Information on spatial and temporal patterns of genetic diversity is a prerequisite to understanding the demography of populations, and is fundamental to successful management and conservation of species. In the sea, it has been observed that oceanographic and other physical forces can constitute barriers to gene flow that may result in similar population genetic structures in different species. Such similarities among species would greatly simplify management of genetic biodiversity. Here, we tested for shared genetic patterns in a complex marine area, the Baltic Sea. We assessed spatial patterns of intraspecific genetic diversity and differentiation in seven ecologically important species of the Baltic ecosystem—Atlantic herring (Clupea harengus), northern pike (Esox lucius), European whitefish (Coregonus lavaretus), three-spined stickleback (Gasterosteus aculeatus), nine-spined stickleback (Pungitius pungitius), blue mussel (Mytilus spp.), and bladderwrack (Fucus vesiculosus). We used nuclear genetic data of putatively neutral microsatellite and SNP loci from samples collected from seven regions throughout the Baltic Sea, and reference samples from North Atlantic areas. Overall, patterns of genetic diversity and differentiation among sampling regions were unique for each species, although all six species with Atlantic samples indicated strong resistence to Atlantic-Baltic gene-flow. Major genetic barriers were not shared among species within the Baltic Sea; most species show genetic heterogeneity, but significant isolation by distance was only detected in pike and whitefish. These species-specific patterns of genetic structure preclude generalizations and emphasize the need to undertake genetic surveys for species separately, and to design management plans taking into consideration the specific structures of each species.  相似文献

The use of colorimetric methods for protein quantification in microalgae is hindered by their elevated amounts of membrane‐embedded intracellular proteins. In this work, the protein content of three species of microalgae was determined by the Lowry method after the cells were dried, ball‐milled, and treated with the detergent sodium dodecyl sulfate (SDS). Results demonstrated that the association of milling and SDS treatment resulted in a 3‐ to 7‐fold increase in protein quantification. Milling promoted microalgal disaggregation and cell wall disruption enabling access of the SDS detergent to the microalgal intracellular membrane proteins and their efficient solubilization and quantification.  相似文献

This study aimed to understand the use of “invasive species” as a normative concept and discuss its implications in conservation science, using introduced ascidians worldwide as model species. A specific search in Web of Science was performed and articles suitable for analysis were selected. Each article was classified according to the type of environment, species under study, type of effects and spread that ascidians are linked to. Most of the 184 articles analysed did not consider dispersal or effects as study subject (82 and 71%, respectively). Most research was conducted in laboratory conditions (41%) or human-made environments (32%) or indicating few escapes to natural environments. Almost half of the articles (47%) were made with the six more conspicuous introduced ascidians and this raised to 70% while considering articles that worked with two or more (pooled) species. The normative use of “invasive” is widely used regarding introduced ascidians. Spread and effects, necessary conditions to consider a species as invasive, are notoriously understudied. Most research was not conducted in natural environments and over a few species, weakening the perception of introduced ascidians as a conservation problem. To discuss the extent of the normative use of invasion science is important to distinguish two phenomena: are some species intrinsically problematic for conservation (i.e. invasive) or is the movement of non-native species (i.e. biological invasion) the conservation problem? By using invasive as a normative concept, we risk ending with a weakened concept potentially hindering the progress of invasion science.  相似文献

Invasion science has not been developed without controversies. Two questions that are still unsolved are: what is an invasive species?, and are invasive species an inherent conservation problem? These questions have led to discussions about effects versus origins. In contrast to the definitional problems, a unified framework describing invasion as a step‐by‐step process has been widely accepted. I conducted a bibliographic search with two separate databases searching for (i) evidence of less use of controversial terms over time; (ii) how many articles defined ‘invasive species’; (iii) the criteria used to define a species as invasive; and (iv) in which stage of the invasion continuum were species labelled as invasive located. My results show that controversial terms are widely used, that authors rarely define ‘invasive species’ and, often, it is very complicated to determine which criterion they used. In addition, only a fraction of the species labelled as invasive could be classified as such according to the unified framework of invasion stages. This is not a merely semantic issue, because invasive is a strong and value‐laden term that is used to guide environmental agendas. The uncritical use of a key concept could hamper research, complicate communication among peers and produce mixed results.  相似文献

