Antibodies to histamine were used to examine the localization of the amine in cells of the stomach and upper small intestine of a great variety of species, including cartilaginous and bony fish, amphibia, reptiles (lizard), birds (chicken) and a large number of mammals. In all species gastric histamine was localized in endocrine cells (invariably found in the epithelium) and mast cells (usually with an extra-epithelial localization). The endocrine cells were identified as such by immunostaining with antibodies to chromogranin A and the mast cells were identified by toluidine blue staining. Histamine-immunoreactive endocrine cells were found almost exclusively in the acid-producing part of the stomach; only rarely were such cells observed in the pyloric gland area. They were fairly numerous in the gastric mucosa of the two subclasses of fish as well as in the amphibia and reptile species studied. Here, the majority of the histamine-immunoreactive endocrine cells seemed to have contact with the gastric lumen (open type cells) and were located in the surface epithelium (certain fish only) or together with mucous neck cells at the bottom of the pits. In the chicken, histamine-immunoreactive endocrine cells were numerous and located peripherally in the deep compound glands. They were without contact with the lumen (closed type) and had long basal extensions ("paracrine" appearance), running close to the base of the oxyntic-peptic cells. In mammals, the number of histamine-immunoreactive endocrine cells in the stomach varied greatly. They were particularly numerous in the rat and notably few in the dog, monkey and man. In all mammals, the histamine-immunoreactive endocrine cells were of the closed type and located basally in the oxyntic glands. They often had a "paracrine" appearance with long basal processes. Histamine-storing mast cells, finally, were few in both subclasses of fish as well as in the amphibian species and in the lizard. They were fairly numerous in chicken proventriculus (beneath the surface epithelium), few in the oxyntic mucosa of mouse, rat and hamster, moderate in number in hedgehog, guinea-pig, rabbit, pig and monkey, and numerous in cat, dog and man.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Phyloplankton structure and its relation to physical and chemical properties of the water was studied in 58 central Finnish lakes. The biomass ranged from 0.2 to 14.2 g m−3 and the number of taxa per sample ranged from 33 to 152. The lakes were grouped into 5 types according to their trophic state: eutrophic, dyseutrophic, mesotrophic, oligotrophic, and acid oligotrophic lakes. The average biomass in eutrophic lakes was 5.57 g m−3, in dyseutrophic 3.54 g m−3, 1.23 g m−3 in mesotrophic, 0.52 g m−3 in oligotrophic and 0.39 g −3 in acid oligotrophic lakes. The average number of taxa per sample in the corresponding lake types were 109. 1, 79.3, 97.9, 90.9 and 43.8, respectively. The phytoplankton communities in eutrophic lakes were characterized by blue-green algae (21.2% of total biomass) and green algae (18.7% of total biomass). In dyseutrophic lakes the proportion of green algae was much smaller (7.2% of total biomass) than in eutrophic lakes, whereas the proportion of diatoms and cryptophytes was higher (28.2 and 20.4% of total biomass, respectively). Chrysophytes dominated in the oligotrophic and mesotrophic lakes (27.3–39.9% of total biomass). The contribution of dinoflagellates to the total biomass was highest in the most oligotrophic acidified lakes and in those lakes the relative proportions of blue-green and green algae were much higher than in the typical oligotrophic lakes. The lakes were also grouped into 8 community types according to the dominating algal group. Cyanophyceae- and Chlorophyceae-types characterized the eutrophic lakes, whereas Chrysophyceae-Dinopheceae-type was typical for most oligotrophic lakes. The other 5 types occurred in mesotrophic and oligotrophic lakes but the physical and chemical properties of these lakes did not differ much.  相似文献

Aerobic N2O production was studied in nitrifying humus from urea-fertilized pine forest soil. Acetylene and nitrapyrin inhibited both NH4+ oxidation and N2O production, indicating that N2O production was closely associated with autotrophic NH4+ oxidation. N2O production was enhanced by low soil pH; it was negligible above pH 4.7. When soil pH decreased from 4.7 to 4.1, the relative amount of N2O-N produced from NH4+-N oxidized increased exponentially to 20%. There was also some evidence that N2O formation was stimulated by salts (potassium sulfate and sodium phosphates). The maximum rate of N2O-N production was 0.17 μg of N2O-N per g of soil per h. When humus was treated with NO2, N2O evolved immediately, indicating chemical formation, but no N2O was formed on the addition of NO3. The amount of N2O-N evolved was 0.6 to 4.6% of NO2-N added. A high concentration of NO2 and low soil pH enhanced chemical production of N2O. There was no accumulation of NO2 during nitrification. The calculations indicated that chemical formation of N2O was not the main source of N2O during NH4+ oxidation. After the addition of inhibitors of NH4+ oxidation the soils contained NO3, but no N2O was produced. The results suggest that enhanced autotrophic NH4+ oxidation is a potential source of N2O in fertilized acid forest soil.  相似文献

Diatom assemblages from the topmost sediment of 49 lakes were ordinated by means of a program for detrended correspondence analysis and reciprocal averaging (Decorana). Five lake groups were separated, each having more or less characteristic diatom assemblages and water quality. Shifts from one group to another caused by factors such as acidification, artificial drainage, diffuse nutrient loading and sewage are discussed.  相似文献

