Mitchell, Gary F., Marc A. Pfeffer, Peter V. Finn, andJanice M. Pfeffer. Comparison of techniques for measuringpulse-wave velocity in the rat. J. Appl.Physiol. 82(1): 203-210, 1997.We evaluatedmethods for measuring average and regional pulse-wave velocity alongthe full length of the aorta in 18-mo-old ether-anesthetized malespontaneously hypertensive rats. Catheter-tip manometers were placed inthe ascending and descending thoracic aorta via the right carotid andleft femoral arteries, respectively. As the distal catheter waswithdrawn at 1-cm intervals, the relationship between distal catheterinsertion distance and distance between transducers was determined fromthe intercept of the insertion distance vs. transmission delayregression line. Methods that assessed the foot-to-foot time delaybetween pressures accurately predicted the separation between catheters(measured distance of 14.3 cm; intercept of 14.0 ± 0.5 cm;P = not significant) were highlyreproducible (coefficient of variation of 2.3% for repeated measurements) and showed minimal variability (range 509 ± 30 to 600 ± 29 cm/s) along the full length of the aorta. Methods that madeuse of the pressure-pressure transfer function were spatially (range ofvalues along the aorta 367 ± 17 to 722 ± 39 cm/s) and temporally more variable, especially during vasoconstriction with methoxamine, due to the effects of reflected waves.

Physiol. 82(4): 1250-1255, 1997.Fast-twitch skeletal muscles contain more neuronal-type nitricoxide synthase (nNOS) than slow-twitch muscles because nNOS is presentonly in fast (type II) muscle fibers. Chronic in vivo electricalstimulation of tibialis anterior and extensor digitorum longus musclesof rabbits was used as a method of inducing fast-to-slow fiber typetransformation. We have studied whether an increase in musclecontractile activity induced by electrical stimulation alters nNOSexpression, and if so, whether the nNOS expression decreases to thelevels present in slow muscles. Changes in the expression of myosinheavy chain isoforms and maximum velocity of shortening of skinnedfibers indicated characteristic fast-to-slow fiber type transformationafter 3 wk of stimulation. At the same time, activity of NOS doubled inthe stimulated muscles, and this correlated with an increase in theexpression of nNOS shown by immunoblot analysis. These data suggestthat nNOS expression in skeletal muscle is regulated by muscle activityand that this regulation does not necessarily follow the fast-twitchand slow-twitch pattern during the dynamic phase of phenotypetransformation.

Klebsiella aerogenes forms electron-dense partieles on the cell surface in response to the presence of cadmium ions in the growth medium. These particles ranged from 20 to 200 nm in size, and quantitative energy dispersive X-ray analysis established that they comprise cadmium and sulfur in a 1:1 ratio. This observation leads to the conclusion that the particles are cadmium sulfide crystallites. A combination of atomic absorption spectroscopy, inductively coupled plasma mass spectrometry, and acid-labile sulfide analysis revealed that the total intracellular and bound extracellular cadmium:sulfur ratio is also 1:1, which suggests that the bulk of the cadmium is fixed as extracellular cadmium sulfide. The tolerance of K. acrogenes to cadmium ions and the formation of the cadmium sulfide crystallites were dependent on the buffer composition of the growth medium. The addition of cadmium ions to phosphate-buffered media resulted in cadmium phosphate precipitates that remove the potentially toxic cadmium ions from the growth medium. Electrondense particles formed on the surfaces of bacteria grown under these conditions were a combination of cadmium sulfide and cadmium phosphates. The specific bacterial growth rate in the exponential phase of batch cultures was not affected by up to 2mM cadmium in Tricine-buffered medium, but formation of cadmium sulfide crystallites was maximal during the stationary phase of batch culture. Cadmium tolerance was much lower (10 to 150 M) in growth media buffered with Tris, Bistris propane, Bes, Tes, or Hepes. Trichosporon beigelii SBUG 752 was able to transform diphenyl ether. By TLC, HPLC, GC, GC-MS, NMR- and UV-spectroscopy, several oxidation products were identified. The primary attack was initiated by a monooxygenation step, resulting in the formation of 4-hydroxydiphenyl ether, 2-hydroxydiphenyl ether and 3-hydroxydiphenyl ether (48:47:5). Further oxidation led to 3,4-dihydroxydiphenyl ether. As a characteristic product resulting from the cleavage of an aromatic ring, the lactone of 2-hydroxy-4-phenoxymuconic acid was identified. The possible mechanism of ring cleavage to yield this metabolite is discussed.  相似文献