The question of developmental relationships amongst myelin-related membranes in subfractions of myelinating mouse brain (15 days) was investigated by a time-staggered double isotope protocol using [3H]leucine and [14C]leucine. Preliminary results are interpreted and discussed in the context of a mathematical conceptualization of pulse-labeling kinetic analyses of myelin proteins in subcellular membrane compartments. Differences in ratio of the two leucine labels among proteins of myelin-containing subfractions are interpreted as confirming metabolic differences relating to various stages of development rather than precursor-product relationships. The incorporation into myelin of 14K, 17K, and 18.5K basic proteins (MBPs) occurs with relatively short delay times, following their synthesis (less than 5 min), and seems to occur simultaneously into all compartments. The 21.5K MBP and the proteolipid protein, on the other hand, require 10-14 min and 14-20 min, respectively. A scheme is presented to illustrate the probable assignment of subfractions to various myelin "compartments" during myelination, and to serve as a working hypothesis for studies on precursor-product relationships.  相似文献

This study investigates the effect of intraperitoneal injection of L. bulgaricus and S. thermophilus on interferon production by Swiss mice. The serum from mice given 5 x 10(7) L. bulgaricus in 0.5 ml saline showed a maximal production of 300 U/ml of alpha/beta interferon activity six hours after injection. Cellular integrity appears to be necessary for stimulation; heat-treated bacteria had little effect, while irradiated-bacteria had a greater effect. TNF was also produced, the sera of mice with high IFN also contained 300 U/ml TNF. Streptococcus thermophilus produced no detectable increase in serum IFN, but the 2'-5' A synthetase activity of peritoneal cells was elevated suggesting that small amounts of interferon were produced. Injection of Streptococcus thermophilus plus Lactobacillus bulgaricus did not change the serum interferon response to L. bulgaricus. These observations suggest that non-pathogenic bacteria such as those used in food processing, can stimulate IFN production in mice. There is some evidence that the bacterial cell walls might be responsible for at least part of this effect.  相似文献

We report here the isolation of fractions enriched in components of the myelin-like membranes surrounding the giant axons of the earthworm.Lumbricus terrestris L. The composition and purity of the fractions have been assessed using SDS-protein electrophoresis, Western immunoblots, and electron microscopy. Preliminary enzyme assays indicated that the mitochondrial marker, succinate dehydrogenase, has a similar specific activity distribution in earthworm nerve cord and in mouse liver sedimentation velocity fractions, however, the distribution of the total units of activity among the fractions seems to indicate the existence of smaller mitochondria in earthworm nerve cord compared with mouse liver mitochondria. In earthworm nerve cord fractions, Na+/K+ ATPase and Ca2+/Mg2+ ATPase were found to be enriched exclusively in the fraction containing large plasma and myelin-like membranes, while in the mouse liver fractions, the total units of these two enzymes were found to be distributed broadly among fractions. 5-Nucleotidase activity in the earthworm nerve cord seemed to be restricted to the microsomal fractions (endomembrane network), with a very low activity associated with the large plasma and myelin-like membrane fraction. We have established the presence of keratins or prekeratins in the myelin-like membranes, probably in the form of tonofilaments. However, we could not show that the desmosome-like structures, characteristic of these membranes, are composed of those proteins described for vertebrate epithelial desmosomes.  相似文献

The production of interferon by fasted human subjects in response to lactic bacteria Lactobacillus bulgaricus and Streptococcus thermophilus was evaluated in vivo and in vitro. The 2'-5' A synthetase activity of blood mononuclear cells was used to estimate interferon production following a single ingestion of 10(11) bacteria in yoghurt or sterile milk (controls). The level of the 2'-5' A synthetase of the yoghurt fed subjects was 83% (p = 0.002) higher than that of the milk fed controls 24 hours after ingestion. The baseline value remained unchanged in the control group. Blood mononuclear cells from a second group of subjects, were cultured with lactic bacteria for 48 hours, their cell-free supernatants contained gamma interferon. These results suggest that a transient production of interferon can be induced in healthy subjects by the lactic bacteria used in food processing.  相似文献
