Summary Knowing the ontogenesis of the central monoamine neurons of the rat it is possible to obtain, by free-hand dissection from embryos and newly born animals, pieces containing dopamine (DA), noradrenaline (NA), and 5-hydroxytryptamine (5-HT) neurons that are small enough to permit homologous transplantation to the anterior chamber of the eye of adult animals. With this technique it was established that all three types of immature monoamine neurons are able to survive in the anterior chamber. Fluorescence histochemical analysis of whole mount preparations of the sympathetically denervated host irides revealed that both the catecholamine- and the 5-HT-neurons are able to partly reinnervate the irides, forming networks of varicose nerve terminals similar to the normally present sympathetic adrenergic ground plexus.Monoamine nerve cell bodies are attached to the irides but the majority of fluorescent nerve cell bodies is located within the transplants. Serial sectioning of these transplants showed rather well organized brain tissue, containing groups of fluorescent and non-fluorescent cell bodies, many areas being innervated by monoamine nerve terminals. When brain tissue was transplanted before the normal appearance of fluorescent neuroblasts (embryos with a crown-rump length less than 8 mm) monoamine neurons developed and matured within the eye.The amount of newly formed nerves of central origin recovered on the irides increased with time between the 2nd and 4th postoperative week and persisted after 2 months. The yield of new fibers was better using transplants from embryos with a crown-rump length between 15 and 30 mm than using transplants from larger embryos and newly born animals.If embryonic brain tissue known to be devoid of monoamine nerve cell bodies but containing monoamine nerve terminals in the adult state (cortex cerebri and cerebelli, spinal cord) was transplanted to sympathetically non-denervated eyes, the sympathetic adrenergic fibers seemed to be able to innervate the transplants.This work was supported by grants from the Swedish Medical Research Council (14×–3185), Karolinska Institutets fonder, and Magnus Bergvalls Stiftelse. The effect of nitrogen, phosphorus and potassium nutrition on average seed weight, oil content and fatty acid composition of rape seed (Brassica napus) grown in soil-free culture has been studied. Nitrogen effected an increase in seed weight and a decrease in oil content, while the average amount of oil per seed remained constant. A small, but highly significant, decrease in palmitic and eicosenoic acid content, a significant decrease in oleic acid and a highly significant increase in erucic acid content were observed. This suggests that a decrease in the extent of elongation of oleic acid to erucic acid occurs in seeds developing on plants with sub-optimal levels of nitrogen nutrition. Phosphorus and potassium had very limited effects on fatty acid composition. Significant differences were found only in oleic acid content for phosphorus alone, the nitrogen-phosphorus interaction and the phosphorus-potassium interaction. The effect of various levels of sulfate at optimal levels for nitrogen, phosphorus and potassium, was studied in a separate experiment. Seed from sulfur-starved plants had decreased oil content; oleic acid percentages were increased and erucic acid percentages decreased. Excessive amounts of sulfate had no effect on fatty acid composition.  相似文献

Kidney biopsies from 23 bitches with pyometra and an entire kidney from four pyometra bitches were examined by light microscopy. Kidney tissue was also taken from three bitches at different intervals after ovariohysterectomy for pyometra. All the pyometra bitches had membranous glomerulonephritis or mixed proliferative and membranous glomerulonephritis. Two of the bitches had intraglomerular hyaline nodules resembling those seen in conjunction with diabetes in human beings. The degree of glomerular damage could be correlated with the reduction in glomerular filtration rate determined by function tests. The proximal tubules generally contained numerous hyaline droplets but the degree of this change could not be correlated to the degree of glomerular damage. A yellow pigment, a lipofuscin, was regularly present in the proximal tubules as well as epithelial proliferation and mitoses. Focal atrophy of tubules also occurred, presumably because of obliteration of glomeruli. The cortical interstitium contained collections of mature and immature plasma cells, often surrounding the glomeruli. When the kidneys from three bitches were examined after ovariohysterectomy for pyometra, the glomerular damage in two had regressed to leave only slight thickening of the capillary walls. In the third bitch, examined only 14 days after ovariohysterectomy, healing was partial. Kidney tissue from five bitches was also examined by electron microscopy. The glomerular endothelial cells were swollen and the basement membrane was grealy thickened. With more severe degrees of glomerular damage, an electron-dense material was deposited along the inner surface of the basement membrane and the swollen mesangial cells contained numerous inclusions. There was focal fusion of the foot processes of the glomerular epithelial cells; in one bitch with heavy proteinuria, the fusion was widespread. The proximal tubules contained numerous protein absorption droplets representing resorbed protein. The tubular basement membrane at all levels was thickened. Because of similarities with some other types of renal damage (nephrotoxic nephritis in dogs and acute proliferative glomerulonephritis in human beings), the possibility is broached that the renal lesion in pyometra is the result of an immunobiological process.  相似文献