The daily range of feral goats varied in relation to social class and environmental variables. Mean daily ranges for 5 social classes were: male herd, 3.52 × 105 m2; female herd, 2.12 × 105 m2; composite herd, 1.86 × 105 m2; stayer female, 0.57 × 105 m2; creche group, 0.39 × 105 m2. Total recorded home range was male herd 14.8 × 105 m2 and female herd 9.65 × 105 m2. Female herd daily range varied seasonally in terms of size, location and habitat utilization. Female herd daily range area was correlated significantly with number of days since rain (r = 0.412), amount of rain in the past 30 days (r = ?0.296), maximum daily temperature (r = ?0.409), and minimum daily temperature (r = ?0.523), but not with wind velocity in the morning (r = ?0.064) or afternoon (r = ?0.137).  相似文献

Summary Visual unit activity, EEG changes and sustained potential shifts (SPS) were recorded from the toad tectum whilst the animal was presented with a visual stimulus. Telencephalic EEGs were also recorded.On the surface of the tectum, retinal unit activity preceded a sustained negative shift in potential and an increase in the amplitude and dominant frequency of the EEG. In deeper layers of the tectum, T5 units with configurational selectivity for wormlike stimuli were found. The activity of these units followed a pronounced SPS and EEG change.Visual unit activity was most pronounced during the negative-going phase of the synchronised EEG, when there was also a small decrease in amplitude of neuronal spikes. Similarities between the latencies and durations of EEGs and SPSs, and their response decrements, on repeated stimulus presentation, implies a close relationship between them not shared by the visual units studied. Using a 200 kV electron microscope (JEM 200 A), thick (up to 0.4 μm) crosssections of the myosin filaments of vertebrate striated muscle were studied. It was found that: (a) with increasing section thickness the cross-sectional profiles of the shaft of the filament were increasingly more triangular and in sections 0.4 μm thick each apex of the triangle was clearly blunted. This unique cross-sectional profile is predicted by the model proposed by Pepe (1966,1967) in which 12 parallel structural units are packed to form a triangular profile with a structural unit missing at each apex of the triangle. (b) With increasing section thickness the substructure of the myosin filament was enhanced, with the best substructure visible in sections 0.2 μm to 0.3 μm thick. This strongly supports parallel alignment of structural units in the shaft of the filament as proposed by Pepe (1966,1967). (c) The substructure spacing, determined by optical diffraction from electron micrographs of cross-sections of individual myosin filaments or groups of filaments is about 4 nm. (d) The different optical diffraction patterns observed from individual myosin filaments can be explained if the projection of each structural unit in the plane of the section has an elongated profile. With a substructure spacing of 4 nm an elongated cross-sectional profile could be produced by having two myosin molecules per structural unit. Models drawn with two myosin molecules per structural unit in the model proposed by Pepe (1966,1967) gave optical diffraction patterns similar to those observed from individual filaments. (e) The different optical diffraction patterns observed from individual myosin filaments can be explained if the elongated profiles in each structural unit are similarly oriented but with the orientation changing along the length of the filament. The change in orientation per unit length of the filament must be small enough to maintain an elongated profile for the projection of the structural unit in the plane of the sections 0.3 μm thick. All of these observations and conclusions strongly support the model for the myosin filament proposed by Pepe (1966,1967).  相似文献
