Upon penetration in an unfertilized Xenopus egg bathed in 1/10 Ringer, the voltage recorded by a microelectrode shows an abrupt jump to a negative voltage (Ep) followed by a rapid depolarization to a steady value (Er) (Ep = -39.4 +/- 1.9 mV and Er = -11.5 +/- 0.5 SE, 54 eggs from 9 females). The same is true for fertilized eggs impaled 16-35 min after insemination (Ep = -29.5 +/- 2.1 mV, Er = -11.5 +/- 0.9 mV, SE, 18 eggs from 3 females). The voltage recorded by a second microelectrode inserted into the same egg does not show the transient initial negativity. The stationary level of the membrane potential is close to the diffusion potential calculated from the Goldman equation with equal permeabilities for all the relevant ions. It is concluded that the low resting potentials measured in Xenopus eggs before and after fertilization are largely due to damage caused by the electrode. Using an upper limit of -39 mV for the true membrane potential and correlating the input resistance with the stationary membrane potential, a lower limit of 22 M omega (about 1 M omega cm2) for the membrane resistance can be obtained. Insertion of a microelectrode during the first 3 min after insemination shows a steady positive potential while, at later times (3-16 min post-insemination), a positive peak followed by a repolarization can be observed. This indicates that the measurement of the peak of the fertilization potential is not seriously affected by the electrode penetration while its time course after the first 3 min may be deformed by the presence of a large leakage conductance.  相似文献

Aim Large herbivores have important effects upon Paleotropical ecosystems, but attain much lower biomass densities in the Neotropics. We assess how this difference in herbivore activity has generated different ecological and evolutionary trajectories in the New and Old World tropics. We also propose an explanation for how the greater biomass density in the Old World came about. Location Data were compiled primarily from moist tropical forests, although more of the relevant information to address most of our hypotheses was available from the mainland areas of Africa, Asia, and South America than elsewhere. Methods We gleaned data from published information and personal communication. We compared body masses and a variety of other types of information for the New‐ and Old‐World tropics. We proposed that interhemispheric differences exist in a variety of processes, including herbivory, frugivory, and flower visitation. We erected hypotheses and evaluated them qualitatively, and, when information was available, tested them using simple ratios of species in various taxonomic and trophic categories. To make the comparisons more meaningful, we specified appropriate data selection criteria. Results A general pattern of differences emerges from this review. Compared with Neotropical forests, the much greater biomass densities of large herbivores in Paleotropical forests are associated with a lesser diversity of small herbivores, different hunting methods used by indigenous humans, larger arboreal vertebrates, larger fruits, different patterns of fruit and flower dispersion in space and time, a lesser abundance of most types of reproductive plant parts, and other features. The existence of a species‐rich fauna of large herbivores in the pre‐Holocene Neotropical rain forest was not supported. Main conclusions The potential for large herbivores to cause functional differences between the New and Old World tropical forests has been virtually unexplored, despite the well‐known importance of large herbivores in the Old World tropics. The evaluations of our hypotheses suggest that the abundance of large herbivores in the Old World tropics has launched it onto a different evolutionary trajectory than that of the NewWorld tropics. The relevant evidence, although scanty, suggests that the interhemispheric ecological differences are not an artefact of recent megafaunal extinctions in the New World. Recent human activities have, however, reduced population sizes of large wild herbivores in the Old World, and increased population sizes of livestock. This has likely created a rather homogeneous, anthropogenic selection pressure that tends to erase the evolutionary differences between the two tropical worlds.  相似文献

A direct numerical integration method, as modified by Du Fort and Frankel (1953), has been used to solve the partial differential equation system which describes the spread of action potential in a mammalian nerve terminal. Branching of the terminal as well as inhomogeneous distributions of Na+ and K+ voltage-dependent channels (Brigant and Mallart 1982) have been incorporated in the model.Using the channel densities and the kinetic parameters measured in the node of Ranvier, the depolarization in the terminal branches has an amplitude of only 60% of the action potential in the node. Furthermore, the time courses of the calculated membrane currents differ considerably from the ones measured by Brigant and Mallart (1982) and by Konishi and Sears (1984).Increasing the Na+ and K+ channel densities may considerably increase the terminal depolarization and also reproduce qualitatively the current waveforms observed experimentally. The model can also reproduce some of the effects of pharmacological channel blocks.The simulation allows a new interpretation of the different components of membrane current along the terminal.  相似文献

1. Peripheral blood reiculocytes of normal and bled rabbits and of rabbits with phenylhydrazine-induced anemia, were morphologically analysed, through silver sections, for a relative evaluation of hemoglobin (Hb) biosynthesis activity. 2. Reticulocytes of maturation degrees within the range of 35-60 polysomes/microns2, were compared as to their mean numbers of hemosomes (sites of heme integration into the globin chains), and mitochondria (indirect precursors for hemosome formation). 3. The results on the mean numbers of hemosomes per reticulocyte section, correlated to several physiological data under those three conditions, suggested a close relationship between Hb biosynthesis activity and hemosome frequency. 4. In bled rabbits, reticulocytes showing a low mean number of hemosomes (means hB/section = 0.32), as compared to reticulocytes of normal rabbits (means hN/section = 0.70) and to reticulocytes of rabbits with hemolytic anemia (means hH/section = 2.10), gave rise to a new erythrocyte population characterized by a low Hb content. 5. Hb concentration differences were verified by confronting hematological data before bleeding with those obtained after the regression of anemia.  相似文献

Uncovering what predicts genetic diversity (GD) within species can help us access the status of populations and their evolutionary potential. Traits related to effective population size show a proportional association to GD, but evidence supports life-history strategies and habitat as the drivers of GD variation. Instead of investigating highly divergent taxa, focusing on one group could help to elucidate the factors influencing the GD. Additionally, most empirical data is based on vertebrate taxa; therefore, we might be missing novel patterns of GD found in neglected invertebrate groups. Here, we investigated the predictors of the GD in crabs (Brachyura) by compiling the most comprehensive cytochrome c oxidase subunit I (COI) available. Eight predictor variables were analysed across 150 species (16 992 sequences) using linear models (multiple linear regression) and comparative methods (PGLS). Our results indicate that population size fluctuation represents the most critical trait predicting GD, with species that have undergone bottlenecks followed by population expansion showing lower GD. Egg size, pelagic larval duration and habitat might play a role probably because of their association with how species respond to disturbances. Ultimately, K-strategists that have undergone bottlenecks are the species showing lower GD. Some variables do not show an association with GD as expected, most likely due to the taxon-specific role of some predictors, which should be considered in further investigations and generalizations. This work highlights the complexity underlying the predictors of GD and adds results from a marine invertebrate group to the current understanding of this topic.  相似文献

Estradiol directly stimulated the secretion of prolactin by the adenohypophyseal cells of intact rats in a monolayer culture. Complex interactions between estradiol and some other regulators of the hypophyseal lactotropic function were revealed. It was established that changes in prolactin synthesis and secretion in ovariectomized rats persisted for 3--4 days of adenohypophyseal cell cultivation in vitro.  相似文献
