chlamydomonas and Navicula abundances were inversely related to temperature. Other physical/chemical factors that were significantly positively ( + ) or negatively ( - ) correlated with algal abundances included discharge and alkalinity (Oscillatoria, +), pH CChlamydomonas and Cyclotella, (both + ), turbidity (Anacystis, \s- ; Navicula + ), silica (Cyclotella, - ), and ammonium/organic N (Anacystis, -). Genera within the same algal division exhibited different seasonal patterns and responded to different physical/chemical parameters.  相似文献

At least two different class II histocompatibility antigens, I-A and I-E, are encoded by the murine major histocompatibility complex. Both types of class II antigens are composed of polypeptide chains called alpha and beta. Class II antigens display extensive genetic polymorphism, the main part of which resides in the NH2-terminal domains of the A alpha, A beta and E beta chains. Recently it was shown that the mutant gene Abm 12 beta differed from the wild-type gene Ab beta by three nucleotide substitutions, which all occur within a stretch of 14 nucleotides. Multiple substitutions of the type found in the Abm 12 beta gene suggest that the mutant arose by a gene conversion-like event. To examine whether the Eb beta gene may have served as the donor gene in the generation of the Abm 12 beta gene, we have isolated and sequenced a cDNA clone corresponding to the Eb beta gene. Comparisons of the Eb beta, the Ab beta and the Abm 12 beta nucleotide sequences revealed that the Eb beta sequence is identical to that of Abm 12 beta in the positions where the latter differs from the Ab beta sequence. This observation is consistent with the notion that the Abm 12 beta mutant gene arose by a gene conversion-like event involving the Eb beta gene.  相似文献

A comparison of seven human DR and DC class II histocompatibility antigen beta-chain amino acid sequences indicates that the allelic variation is of comparable magnitude within the DR and DC beta-chain genes. Silent and replacement nucleotide substitutions in six DR and DC beta-chain sequences, as well as in seven murine class II sequences (three I-A beta and four I-A alpha alleles) were analyzed. The results suggest that the mutation rates are of a comparable magnitude in the nucleotide sequences encoding the first and second external domains of the class II molecules. Nevertheless, the allelic amino acid replacements are predominantly located in the first domains. We conclude that a conservative selective pressure acts on the second domains, whereas in many positions in the first domains replacement substitutions are selectively neutral or maybe even favoured. Thus, the difference between the first and second domains as regards the number of amino acid replacements is mainly due to selection.  相似文献

Previous work showed that the segl mutant of barley (Hordeum vulgare cv Betzes) did not differ from normal Betzes in plant growth, photosynthesis, or fertility, but it produced only shrunken seeds regardless of pollen source. To determine whether defects in sucrose uptake or starch synthesis resulted in the shrunken condition, developing grains of Betzes and segl were cultured in [14C]sucrose solutions after slicing transversely to expose the endosperm cavity and free space. In both young grains (before genotypes differed in dry weight) and older grains (17 days after anthesis, when segl grains were smaller than Betzes), sucrose uptake and starch synthesis were similar in both genotypes on a dry weight basis. To determine if sucrose was hydrolyzed during uptake, spikes of Betzes and segl were allowed to take up [fructose-U-14C]sucrose 14 days after anthesis and the radioactivity of endosperm sugars was examined during 3 hours of incubation. Whereas less total radioactivity entered the endosperm and the endosperm cavity (free space) of segl, in both genotypes over 96% of the label of endosperm sugars was in sucrose, and there was no apparent initial or progressive randomization of label among hexose moieties of sucrose as compared to the free space sampled after 1 hour of incubation. We conclude that segl endosperms are capable of normal sucrose uptake and starch synthesis and that hydrolysis of sucrose is not required for uptake in either genotype. Evidence suggests abnormal development of grain tissue of maternal origin during growth of segl grains.  相似文献

Plants of Agrostis tenuis Sibth., Hordeum vulgare L., Lycopersicon esculentum Mill. and Raphanus sativus L. were grown hydroponically in sealed systems and fumigated with 8 g m-3 [75Se]-dimethylselenide. The accumulation of 75Se was measured and the shoot tissues were extracted to examine the products of the 75Se assimilation. Characteristic differences were observed between species in the accumulation of 75Se and the transport from shoots to roots. High-voltage electrophoresis and chromatography of extracts made with 80% aqueous ethanol revealed the presence of inorganic selenite as an assimilation product as well as the selenium analogues of glutathione and methionine. ABSTRACT Habitat quality, as indexed by the reproductive success of individuals, can greatly influence population growth, especially for rare species near the limits of their range. Along the Pacific coast, the Snowy Plover (Charadrius alexandrinus nivosus) is a threatened species that, in recent years, has been breeding on both riverine gravel bars and ocean beaches in northern California. From 2001 to 2009, we compared the habitat characteristics, breeding phenology, reproductive success, and abundance of Western Snowy Plovers occupying these two habitats. Similar percentages of yearling and adult plovers returned to gravel bars and beaches, but plovers breeding on gravel bars arrived and initiated first clutches 2–3 weeks later than those breeding on beaches. Despite this delay, however, the mean annual fledging success of plovers on gravel bars (1.4 ± 0.4 [SD]) was double that on beaches (0.7 ± 0.3). Differences in cumulative reproductive success produced a stronger pattern. By their sixth year, males on gravel bars had fledged 14.5 ± 2.1 chicks, more than four times the number of young fledged by males on beaches (3.3 ± 3.1). Over 9 years, local population size decreased by about 75%, coincident with a shift in breeding distribution away from high‐quality gravel bars to ocean beaches. This unexpected population decline and shift to poorer quality beaches may have been related to occasional low survival of plovers that over‐winter exclusively on beaches in our study area. Consistently low productivity of plovers breeding on ocean beaches suggests the need for intensified management to ameliorate the negative impacts of predation and human activity on the recovery of this population.  相似文献

To establish a suitable experimental system for studies of the interaction of retinol-binding protein (RBP) with transthyretin (TTR) we have expressed the corresponding cDNAs in HeLa cells. To investigate whether complex formation might occur already in the endoplasmic reticulum (ER), the C-terminal ER retention signal, KDEL, was attached to TTR. The tetrameric TTR-KDEL fusion protein was retained in the ER of HeLa cells. When RBP was co-expressed with TTR-KDEL, RBP was retained intracellularly. A cDNA-encoding purpurin, a protein which is 50% identical to RBP, was then expressed together with TTR-KDEL. Purpurin was not retained intracellularly and did not bind to TTR coupled to Sepharose. The effect of the vitamin A status on the secretion of TTR and RBP was examined. While TTR expressed alone was not retained intracellularly, TTR was retained in vitamin A-deficient cells when co-expressed with RBP. Addition of retinol stimulated rapid secretion of both proteins. These results demonstrate that TTR can form a complex with RBP in the ER. The data suggest that RBP and TTR are secreted as a complex.  相似文献