Abstract Chromosomal DNA fragments from Chlamydia trachomatis serotype L2 were shotgun-cloned into pBR322. A C. trachomatis -specific clone was further subcloned to produce specific DNA reagents for the identification of C. trachomatis by nucleic acid sandwich hybridization. The chosen DNA reagents from serotype L2 also hybridized with all the other chlamydial serotypes tested, but not with the DNA from 41 unrelated organisms. The sensitivity of the sandwich hybridization test was 106 DNA molecules. The applicability of the test for routine diagnostic use was demonstrated by a pilot study in which C. trachomatis was directly detected from genital specimens.  相似文献

Factors affecting the activity of human lysozyme (EC 3.2.1.17) toward cell suspensions of Micrococcus lysodeikticus were reexamined. Effects of substrate concentration, pH, and ionic strength and matrix effects of protein were assessed with special emphasis on the interdependence of various parameters. On the basis of these evaluations, an optimized kinetic turbidimetric method for lysozyme assay was set up. The method was applied for automation with a System Olli 3000 analyzer. The new automated lysozyme assay proved good for routine clinical use in regard to analysis speed, sensitivity, linearity, and reproducibility. Reference values for serum, urinary, and cerebrospinal fluid lysozyme were assessed with the automated method.  相似文献

Summary The coexistence of histamine, histidine decarboxylase (the enzyme synthesizing histamine), 5-hydroxytryptamine and tyrosine hydroxylase (the rate-limiting enzyme in catecholamine synthesis), was studied in the rat superior cervical ganglion with the indirect immunofluorescence method. Possible colocalization was examined by staining consecutive sections with two different antibodies, or alternatively in the same section by eluting the first antibody with a mild solution containing potassium permanganate and sulphuric acid, and by staining the same section with another antibody. It was shown that tyrosine hydroxylase immunoreactivity was found both in large principal nerve cells and in small cells, which on the basis of their size and high nucleus—cytoplasm ratio corresponded to small intensely fluorescent (SIF) cells. Histamine, histidine decarboxylase and 5-hydroxytryptamine immunoreactivities were observed only in SIF cells. Those SIF cells which were immunoreactive for histamine, histidine decarboxylase or 5-hydroxytryptamine also contained tyrosine hydroxylase immunoreactivity. On the other hand, all tyrosine hydroxylase-immunoreactive SIF cells were also immunoreactive for histidine decarboxylase or 5-hydroxytryptamine. Some of the SIF cells, which were non-reactive for histamine, were immunoreactive for tyrosine hydroxylase.  相似文献

A palaeolimnological study of Lake Iso-Hietajärvi in Patvinsuo National Park, Lieksa, East Finland was conducted. The drainage area of the lake is one of four Integrated Monitoring areas established in Finland. Lead-210 dating reveals a period of increased sedimentation in the lake from 1920 to 1950. Increased atmospheric burden of several heavy metals in the order Pb > Cu> Zn > Ti = Al > Cr = Ni = V is recorded. The first to expand during the 19th century is Pb, whilst V increases after 1950. Sedimentary chlorophyll derivatives expand in the early part of the 20th century. Assemblages of diatoms and Cladocera were also changed somewhat during this time, but water quality seems not to have varied much: e.g. the diatom-inferred pH has remained in the range 6.4–6.8 (with a slight decrease) throughout the period of study.  相似文献

Summary The association between mast cells (visualized by routine staining and immunohistochemistry for histamine) and capsaicin-sensitive nerves (containing calcitonin gene-related peptide (CGRP) and substance P (SP)) was studied in the pig. In the 1-ethyl-3(3-diethylaminopropyl)carbodiimide (EDCDI)-fixed skin tissue, histamine-containing mast cells and CGRP/SP-positive nerves were found in close association around blood vessels. In the EDCDI-fixed airway mucosa, only single histamine-containing mast cells were detected. However, many alcian blue-positive mast cells were found, sometimes close to the airway epithelium where CGRP/SP-containing nerve fibres were absent 2 days after systemic capsaicin pretreatment, but no changes in the number and distribution of tissue mast cells, granulocytes or lymphocytes, or the number of blood leukocytes were detected. Local injection of allergen, histamine and capsaicin into the skin of pigs actively sensitized with ascaris antigen caused a rapid light red-flare (vasodilation) reaction. Allergen and histamine, but not capsaicin, also produced plasma protein extravasation. In contrast to the absent flare, the protein extravasation response still occurred in capsaicin-treated pigs. The sensitivity to ascaris antigen was mediated by an IgE-like antibody. We conclude that a functional and morphological relationship exists between histamine-containing mast cells and capsaicin-sensitive sensory nerves in the pig skin. Mast cells and sensory nerves are also found in the airway mucosa and appear to be closely associated with the epithelium.  相似文献

A linkage map spanning the locus for diastrophic dysplasia (DTD)   总被引:2,自引:0,他引:2  
Diastrophic dysplasia (DTD) is an autosomal recessive osteochondrodysplasia. Patients have short-limbed short stature and suffer from generalized joint dysplasia. We have recently mapped DTD to the distal long arm of chromosome 5. Here we report the localization of DTD in relation to 16 polymorphic markers on distal 5q. No recombinations occurred with two loci, D5S72 and D5S66. One presumptive candidate gene, osteonectin (SPARC), could be excluded on the basis of three recombinations with the DTD locus. Multipoint linkage analysis performed against a fixed order of markers placed DTD between glucocorticoid receptor (GRL) and SPARC favored by the odds of 33:1 over the next best location of DTD between D5S72 and D5S55. The sex-averaged distance between the definite flanking markers, GRL and D5S55, is 17.5 cM. From previously reported data on the physical localization of markers, we conclude that the DTD locus is in 5q31-q34.  相似文献