This report deals with some clinical features of bitches with pyometra accompanied by polyuria. According to previous morphological studies a glomerulonephritis often acompanies pyometra in bitches. The renal dysfunction of pyometra has been compared with observations made on human beings with morphologically similar nephropathies. Reduction of the glomerular filtration rate and filtration fraction, the occurrence of proteinuria, and hypercholesterolaemia are clinical features in common. The reduced concentrating ability of pyometra bitches — probably the result of toxin released from the uterus — gives polyuria, a clinical sign which does not appear in human beings with glomerulonephritis. Other differences are the absence of oedema and hypertension in pyometra bitches. These differences do not necessarily imply a difference in the nature of the renal damage; they more likely represent species differences in reaction patterns. Possible causal relationships between the different functional abnormalities and between the functional abnormalities and the morphological changes are discussed.  相似文献

It was previously found that cations introduced into a discontinuous sucrose gradient exert a very pronounced effect on microsomal vesicles, and this principle proved to be effective in microsomal subfractionation. The mechanism of the cation effect was investigated. By using the radioactive isotopes 137Cs and 85Sr, it could be calculated that the amount of ions bound to the various subfractions increases their density by 0.14%, thereby enhancing the sedimentation velocity by only ~7%. In the presence of Cs+ the total volume of the microsomal pellet was decreased by ~15%. Assuming this change in volume to be due to a contraction of the individual vesicles, a roughly 2½-fold increase in sedimentation velocity would be expected. It is further demonstrated, on the basis of light scattering and millipore filtration experiments, that monovalent cations cause an extensive aggregation of rough microsomes and a less pronounced aggregation of smooth microsomes. The mean radius of the sedimenting particles of rough microsomes was found to be at least doubled or trebled in the presence of Cs+, which would give a 4- to 9-fold increase in the sedimentation velocity. Aggregation, therefore, appears to be the main factor in the accelerated sedimentation of rough microsomes in the presence of CsCl. Divalent cations exert a similar effect on a subfraction of the smooth microsomes. Isolated smooth microsomes are very unstable and often exhibit spontaneous aggregation. The presence of attached ribosomes, however, appears to impart greater stability to the rough microsomes as well as increasing their ability to bind monovalent cations. The primary cause of the aggregation of microsomal vesicles is probably due to a change in net charge.  相似文献

From the apical end of the inner hair cell of the organ of Corti in the guinea pig cochlea protrude four to five rows of stereocilia shaped in a pattern not unlike the wings of a bird. In the area devoid of cuticular substance facing toward the tunnel of Corti lies a consistently present centriole. The ultrastructure of this centriole is similar to that of the basal body of the kinocilium located in the periphery of the sensory hair bundles in the vestibular and lateral line organ sensory cells and to that of the centrioles of other cells. The physiological implications of the anatomical orientation of this centriole are discussed in terms of directional sensitivity.  相似文献

A case of bilateral carcinoid tumors of the breast was studied by cytology, histology, immunohistochemistry and electron microscopy. The preoperative aspiration cytologic findings strongly suggested a carcinoid tumor of the breast. The differential diagnosis with fine needle aspiration cytology of other breast lesions, the bilaterality of the condition and terminology are discussed.  相似文献

The activity of the eukaryotic elongation factor 2 (eEF-2)-specific Ca(2+)- and calmodulin-dependent protein kinase III (CaM PK III) is regulated by phosphorylation. The kinase can be inactivated by treatment with alkaline phosphatase and subsequently reactivated by endogenous protein kinase. This kinase can be substituted for by the catalytic subunit of cAMP-dependent protein kinase but not by casein kinase II. The purified kinase preparation contains only one protein as judged by gel electrophoresis. This protein has a molecular mass of approximately 90 kDa and an isoelectric point of 5.2. Reactivation of the eEF-2 kinase is associated with the phosphorylation of this protein. The amino acid sequence obtained from the 90-kDa protein reveals substantial homology with that of murine heat shock protein 86 (HSP 86) a member of the HSP 90-family. Conventional preparations of HSP 90 contain an inactive eEF-2 kinase that could be activated after dephosphorylation and phosphorylation by the catalytic subunit of cAMP-dependent protein kinase.  相似文献
